Authors: N. H. Hamid, M. S. Syaref, M. I. Adiyanto, M. Mohamed
Abstract:
A one-half scale of single-bay two-storey RC frame together with foundation beam and mass concrete block is investigated. Moment resisting RC frame was designed using EC8 by including the provision for seismic loading and detailing of its connection. The objective of the experimental work is to determine seismic behaviour RC frame under in-plane lateral cyclic loading using displacement control method. A double actuator is placed at centre of the mass concrete block at top of frame to represent the seismic load. The percentage drifts are starting from ±0.01% until ±2.25% with increment of ±0.25% drift. The ultimate lateral load of 158.48 kN was recorded at +2.25% drift in pushing and -126.09 kN in pulling direction. From the experimental hysteresis loops, the parameters such as lateral strength capacity, stiffness, ductility and equivalent viscous damping can be obtained. RC frame behaves in the elastic manner followed by inelastic behaviour after reaches the yield limit. The ductility value for this type frame is 4 which lies between the limit 3 and 6. Therefore, it is recommended to build this RC frame for moderate seismic regions under Ductility Class Medium (DCM) such as in Sabah, East Malaysia. Keywords: single bay, moment resisting RC frame, ductility class medium, inelastic behavior, seismic load

Authors: Minsu Kim, Hae-Chang Cho, Jae Hoon Chung, Inwook Heo, Kang Su Kim
Abstract:
For safe design of the UHPC flexural members, accurate estimations of their shear strengths are very important. However, since the shear strengths are significantly affected by various factors such as tensile strength of concrete, shear span to depth ratio, volume ratio of steel fiber, and steel fiber factor, the accurate estimations of their shear strengths are very challenging. In this study, therefore, the Adaptive Neuro-Fuzzy System (ANFIS), which has been widely used to solve many complex problems in engineering fields, was introduced to estimate the shear strengths of UHPC flexural members. A total of 32 experimental results has been collected from previous studies for training of the ANFIS algorithm, and the well-trained ANFIS algorithm provided good estimations on the shear strengths of the UHPC test specimens. Procedia PDF Downloads 4021777 BIM-based Construction Noise Management Approach With a Focus on Inner-City Construction
Authors: Nasim Babazadeh
Abstract:
Growing demand for a quieter dwelling environment has turned the attention of construction companies to reducing the propagated noise of their project. In inner-city constructions, close distance between the construction site and surrounding buildings lessens the efficiency of passive noise control methods. Dwellers of the nearby areas may file complaints and lawsuits against the construction companies due to the emitted construction noise, thereby leading to the interruption of processes, compensation costs, or even suspension of the project. Therefore, construction noise should be predicted along with the project schedule. The advantage of managing the noise in the pre-construction phase is two-fold. Firstly, changes in the time plan and construction methods can be applied more flexibly. Thus, the costs related to rescheduling can be avoided. Secondly, noise-related legal problems are expected to be reduced. To implement noise mapping methods for the mentioned prediction, the required detailed information (such as the location of the noisy process, duration of the noisy work) can be exported from the 4D BIM model. The results obtained from the noise maps would be used to help the planners to define different work scenarios. The proposed approach has been applied for the foundation and earthwork of a site located in a residential area, and the obtained results are discussed. Keywords: building information modeling, construction noise management, noise mapping, 4D BIM
Procedia PDF Downloads 1861776 Dynamic Analysis of Nanosize FG Rectangular Plates Based on Simple Nonlocal Quasi 3D HSDT
Authors: Sabrina Boutaleb, Fouad Bourad, Kouider Halim Benrahou, Abdelouahed Tounsi
Abstract:
In the present work, the dynamic analysis of the functionally graded rectangular nanoplates is studied. The theory of nonlocal elasticity based on the quasi 3D high shear deformation theory (quasi 3D HSDT) has been employed to determine the natural frequencies of the nanosized FG plate. In HSDT, a cubic function is employed in terms of thickness coordinates to introduce the influence of transverse shear deformation and stretching thickness. The theory of nonlocal elasticity is utilized to examine the impact of the small scale on the natural frequency of the FG rectangular nanoplate. The equations of motion are deduced by implementing Hamilton’s principle. To demonstrate the accuracy of the proposed method, the calculated results in specific cases are compared and examined with available results in the literature, and a good agreement is observed. Finally, the influence of the various parameters, such as the nonlocal coefficient, the material indexes, the aspect ratio, and the thickness-to-length ratio, on the dynamic properties of the FG nanoplates is illustrated and discussed in detail. Keywords: nonlocal elasticity theory, FG nanoplate, free vibration, refined theory, elastic foundation

