1297 A CFD Study of the Performance Characteristics of Vented Cylinders as Vortex Generators
Authors: R. Kishan, R. M. Sumant, S. Suhas, Arun Mahalingam
Abstract:
This paper mainly researched on influence of vortex generator on lift coefficient and drag coefficient, when vortex generator is mounted on a flat plate. Vented cylinders were used as vortex generators which intensify vortex shedding in the wake of the vented cylinder as compared to base line circular cylinder which ensures more attached flow and increases lift force of the system. Firstly vented cylinders were analyzed in commercial CFD software which is compared with baseline cylinders for different angles of attack and further variation of lift and drag forces were studied by varying Reynolds number to account for influence of turbulence and boundary layer in the flow. Later vented cylinders were mounted on a flat plate and variation of lift and drag coefficients was studied by varying angles of attack and studying the dependence of Reynolds number and dimensions of vortex generator on the coefficients. Mesh grid sensitivity is studied to check the convergence of the results obtained It was found that usage of vented cylinders as vortex generators increased lift forces with small variation in drag forces by varying angle of attack. Keywords: CFD analysis, drag coefficient, FVM, lift coefficient, modeling, Reynolds number, simulation, vortex generators, vortex shedding

Authors: Hanqing Yang, Xiang Meng, Qi Li, Weirong Chen
Abstract:
The SOC (state of charge) based droop control has limitations on the load power sharing among different energy storage units, due to the line impedance. In this paper, a distributed control strategy for dispersed energy storage units in the DC microgrid based on discrete consensus is proposed. Firstly, a sparse information communication network is built. Thus, local controllers can communicate with its neighbors using voltage, current and SOC information. An average voltage of grid can be evaluated to compensate voltage offset by droop control, and an objective virtual resistance fulfilling above requirement can be dynamically calculated to distribute load power according to the SOC of the energy storage units. Then, the stability of the whole system and influence of communication delay are analyzed. It can be concluded that this control strategy can improve the robustness and flexibility, because of having no center controller. Finally, a model of DC microgrid with dispersed energy storage units and loads is built, the discrete distributed algorithm is established and communication protocol is developed. The co-simulation between Matlab/Simulink and JADE (Java agent development framework) has verified the effectiveness of proposed control strategy. Keywords: dispersed energy storage units, discrete consensus algorithm, state of charge, communication delay

Authors: A. Jarrou, A. Iranzo, C. Nana
Abstract:
The rapid development of the onshore/offshore wind industry and the continuous, strong, and long-term support from governments have made it possible to create factories specializing in the manufacture of the different parts of wind turbines, but in the literature, Life Cycle Assessment (LCA) analyzes consider the wind turbine as a whole and do not allow the allocation of impacts to the different components of the wind turbine. Here we propose to treat each part of the wind turbine as a system in its own right. This is more in line with the current production system. Environmental Life Cycle Assessment of two technological scenarios of wind turbine blades composition for an optimized wind turbine design using the impact 2002+ method and using 15 environmental impact indicators. This article aims to assess the environmental impacts associated with 1 kg of wind turbine blades. In order to carry out a realistic and precise study, the different stages of the life cycle of a wind turbine installation are included in the study (manufacture, installation, use, maintenance, dismantling, and waste treatment). The Impact 2002+ method used makes it possible to assess 15 impact indicators (human toxicity, terrestrial and aquatic ecotoxicity, climate change, land use, etc.). Finally, a sensitivity study is carried out to analyze the different types of uncertainties in the data collected. Keywords: life cycle assessment, wind turbine, turbine blade, environmental impact
Procedia PDF Downloads 1781294 Analysis of an High Voltage Direct Current (HVDC) Connection Using a Real-Time Simulator Under Various Disturbances
Authors: Mankour Mohamed, Miloudi Mohamed
Abstract:
A thorough and accurate simulation is necessary for the study of a High Voltage Direct Current (HVDC) link system during various types of disturbances, including internal faults on both converters, either on the rectifier or on the inverter, as well as external faults, such as AC or DC faults on both converter sides inside the DC link party. In this study, we examine how an HVDC inverter responds to three different types of failures, including faults at the inverter valve, system control faults, and single-phase-to-ground AC faults at the sending end of the inverter side. As this phenomenon represents the most frequent problem that may affect inverter valves, particularly those based on thyristor valves (LCC (line-Commutated converter)), it is more precise to explore which circumstance generates and raises the commutation failure on inverter valves. Because of the techniques used to accelerate the simulation, digital real-time simulators are now the most potent tools that provide simulation results. The real-time-lab RT-LAB platform HYPERSIM OP-5600 is used to implement the Simulation in the Loop (SIL) technique, which is used to validate the results. It is demonstrated how to recover from both the internal faults and the AC problem. The simulation findings show how crucial a role the control system plays in fault recovery. Keywords: hypersim simulator, HVDC systems, mono-polar link, AC faults, misfiring faults