Authors: K. Nirmalkumar
Abstract:
An acute water scarcity is prevailing in the dry season in and around Perundurai (Erode district, Tamil Nadu, India) where there are more number of tannery units. Hence an attempt was made to use the effluent from the tannery industry for construction purpose. The mechanical properties such as compressive strength, tensile strength, flexural strength and the special properties such as chloride attack, sulphate attack and chemical attack were studied by casting various concrete specimens in form of cube, cylinders and beams, etc. It was observed that the concrete had some reduction in strength while subjected to chloride attack, sulphate attack and chemical attack. So admixtures were selected and optimized in suitable proportion to counter act the adverse effects and the results were found to be satisfactory. In this research study the corrosion results of specimens prepared by using treated and untreated tannery effluent were compared with the concrete specimens prepared by using potable water. It was observed that by the addition of admixtures, the adverse effects due to the usage of the treated and untreated tannery effluent are counteracted. Keywords: corrosion, calcium nitrite, concrete, fly ash

Authors: Vanessa L. Haro
Abstract:
Beginning with a historical foundation of Mexico, this marks the start of a close examination of this major Latin American country by providing the context needed to understand the reasons for Mexico’s strengths and struggles today, specific to their response to the issue of gender violence. Responding to the challenge of combating gender violence and inequality, Mexico has created social programs and initiatives in hopes of addressing these issues and modernizing their gender norms, which currently disempower and dehumanize women, while simultaneously denying women the necessary tools needed to fight back or bring balance to the gender scales. Nevertheless, women in Mexico have made their voices heard with the most salient image of that of the mothers protesting while holding the photos of their young daughters who lost their lives. This case study on gender issues in Mexico works to acknowledge the diverse forces that contribute to the issue of gender violence, and to make a statement that this is a crisis that requires a more dynamic response within Mexico’s social welfare policies, and should not be allowed to continue to progress as a normative phenomenon. As the advocacy groups and protesters cry out, “Ni una menos! (Not one less), meaning we will not lose one more woman and making the statement that all women’s lives matter. Keywords: gender issues, Mexico, poverty, social welfare

Abstract:
Kedulan Temple is located in Dusun Kedulan, Sleman, Yogyakarta, Indonesia at coordinates S 07o 44’ 57’, E 110o 28’ 17’. Kedulan Temple is a trace of the relics of life in the 3 century AD. The Kedulan Temple including exhumed landforms, which the primordial landform is first surface topography, then buried under cover mass and exposed or re-inscribed. Recognized by the existence of ancient soil (paleosoil) and ancient objects. Seen from the type of soil that closes the temple, there are 13 layers of lava type, so it is estimated that the lava that buried the temple came from 13 times the eruption of Mount Merapi. The material that buries the base of this temple is the pyroclastic surge deposits in 3 layers, each of which is limited by a thin layer of paleosol, the sediments are 1445+/-50 yBP, 1175+/-50 yBP, and 1060+/-40 yBP. This temple is buried and dug again at 940+/-100 yBP. Furthermore, the temple affected by earthquake, so the floor and foundation becomes bumpy and most of the temple stone are thrown. The temple is left alone, until exposed to hot clouds at 1285 M (740+/-50yBP). Next, repeatedly buried lava in 4 periods, in 1587 M (360+/-50 yBP, 240+/-50 yBP, 200+/-50 yBP and unknown date). From studying this temple, can be known paleogeomorphology process that occurred in Yogyakarta, especially related to the volcanic activity of Mount Merapi. Until now, the water is still flowing around the temple so there is a fluvial process that began to take a role in the temple. Keywords: Kedulan temple, paleogeomorphology, buried, mount Merapi, Yogyakarta

Authors: Lalla Nezha Lakmiti, Abdelkahar Zahid
Abstract:
Due to the setbacks on the international scene and the wave of cacophonic financial scandals affecting large international groups, the new Islamic finance industry is not immune despite its initial resistance. The purpose of this paper is to understand and analyze the meaning of the Corporate Governance (CG) concept in Moroccan Islamic banking systems with specific reference to their institutions. The research objective is to identify also the path taken and adopted by these banks recently set up in Morocco. The foundation is rooted in shari'a, in particular, no stakeholder (the shareholding approach) must be harmed, and the ethical value is reflected into these parties’ behavior. We chose a qualitative method, semi-structured interviews where six managers provided answers about their banking systems. Since these respondents held a senior position (directors) within their organizations, it is felt that they are well placed and have the necessary knowledge to provide us with information to answer the questions asked. The results identified the orientation of participating banks and assessing how governance works, while determining which party is fovoured: shareholders, stakeholders or both. This study discusses the favorable condition to the harmonization of the regulations and therefore a better integration between Islamic finance and conventional ones in the economic context of Morocco. Keywords: corporate governance, Islamic Banks, stakeholders, shareholders

Abstract:
This article details the development of the X-ray response team (XRT), a service that utilises innovative technology to safely deliver acute and elective imaging and medical assessment service in the pre-hospital and community setting. This involves a partnership between Northumbria Healthcare NHS Foundation Trust’s Radiology and Emergency Medicine departments and the North East Ambulance Service to create a multidisciplinary prehospital team. The team committed to the delivery of a two-day acute service every week, alongside elective referrals, starting in November 2020. The service was originally made available to a 15-mile radius surrounding the Northumbria Hospital. Due to demand, this was expanded to include the North Tyneside and Northumberland regions. The target population was specified as frail and vulnerable patients, as well as those deemed to benefit from staying in their own environment. Within the first two months, thirty-six percent of patients assessed were able to stay at home due to the provision of off-site imaging. In the future, this service aims to allow patient transfer directly to an appropriate ward or clinic, bypassing the emergency department to improve the patient journey and reduce emergency care pressures. Keywords: frailty, imaging, pre-hospital, X-ray

Procedia PDF Downloads 1211769 Integrating Assurance and Risk Management of Complex Systems
Authors: Odd Ivar Haugen
Abstract:
This paper explores the relationship between assurance, risk, and risk management in the context of complex safety-related systems. It introduces a nuanced understanding of assurance and argues that the foundation for grounds for justified confidence in claims made about a complex system is related to the system behaviour. It emphasises the importance of knowledge as the cornerstone of assurance. The paper addresses the challenges of epistemic and aleatory uncertainties inherent in safety-critical systems. A systems approach is proposed to model emergent properties and complexity using the composition, environment, structure, mechanisms (CESM) metamodel, offering a structured framework for analysing system behaviour. The interplay between assurance and risk management is conceptualised through two models: the domain model and the control model. Assurance and risk management are mutually dependent on each other to reduce uncertainty and control risk levels. This work highlights the dual roles of assurance in risk management, acting as an epistemic actuator on the one side and providing feedback about the strength of the justification on the other. Assurance and risk management have inseparable roles in ensuring safety in complex systems. Keywords: assurance, CESM metamodel, confidence, emergent properties, knowledge, objectivity, risk, system behaviour, system safety

Authors: A. Hamam, D. Oukil, A. Dib, L. Makhloufi
Abstract:
The aim of this work is to synthesize modified Polypyrrole films (PPy) containing nanoparticles of copper oxides onto a non conducting cellulosic substrate. Firstly, the chemical polymerization of polypyrrole onto cellulosic substrate is carried out using FeCl3 as an oxidant and Pyrrole as monomer. Different parameters were optimized (monomer concentration, duration of the experiment, nature of supporting electrolyte, temperature, etc.) in order to obtain films with different thickness and different morphologies. Thickness and topography of different PPy deposits were estimated by a profilometer apparatus. The electrochemical reactivity of the obtained electrodes were tested by cyclic voltammetry technique (CV) and electrochemical impedance spectroscopy (EIS). Secondly, the modification of the PPy film surface by incorporation of copper oxide nanonoparticles is conducted by applying a galvanostatic procedure from CuCl2 solution. Surface characterization has been carried out using scanning microscope (SEM) coupled with energy dispersive X-ray analysis (EDX), Fourier-transform infrared spectroscopy (FTIR) and X-ray diffraction (XRD). The analysis showed the presence of the copper oxide nanoparticles (CuO) in the polymer films with dimensions less than 50 nm.Keywords: polypyrrole, modified electrode, cellulosic substrate, copper oxide