Authors: Mudavath Raju, Bhaskar Gugulothu, B. Rajendra Naik
Abstract:
The rapid advances in Very Large Scale Integration (VLSI) technology has resulted in the reduction of minimum feature size to sub-quarter microns and switching time in tens of picoseconds or even less. As a result, the degradation of high-speed digital circuits due to signal integrity issues such as coupling effects, clock feedthrough, crosstalk noise and delay uncertainty noise. Crosstalk noise in VLSI interconnects is a major concern and reduction in VLSI interconnect has become more important for high-speed digital circuits. It is the most effectively considered in Deep Sub Micron (DSM) and Ultra Deep Sub Micron (UDSM) technology. Increasing spacing in-between aggressor and victim line is one of the technique to reduce the crosstalk. Guard trace or shield insertion in-between aggressor and victim is also one of the prominent options for the minimization of crosstalk. In this paper, far end crosstalk noise is estimated with mutual inductance and capacitance RLC interconnect model. Also investigated the extent of crosstalk in capacitive and inductively coupled interconnects to minimizes the same through shield insertion technique. Keywords: VLSI, interconnects, signal integrity, crosstalk, shield insertion, guard trace, deep sub micron

Authors: Khalid Mahmud
Abstract:
In this paper, an analytic approach is presented to evaluate the Bit Error Rate (BER) and round trip distance for a UWB chipless RFID system using diversity technique at the reader receiver using different modulation technique. The analysis is carried out with multiresonator based chipless RFID tags using frequency range from 3 GHz − 6 GHz and bandwidth of 500 M Hz in outdoor non-line-of-sight (NLOS) environment. SISO configuration is used to communicate from the reader to the tag and SIMO configuration is used do vice versa. Maximal Ratio Combining (MRC) technique is used in the reader. MPSK, DQPSK, DBPSK, BPSK, QPSK and DMPSK modulation techniques are considered with coherent demodulation to evaluate the BER performance. From the numerical analysis of the results, it is found that at a given BER maximum possible round trip distance can be achieved using DMPSK modulation technique. In addition, it has been proved that, while using DMPSK modulation technique, the application of diversity has very little effect on the overall improvement in reader receiver sensitivity and achievable distance. Finally the method not only proves to be a very good way for tag detection in case of a chipless RFID system but also gives a clear insight regarding the interrelationship between BER, read range, reader received power, number of receiving antenna in outdoor NLOS environment. Keywords: EGC, MRC, BER, read range, diversity