Procedia PDF Downloads 1951766 Estimation of Fourier Coefficients of Flux Density for Surface Mounted Permanent Magnet (SMPM) Generators by Direct Search Optimization
Authors: Ramakrishna Rao Mamidi
Abstract:
It is essential for Surface Mounted Permanent Magnet (SMPM) generators to determine the performance prediction and analyze the magnet’s air gap flux density wave shape. The flux density wave shape is neither a pure sine wave or square wave nor a combination. This is due to the variation of air gap reluctance between the stator and permanent magnets. The stator slot openings and the number of slots make the wave shape highly complicated. To reduce the complexity of analysis, approximations are made to the wave shape using Fourier analysis. In contrast to the traditional integration method, the Fourier coefficients, an and bn, are obtained by direct search method optimization. The wave shape with optimized coefficients gives a wave shape close to the desired wave shape. Harmonics amplitudes are worked out and compared with initial values. It can be concluded that the direct search method can be used for estimating Fourier coefficients for irregular wave shapes. Keywords: direct search, flux plot, fourier analysis, permanent magnets
Procedia PDF Downloads 2161765 Migration as a Climate Change Adaptation Strategy: A Conceptual Equation for Analysis
Authors: Elisha Kyirem
Abstract:
Undoubtedly, climate change is a major global challenge that could threaten the very foundation upon which life on earth is anchored, with its impacts on human mobility attracting the attention of policy makers and researchers. There is an increasing body of literature and case studies suggesting that migration could be a way through which the vulnerable move away from areas exposed to climate extreme events to improve their lives and that of their families. This presents migration as a way through which people voluntarily move to seek opportunities that could help reduce their exposure and avoid danger from climate events. Thus, migration is seen as a proactive adaptation strategy aimed at building resilience and improving livelihoods to enable people to adapt to future changing events. However, there has not been any mathematical equation linking migration and climate change adaptation. Drawing from literature in development studies, this paper develops an equation that seeks to link the relationship between migration and climate change adaptation. The mathematical equation establishes the linkages between migration, resilience, poverty reduction and vulnerability, and these the paper maintains, are the key variables for conceptualizing the migration-climate change adaptation nexus. The paper then tests the validity of the equation using the sustainable livelihood framework and publicly available data on migration and tourism in Ghana. Keywords: migration, adaptation, climate change, adaptation, poverty reduction
Procedia PDF Downloads 3971764 Advantages of Matrix Solid Phase Dispersive (MSPD) Extraction Associated to MIPS versus MAE Liquid Extraction for the Simultaneous Analysis of PAHs, PCBs and Some Hydroxylated PAHs in Sediments
Authors: F. Portet-Koltalo, Y. Tian, I. Berger, C. Boulanger-Lecomte, A. Benamar, N. Machour
Abstract:
Sediments are complex environments which can accumulate a great variety of persistent toxic contaminants such as polychlorobiphenyles (PCBs), polycyclic aromatic hydrocarbons (PAHs) and some of their more toxic degradation metabolites such as hydroxylated PAHs (OH-PAHs). Owing to their composition, fine clayey sediments can be more difficult to extract than soils using conventional solvent extraction processes. So this study aimed to compare the potential of MSPD (matrix solid phase dispersive extraction) to extract PCBs, PAHs and OH-PAHs, in comparison with microwave assisted extraction (MAE). Methodologies: MAE extraction with various solvent mixtures was used to extract PCBs, PAHs and OH-PAHs from sediments in two runs, followed by two GC-MS analyses. MSPD consisted in crushing the dried sediment with dispersive agents, introducing the mixture in cartridges and eluting the target compounds with an appropriate volume of selected solvents. So MSPD combined with cartridges containing MIPs (molecularly imprinted polymers) designed for OH-PAHs was used to extract the three families of target compounds in only one run, followed by parallel analyses in GC-MS for PAHs/PCBs and HPLC-FLD for OH-PAHs. Results: MAE extraction was optimized to extract from clayey sediments, in two runs, PAHs/PCBs in one hand and OH-PAHs in the other hand. Indeed, the best conditions of extractions (mixtures of extracting solvents, temperature) were different if we consider the polarity and the thermodegradability of the different families of target contaminants: PAHs/PCBs were better extracted using an acetone/toluene 50/50 mixture at 130°C whereas OH-PAHs were better extracted using an acetonitrile/toluene 90/10 mixture at 100°C. Moreover, the two consecutive GC-MS analyses contributed to double the total analysis time. A matrix solid phase dispersive (MSPD) extraction procedure was also optimized, with the first objective of increasing the extraction recovery yields of PAHs and PCBs from fine-grained sediment. The crushing time (2-10 min), the nature of the dispersing agents added for purifying and increasing the extraction yields (Florisil, octadecylsilane, 3-chloropropyle, 4-benzylchloride), the nature and the volume of eluting solvents (methylene chloride, hexane, hexane/acetone…) were studied. It appeared that in the best conditions, MSPD was a better extraction method than MAE for PAHs and PCBs, with respectively, mean increases of 8.2% and 71%. This method was also faster, easier and less expensive. But the other advantage of MSPD was that it allowed to introduce easily, just after the first elution process of PAHs/PCBs, a step permitting the selective recovery of OH-PAHs. A cartridge containing MIPs designed for phenols was coupled to the cartridge containing the dispersed sediment, and various eluting solvents, different from those used for PAHs and PCBs, were tested to selectively concentrate and extract OH-PAHs. Thereafter OH-PAHs could be analyzed at the same time than PAHs and PCBs: the OH-PAH extract could be analyzed with HPLC-FLD, whereas the PAHs/PCBs extract was analyzed with GC-MS, adding only few minutes more to the total duration of the analytical process. Conclusion: MSPD associated to MIPs appeared to be an easy, fast and low expensive method, able to extract in one run a complex mixture of toxic apolar and more polar contaminants present in clayey fine-grained sediments, an environmental matrix which is generally difficult to analyze. Keywords: contaminated fine-grained sediments, matrix solid phase dispersive extraction, microwave assisted extraction, molecularly imprinted polymers, multi-pollutant analysis
Procedia PDF Downloads 3551763 Properties of Preplaced Aggregate Concrete with Modified Binder
Authors: Kunal Krishna Das, Eddie S. S. Lam
Abstract:
Preplaced Aggregate Concrete (PAC) is produced by first placing the coarse aggregate into the formwork, followed by injection of grout to fill in the voids in between the coarse aggregates. In this study, tests were carried out to determine the effects of supplementary cementitious materials on the properties of PAC. Cement was partially replaced by ground granulated blast furnace slag (GGBS) and silica fume (SF) at different proportions. Grout properties were determined by the flow cone test and compressive strength test. Grout proportion was optimized statistically. It was applied to form PAC. Hardened properties of PAC, comprising compressive strength, splitting tensile strength, chloride-ion penetration and drying shrinkage, were evaluated. GGBS enhanced the flowability of the grout, whereas SF enhanced the strength of PAC. Both GGBS and SF improved the resistance to chloride-ion penetration with the drawback of increased drying shrinkage. Nevertheless, drying shrinkage was within the range to be classified as low shrinkage concrete. Keywords: factorial design, ground granulated blast furnace slag, preplaced aggregate concrete, silica fume

Abstract:
Asthma and Chronic obstructive pulmonary disease (COPD) are common lung diseases that have a significant global impact. Pressurized metered dose inhalers (pMDIs) are widely used for treatment, but they can have limitations such as high medication release speed resulting in drug deposition in the mouth or oral cavity and difficulty achieving proper synchronization with inhalation by users. Spacers are add-on devices that improve the efficiency of pMDIs by reducing the release speed and providing space for aerosol particle breakup to have finer and medically effective medication. The aim of this study is to optimize the size and cylindrical shape of spacers to enhance their drug delivery performance. The study was based on fluid dynamics theory and employed Ansys software for simulation and optimization. Results showed that optimization of the spacer's geometry greatly influenced its performance and improved drug delivery. This study provides a foundation for future research on enhancing the efficiency of inhalation therapy for lung diseases. Keywords: asthma, COPD, pressurized metered dose inhalers, spacers, CFD, shape optimization

Authors: Uswah Khairuddin, Rubiyah Yusof, Nenny Ruthfalydia Rosli
Abstract:
This paper presents a comparative study between k-Nearest Neighbour (k-NN) and Multi-Layer Perceptron Neural Network (MLP-NN) classifier using Genetic Algorithm (GA) as feature selector for wood recognition system. The features have been extracted from the images using Grey Level Co-Occurrence Matrix (GLCM). The use of GA based feature selection is mainly to ensure that the database used for training the features for the wood species pattern classifier consists of only optimized features. The feature selection process is aimed at selecting only the most discriminating features of the wood species to reduce the confusion for the pattern classifier. This feature selection approach maintains the ‘good’ features that minimizes the inter-class distance and maximizes the intra-class distance. Wrapper GA is used with k-NN classifier as fitness evaluator (GA-kNN). The results shows that k-NN is the best choice of classifier because it uses a very simple distance calculation algorithm and classification tasks can be done in a short time with good classification accuracy. Keywords: feature selection, genetic algorithm, optimization, wood recognition system