Abstract:
This paper proposes a cellular-based structure health monitoring device (HMD) for temporary bridge monitoring without the requirement of power line and internet service. The proposed HMD includes sensor node, power module, cellular gateway, and rechargeable batteries. The purpose of HMD focuses on short-term collection of civil infrastructure information. It achieves the features of low cost by using three 1-axis accelerometers with data synchronization problem being solved. Furthermore, instead of using data acquisition system (DAQ) sensed data is transmitted to Host through cellular gateway. Compared with 3-axis accelerometer, our proposed 1-axis accelerometers based device achieves 50.5% cost saving with high sensitivity 2000mv/g. In addition to fit different monitoring environments, the proposed system can be easily replaced and/or extended with different PCB boards, such as communication interfaces and sensors, to adapt to various applications. Therefore, with using the proposed device, the real-time diagnosis system for civil infrastructure damage monitoring can be conducted effectively. Keywords: cellular-based structural health monitoring, cost-effective 1-axis accelerometers, short-term monitoring, structural engineering

Authors: Simon Bard, Martin Demleitner, Florian Schonl, Volker Altstadt
Abstract:
For an electrically driven aircraft, whose engine is based on semiconductors, alternative materials are needed. The avoid hotspots in the materials thermally conductive polymers are necessary. Nevertheless, the mechanical properties of these materials should remain. Herein, the work of three years in a project with airbus and Siemens is presented. Different strategies have been pursued to achieve conductive fiber-reinforced composites: Metal-coated carbon fibers, pitch-based fibers and particle-loaded matrices have been investigated. In addition, a combination of copper-coated fibers and a conductive matrix has been successfully tested for its conductivity and mechanical properties. First, prepregs have been produced with a laboratory scale prepreg line, which can handle materials with maximum width of 300 mm. These materials have then been processed to fiber-reinforced laminates. For the PAN-fiber reinforced laminates, it could be shown that there is a strong dependency between fiber volume content and thermal conductivity. Laminates with 50 vol% of carbon fiber offer a conductivity of 0.6 W/mK, those with 66 vol% of fiber a thermal conductivity of 1 W/mK. With pitch-based fiber, the conductivity enhances to 1.5 W/mK for 61 vol% of fiber, compared to 0.81 W/mK with the same amount of fibers produced from PAN (+83% in conducitivity). The thermal conductivity of PAN-based composites with 50 vol% of fiber is at 0.6 W/mK, their nickel-coated counterparts with the same fiber volume content offer a conductivity of 1 W/mK, an increase of 66%.Keywords: carbon, electric aircraft, polymer, thermal conductivity

Authors: Ghodhbani Salah, Jemili Farah
Abstract:
With the growth of cyber attacks, information safety has become an important issue all over the world. Many firms rely on security technologies such as intrusion detection systems (IDSs) to manage information technology security risks. IDSs are considered to be the last line of defense to secure a network and play a very important role in detecting large number of attacks. However the main problem with today’s most popular commercial IDSs is generating high volume of alerts and huge number of false positives. This drawback has become the main motivation for many research papers in IDS area. Hence, in this paper we present a data mining technique to assist network administrators to analyze and reduce false positive alarms that are produced by an IDS and increase detection accuracy. Our data mining technique is unsupervised clustering method based on hybrid ANT algorithm. This algorithm discovers clusters of intruders’ behavior without prior knowledge of a possible number of classes, then we apply K-means algorithm to improve the convergence of the ANT clustering. Experimental results on real dataset show that our proposed approach is efficient with high detection rate and low false alarm rate. Keywords: intrusion detection system, alarm filtering, ANT class, ant clustering, intruders’ behaviors, false alarms

Authors: Joachim Onyike
Abstract:
The Federal Government of Nigeria in 2012 came up with a new National Housing Policy; one of its major objectives was to make housing affordable to the poor. Six years down the line, this study was carried out to find out whether the poor have fared better under the new housing policy. Owerri, the capital of Imo State, was adopted as a case study to mirror the situation nationwide. The study population was made up of low-income civil servants, i.e., grade levels 1–6 in the Imo State Civil Service. The study looked at household size, household income, rental levels, house prices, costs of major building materials, land values, land tenure, the interest rate on mortgages, inflation rate, and the status of government interventions, owing to their obvious effect on housing affordability by the low-income earners. The study made use of physical observations, questionnaires, and interviews as well as library studies to elicit relevant information. Housing affordability by the subject population did not improve. It rather dropped. The study came to the conclusion that in spite of the new National Housing Policy, housing affordability by the low-income earners has not improved. The policy as it affects the poor has not been duly implemented by both Federal and State Governments. Keywords: house prices, housing affordability, housing policy, land values, low-income earners