Abstract:
In this paper, our system is a Markovien system which we can see it like a Dynamic Bayesian Networks. One of the major interests of these systems resides in the complete training of the models (topology and parameters) starting from training data. The Bayesian Networks are representing models of dubious knowledge on complex phenomena. They are a union between the theory of probability and the graph theory in order to give effective tools to represent a joined probability distribution on a set of random variables. The representation of knowledge bases on description, by graphs, relations of causality existing between the variables defining the field of study. The theory of Dynamic Bayesian Networks is a generalization of the Bayesians networks to the dynamic processes. Our objective amounts finding the better structure which represents the relationships (dependencies) between the variables of a dynamic bayesian network. In applications in pattern recognition, one will carry out the fixing of the structure which obliges us to admit some strong assumptions (for example independence between some variables). Keywords: Arabic on line character recognition, dynamic Bayesian network, pattern recognition, networks

Authors: Artem A. Krylov
Abstract:
Instrumental seismological researches in water areas are complicated and expensive, that leads to the lack of strong motion records in most offshore regions. In the same time the number of offshore industrial infrastructure objects, such as oil rigs, subsea pipelines, is constantly increasing. The empirical Green’s function technique proved to be very effective for accelerograms synthesis under the conditions of poorly described seismic wave propagation medium. But the selection of suitable small earthquake record in offshore regions as an empirical Green’s function is a problem because of short seafloor instrumental seismological investigation results usually with weak micro-earthquakes recordings. An approach based on moving average smoothing in the frequency domain is presented for preliminary processing of weak micro-earthquake records before using it as empirical Green’s function. The method results in significant waveform correction for modeled event. The case study for 2009 L’Aquila earthquake was used to demonstrate the suitability of the method. Authors: Benjamin Orimolade, Emily Draper
Abstract:
The interest in all solid electrochromic devices (ECD) is ongoing. This is because these devices offer realistic applications of electrochromic materials in products such as sensors, windows and energy storage devices. The use of quasi-solid (gel) electrolytes for the construction of these ECDs is attractive because of their ease of preparation, availability, low cost, improved electrochromic performance, good ionic conductivity and prevention of leakages in ECDs. Herein, we developed a gel electrolyte consisting of polyacrylamide modified with succinonitrile for an ECD containing leucine-modified naphthalene diimide (NDI-L) as electrochromic material. The amount of succinonitrile in the gel was optimized, and the structure, surface morphology, and ionic conductivity of the electrolytes were assessed using microscopic techniques and electrochemical methods. The ECD fabricated with the gel electrolyte displayed good electrochromic performance with a fast switching response of up to 10 s and outstanding stability. These results add significant insight into understanding the inter- and intra-molecular interaction in succinonitrile gel electrolytes and provide a typical practicable high-performance gel electrolyte material for solid electrochromic devices. Keywords: electrochromic device, gel electrolytes, naphthalene diimide, succinonitrile