Abstract:
Coprinus comatus (O. F. Müll.) Pers.) should not be confused with the common Ink Cap, which contains coprine and can induce coprine poisoning. We study the possibility of applying coprinus mushroom (Coprinus comatus), available in Poland, as food product supporting the treatment of human glioblastoma cells. The U87MG and T98 glioblastoma cell lines were exposed to water (CW) or ethanol 95° (CE) Cantharellus extracts (50-500 μg/ml), with or without temozolomide (TMZ) during 24, 48 or 72 hours. The cell division was examined by the H³-thymidine incorporation. The statistical analysis was performed using Statistica v. 13.0 software. Significant differences were assumed for p < 0.05. We found that both, CW and CE, administrated alone, had inhibitory effect on cell lines growth, but the CE extract had a higher degree of growth inhibition. The anti-tumor effect of TMZ (50 μM) on U87MG was enhanced by mushroom extracts, and the effect was lower to the effect after using Coprinus comatus extracts (CW and CE) alone. A significant decrease (p < 0.05) in pro-MMP2 (82.61 ± 6.3% of control) secretion in U87MG cells was observed after treated with CE (250 μg/ml). We conclude that extracts of Coprinus comatus, edible mushroom, present cytotoxic properties on U87MG and T98 cell lines and may cooperate with TMZ synergistically enhancing its growth inhibiting activity against glioblastoma U87MG cell line. Keywords: anticancer, glioma, mushroom, temozolomide

Abstract:
This work aims to develop multi-criteria decision making (MCDM) and spatial data warehouse (SDW) methods, which will be integrated into a GIS according to a ‘GIS dominant’ approach. The GIS operating tools will be operational to operate the SDW. The MCDM methods can provide many solutions to a set of problems with various and multiple criteria. When the problem is so complex, integrating spatial dimension, it makes sense to combine the MCDM process with other approaches like data mining, ascending analyses, we present in this paper an experiment showing a geo-decisional methodology of SWD construction, On-line analytical processing (OLAP) technology which combines both basic multidimensional analysis and the concepts of data mining provides powerful tools to highlight inductions and information not obvious by traditional tools. However, these OLAP tools become more complex in the presence of the spatial dimension. The integration of OLAP with a GIS is the future geographic and spatial information solution. GIS offers advanced functions for the acquisition, storage, analysis, and display of geographic information. However, their effectiveness for complex spatial analysis is questionable due to their determinism and their decisional rigor. A prerequisite for the implementation of any analysis or exploration of spatial data requires the construction and structuring of a spatial data warehouse (SDW). This SDW must be easily usable by the GIS and by the tools offered by an OLAP system. Keywords: data warehouse, GIS, MCDM, SOLAP

Authors: Mehdi Montakhabrazlighi, Ercan Balikci
Abstract:
The neural network (NN) method is applied to alloy development of single crystal Ni-base Superalloys with low density and improved mechanical strength. A set of 1200 dataset which includes chemical composition of the alloys, applied stress and temperature as inputs and density and time to rupture as outputs is used for training and testing the network. Thermodynamic phase diagram modeling of the screened alloys is performed with Thermocalc software to model the equilibrium phases and also microsegregation in solidification processing. The model is first trained by 80% of the data and the 20% rest is used to test it. Comparing the predicted values and the experimental ones showed that a well-trained network is capable of accurately predicting the density and time to rupture strength of the Ni-base superalloys. Modeling results is used to determine the effect of alloying elements, stress, temperature and gamma-prime phase volume fraction on rupture strength of the Ni-base superalloys. This approach is in line with the materials genome initiative and integrated computed materials engineering approaches promoted recently with the aim of reducing the cost and time for development of new alloys for critical aerospace components. Authors: Zehai Yu, Hui Zhu, Linglong Lin, Huawei Liang, Biao Yu, Weixin Huang
Abstract:
With the development of intelligent vehicle systems, a high-precision road map is increasingly needed in many aspects. The automatic lane lines extraction and modeling are the most essential steps for the generation of a precise lane-level road map. In this paper, an automatic lane-level road map generation system is proposed. To extract the road markings on the ground, the multi-region Otsu thresholding method is applied, which calculates the intensity value of laser data that maximizes the variance between background and road markings. The extracted road marking points are then projected to the raster image and clustered using a two-stage clustering algorithm. Lane lines are subsequently recognized from these clusters by the shape features of their minimum bounding rectangle. To ensure the storage efficiency of the map, the lane lines are approximated to cubic polynomial curves using a Bayesian estimation approach. The proposed lane-level road map generation system has been tested on urban and expressway conditions in Hefei, China. The experimental results on the datasets show that our method can achieve excellent extraction and clustering effect, and the fitted lines can reach a high position accuracy with an error of less than 10 cm.Keywords: curve fitting, lane-level road map, line recognition, multi-thresholding, two-stage clustering
Procedia PDF Downloads 1281278 Merging Appeal to Ignorance, Composition, and Division Argument Schemes with Bayesian Networks
Authors: Kong Ngai Pei
Abstract:
The argument scheme approach to argumentation has two components. One is to identify the recurrent patterns of inferences used in everyday discourse. The second is to devise critical questions to evaluate the inferences in these patterns. Although this approach is intuitive and contains many insightful ideas, it has been noted to be not free of problems. One is that due to its disavowing the probability calculus, it cannot give the exact strength of an inference. In order to tackle this problem, thereby paving the way to a more complete normative account of argument strength, it has been proposed, the most promising way is to combine the scheme-based approach with Bayesian networks (BNs). This paper pursues this line of thought, attempting to combine three common schemes, Appeal to Ignorance, Composition, and Division, with BNs. In the first part, it is argued that most (if not all) formulations of the critical questions corresponding to these schemes in the current argumentation literature are incomplete and not very informative. To remedy these flaws, more thorough and precise formulations of these questions are provided. In the second part, how to use graphical idioms (e.g. measurement and synthesis idioms) to translate the schemes as well as their corresponding critical questions to graphical structure of BNs, and how to define probability tables of the nodes using functions of various sorts are shown. In the final part, it is argued that many misuses of these schemes, traditionally called fallacies with the same names as the schemes, can indeed be adequately accounted for by the BN models proposed in this paper. Keywords: appeal to ignorance, argument schemes, Bayesian networks, composition, division

Abstract:
The production of rainbow trout in the Lake Titicaca represents an important economic activity for Peru. The city of Puno is responsible for 83% of this production, so the use of antibiotics within the aquaculture system is not alien to this reality. Meanwhile, the waters of Lake Titicaca represent an important source for the supply of drinking water for 80% of the population of the Puno city. In this paper, twelve antibiotics for veterinary use were monitored in water samples during two seasons: dry (July 2015) and rainy (February 2016), water samples from trout production systems, near the water catching point in the lake and drinking water in the city house of Puno were considered. The samples were analyzed using liquid chromatography coupled to mass spectrometry and solid online phase extraction (On-line SPE-LC-MS/MS), all samples analyzed showed concentrations of Ciprofloxacin up to 65.2 ng L⁻¹ at the rainy season. On the other hand, 63% of water samples from the dry season and 36 % from the rainy season showed Chlortetracycline up to 8.7 and 6.1 ng L⁻¹, respectively. The presence of residues of veterinary antibiotics in drinking water means a serious health risk for 80% of the population of Puno since all these people are supplied from this source. Keywords: chromatography, DNA damage, environmental risk, water pollution