Abstract:
Alfalfa is renowned for its nutritional and biopharmaceutical value as a perennial forage legume. However, establishing a rapid plant regeneration protocol using somatic embryogenesis and efficient transformation frequency are the crucial prerequisites for gene editing in alfalfa. This study was undertaken to establish and improve the protocol for somatic embryogenesis and subsequent plant regeneration. The experiments were conducted in response to natural sensitivity using various antibiotics such as cefotaxime, carbenicillin, gentamycin, hygromycin, and kanamycin. Using 3-week-old leaf tissue, somatic embryogenesis was initiated on Gamborg’s B5 basal (B5H) medium supplemented with 3% maltose, 0.9µM Kinetin, and 4.5µM 2,4-D. Embryogenic callus (EC) obtained from the B5H medium exhibited a high rate of somatic embryo formation (97.9%) after 3 weeks when the cultures were placed in the dark. Different developmental stages of somatic embryos and cotyledonary stages were then transferred to Murashige and Skoog’s (MS) basal medium under light, resulting in a 94% regeneration rate of plantlets. Our results indicate that leaf segments can grow (tolerate) up to 450 mg/L of cefotaxime and 400 mg/L of carbenicillin in the culture medium. However, the survival threshold for hygromycin at 12.5 mg/L, kanamycin at 250 mg/L, gentamycin at 50 mg/L, and timentin (300 mg/L). The experiment to improve the protocol for achieving efficient transient gene expression in alfalfa through genetic transformation with the Agrobacterium tumefaciens pCAMBIA1304 vector was also conducted. The vector contains two reporter genes such as β-glucuronidase (GUS) and green fluorescent protein (GFP), along with a selectable hygromycin B phosphotransferase gene (HPT), all driven under the CaMV 35s promoter. Various transformation parameters were optimized using 3-week-old in vitro-grown plantlets. The different parameters such as types of explant, leaf ages, preculture days, segment sizes, wounding types, bacterial concentrations, infection periods, co-cultivation periods, different concentrations of acetosyringone, silver nitrate, and calcium chloride were optimized for transient gene expression. The transient gene expression was confirmed via histochemical GUS and GFP visualization under fluorescent microscopy. The data were analyzed based on the semi-quantitative observation of the percentage and number of blue GUS spots on different days of agro-infection. The highest percentage of GUS positivity (76.2%) was observed in 3-week-old leaf segments wounded using a scalpel blade of 11 size- after 3 days of post-incubation at a bacterial concentration of 0.6, with 2 days of preculture, 30 min of bacterial-leaf segment co-cultivation, with the addition of 150 µM acetosyringone, 4 mM calcium chloride, and 75 µM silver nitrate. Our results suggest that various factors influence T-DNA delivery in the Agrobacterium-mediated transformation of alfalfa. The stable gene expression in the putative transgenic tissue was confirmed using PCR amplification of both marker genes, indicating that gene expression in explants was not solely due to Agrobacterium, but also from transformed cells. The improved protocol could be used for generating transgenic alfalfa plants using genome editing techniques such as CRISPR/Cas9.Keywords: Medicago sativa l. Procedia PDF Downloads 141752 A Novel Alginate/Tea Waste Complex for Restoration and Conservation of Historical Textiles Using Immobilized Enzymes
Authors: Mohamed E. Hassan
Abstract:
Through numerous chemical linkages, historical textiles in burial contexts or in museums are exposed to many different forms of stains and filth. The cleaning procedure must be carried out carefully without causing any irreparable harm, and sediments must be removed without damaging the surface's original material. Science and technology continue to develop novel methods for cleaning historical textiles and artistic surfaces biologically (using enzymes). Lipase and α-amylase were immobilized on nanoparticles of alginate/tea waste nanoparticle complex and used in historical textile cleaning. The preparation of nanoparticles, activation, and enzyme immobilization were characterized. Optimization of loading times and units of the two enzymes was done. It was found that the optimum time and units of amylase were 3 hours and 30 U, respectively. While the optimum time and units of lipase were 2.5 hours and 20 U, respectively, FT-IR and TGA instruments were used in proving the preparation of nanoparticles and the immobilization process. SEM was used to examine the fibres before and after treatment. In conclusion, a new carrier was prepared from alginate/Tea waste and optimized to be used in the restoration and conservation of historical textiles using immobilized lipase and α-amylase.Keywords: alginate/tea waste, nanoparticles, immobilized enzymes, historical textiles

Abstract:
Gate Assignment Problem (GAP) is one of the most substantial issues in airport operation. In principle, GAP intends to maintain the maximum capacity of the airport through the best possible allocation of the resources (gates) in order to reach the optimum outcome. The problem involves a wide range of dependent and independent resources and their limitations, which add to the complexity of GAP from both theoretical and practical perspective. In this study, GAP was mathematically formulated as a three-objective problem. The preliminary goal of multi-objective formulation was to address a higher number of objectives that can be simultaneously optimized and therefore increase the practical efficiency of the final solution. The problem is solved by applying the second version of Non-dominated Sorting Genetic Algorithm (NSGA-II). Results showed that the proposed mathematical model could address most of major criteria in the decision-making process in airport management in terms of minimizing both airport/airline cost and passenger walking distance time. Moreover, the proposed approach could properly find acceptable possible answers. Keywords: airport management, gate assignment problem, mathematical modeling, genetic algorithm, NSGA-II