Abstract:
Cationic niosomes formulated with Span20, cholesterol and novel synthesized spermine-cationic lipids (2-hydrocarbon tail and 4- hydrocarbon tail) in a molar ratio of 2.5:2.5:1 can mediate high gene transfection in vitro. However, the uptake mechanisms of these systems are not well clarified. In the present study, effect of endocytic inhibitors on the transfection efficiency of niosomes/DNA complexes was determined on a human cervical carcinoma cell line (HeLa cells) using the inhibitors of macropinocytosis (wortmannin), clathrin- and caveolae-mediated endocytosis (methyl-β-cyclodextrin), clathrin-mediated endocytosis (chlorpromazine), caveolae-mediated endocytosis (genistein and filipin), cytosolic transfer (ammonium chloride) and microtubules polymerization (nocodazole). Authors: Abdull Halim
Abstract:
The Standard Penetration Test (SPT-N) is the most common in situ test for soil investigations. The Shearing Seismic Standard Penetration Test (SSPT-N), on the other hand, is a new method using shearing wave with propagation exponent equation between the shearing wave, Vs., and hardness, N values without any need for borehole data. Due to the fast and accurate results that can be obtained, the SSPT has found many applications such as in the field rectification buried pipe line, the acid tank settlement and foundation design analyses, and the quality control assessment. Many geotechnical regimes and properties have attempted to correlate both the SSPT and the SPT-N values. Various foundation design methods have been developed based on the outcomes of these tests. Hence, it is pertinent to correlate these tests so that either one of the test can be used in the absence of the other, especially for preliminary evaluation and design purposes. The primary purpose of this study was to investigate the relationship between the SSPT-N and SPT-N values for different types of cohesive soil in Peninsular Malaysia. Data were collected from four different sites, and the correlations were established between the hardness N values, principal stress-strain Mohr circle curve, cohesion, friction angle and vertical effective stress. A positive exponent relationship was found between the shearing wave, sVs., and the hardness N values of the soil. In general, the SSPT-N value was slightly lower than the SPT-N value due to the upper limit boundary of the soil layer. Keywords: InsituSoil determination; shearing wave; hardness; correlation, SSPT-N, SPT-N

Authors: Elham S. Hasham, Anthony S. Hasham
Abstract:
The construction industry is one of Australia’s fastest growing industries and its success is a result of a firm’s client satisfaction with focus on product determinants such as price and quality. Ensuring quality at every phase is a must and building rapport with the client will go a long way. To capitalise on the growing demand for Engineering Consulting Firms (ECFs), we should “redefine the bottom line by allowing client satisfaction, high-quality standards, and profits to be the top priorities”. Consequently, the emphasis should be on improving employee skills through various training provisions. Clients seek consistency and thus expect that all services should be similar in respect to quality and the ability of the service to meet their needs. This calls for empowerment and comfortable work conditions to motivate employees and give them incentive to deliver quality and excellent output. The methodology utilized is triangulation-a combination of both quantitative and qualitative research. The case study-Australian Consulting Engineers (ACE) was established in 1995 and has operations throughout Australia, the Philippines, Europe, U.A.E., K.S.A., and Lebanon. ACE is affiliated with key agencies and support organizations in the engineering industry with International Organization for Standardization (ISO) certifications in Safety and Quality Management. The objective of this study is significant as it sheds light on employee motivation and client satisfaction as imperative determinants of the success of an organization. Keywords: leadership, motivation, organizational behavior, satisfaction

Authors: Ghodbane Azeddine, Saad Maarouf, Boland Jean-Francois, Thibeault Claude
Abstract:
This paper proposes a new design of an active fault-tolerant flight control system against abrupt actuator faults. This overall system combines a multiple time scale sliding mode controller for fault compensation and a geometric approach for fault detection and diagnosis. The proposed control system is able to accommodate several kinds of partial and total actuator failures, by using available healthy redundancy actuators. The overall system first estimates the correct fault information using the geometric approach. Then, and based on that, a new reconfigurable control law is designed based on the multiple time scale sliding mode technique for on-line compensating the effect of such faults. This approach takes advantages of the fact that there are significant difference between the time scales of aircraft states that have a slow dynamics and those that have a fast dynamics. The closed-loop stability of the overall system is proved using Lyapunov technique. A case study of the non-linear model of the F16 fighter, subject to the rudder total loss of control confirms the effectiveness of the proposed approach. Keywords: actuator faults, fault detection and diagnosis, fault tolerant flight control, sliding mode control, multiple time scale approximation, geometric approach for fault reconstruction, lyapunov stability

Authors: Jeanne Grace, Shereen Currie
Abstract:
Background: Obesity associated comorbidities increase the risk of morbidity and mortality among employees in the workplace. Objectives: The study aimed to determine the prevalence of obesity and comorbidities like diabetes, hypertension, and hypercholesterolemia associated with obesity within the workplace in South Africa. Methods: A total of 17359 male (n = 8561) and female (n = 8798) employees, aged between 18-64 years (40.8 ± 11.0), from various corporate and industrial companies in South Africa participated in the study. Subjects were assigned to one of five body mass index (BMI) categories, according to their BMI: normal weight, BMI of 18.5‒24.9 kg/m² (n = 7338); overweight, BMI of 25.0‒29.9 kg/m² (n = 6323); obese class I, BMI of 30.0-34.9 kg/m² (n = 2552); obese class II, BMI of 35.0-39.9 kg/m² (n = 782); and obese class III, BMI of ≥ 40 kg/m² (n = 364). Height, weight, blood pressure, random blood glucose, and total cholesterol were measured. Results: The prevalence of normal weight men was 29.2% and women 55.0%; overweight men 46.4% and women 26.7%, obese men 24.4% and women 18.3%. A significant association (p<0.01) of BMI with diabetes, systolic and diastolic hypertension, and hypercholesterolemia were noted. Conclusion: Obesity is strongly associated with adverse comorbidities that may impact employees’ quality of life and performance. If unaddressed, it can increase comorbidities, not only affecting the bottom line of companies but causing morbidity and mortality, including sudden death. Keywords: body mass index, cholesterol, blood glucose, workplace

Abstract:
In the present study, we attempted to develop Mycobacterium tuberculosis (Mtb) inhibitors by exploring the pharmaceutically underexploited enzyme targets which are majorly involved in cell wall biosynthesis of mycobacteria. For this purpose, glutamate racemase (coded by MurI gene) was selected. This enzyme racemize L-glutamate to D-glutamate required for the construction of peptidoglycan in the bacterial cell wall synthesis process. Furthermore this enzyme is neither expressed nor its product, D-glutamate is normally found in mammals, and hence designing inhibitors against this enzyme will not affect the host system as well act as potential antitubercular drugs. A library of BITS in house compounds were screened against Mtb MurI enzyme. Based on docking score, interactions and synthetic feasibility one hit lead was identified. Further optimization of lead was attempted and its derivatives were synthesized. Forty eight derivatives of 2-phenylbenzo[d]oxazole and 2-phenylbenzo[d]thiazole were synthesized and evaluated for Mtb MurI inhibition study, in vitro activities against Mtb, cytotoxicity against RAW 264.7 cell line. Chemical derivatization of the lead resulted in compounds NR-1213 AND NR-1124 as the potent M. tuberculosis glutamate racemase inhibitors with IC50 of 4-5µM which are remarkable and were found to be non-cytotoxic. Molecular dynamics, dormant models and cardiotoxicity studies of the most active molecules are in process. Keywords: cell wall biosynthesis, dormancy, glutamate racemase, tuberculosis
