Authors: M. Bertsiou, E. Feloni, E. Baltas
Abstract:
The proper management of the water supply and electricity is the key issue, especially in small islands, where sustainability has been combined with the autonomy and covering of water needs and the fast development in potential sectors of economy. In this research work a hybrid system in Fournoi island (Icaria), a small island of Aegean, has been evaluated in order to produce hydropower and cover water demands, as it can provide solutions to acute problems, such as the water scarcity or the instability of local power grids. The meaning and the utility of hybrid system and the cooperation with a desalination plant has also been considered. This kind of project has not yet been widely applied, so the consideration will give us valuable information about the storage of water and the controlled distribution of the generated clean energy. This process leads to the conclusions about the functioning of the system and the profitability of this project, covering the demand for water and electricity. Keywords: hybrid system, water, electricity, island

Authors: Sarwan Dhir, Suma Basak, Dipika Parajulee
Abstract:
Alfalfa is renowned for its nutritional and biopharmaceutical value as a perennial forage legume. However, establishing a rapid plant regeneration protocol using somatic embryogenesis and efficient transformation frequency are the crucial prerequisites for gene editing in alfalfa. This study was undertaken to establish and improve the protocol for somatic embryogenesis and subsequent plant regeneration. The experiments were conducted in response to natural sensitivity using various antibiotics such as cefotaxime, carbenicillin, gentamycin, hygromycin, and kanamycin. Using 3-week-old leaf tissue, somatic embryogenesis was initiated on Gamborg’s B5 basal (B5H) medium supplemented with 3% maltose, 0.9µM Kinetin, and 4.5µM 2,4-D. Embryogenic callus (EC) obtained from the B5H medium exhibited a high rate of somatic embryo formation (97.9%) after 3 weeks when the cultures were placed in the dark. Different developmental stages of somatic embryos and cotyledonary stages were then transferred to Murashige and Skoog’s (MS) basal medium under light, resulting in a 94% regeneration rate of plantlets. Our results indicate that leaf segments can grow (tolerate) up to 450 mg/L of cefotaxime and 400 mg/L of carbenicillin in the culture medium. However, the survival threshold for hygromycin at 12.5 mg/L, kanamycin at 250 mg/L, gentamycin at 50 mg/L, and timentin (300 mg/L). The experiment to improve the protocol for achieving efficient transient gene expression in alfalfa through genetic transformation with the Agrobacterium tumefaciens pCAMBIA1304 vector was also conducted. The vector contains two reporter genes such as β-glucuronidase (GUS) and green fluorescent protein (GFP), along with a selectable hygromycin B phosphotransferase gene (HPT), all driven under the CaMV 35s promoter. Various transformation parameters were optimized using 3-week-old in vitro-grown plantlets. The different parameters such as types of explant, leaf ages, preculture days, segment sizes, wounding types, bacterial concentrations, infection periods, co-cultivation periods, different concentrations of acetosyringone, silver nitrate, and calcium chloride were optimized for transient gene expression. The transient gene expression was confirmed via histochemical GUS and GFP visualization under fluorescent microscopy. The data were analyzed based on the semi-quantitative observation of the percentage and number of blue GUS spots on different days of agro-infection. The highest percentage of GUS positivity (76.2%) was observed in 3-week-old leaf segments wounded using a scalpel blade of 11 size- after 3 days of post-incubation at a bacterial concentration of 0.6, with 2 days of preculture, 30 min of bacterial-leaf segment co-cultivation, with the addition of 150 µM acetosyringone, 4 mM calcium chloride, and 75 µM silver nitrate. Our results suggest that various factors influence T-DNA delivery in the Agrobacterium-mediated transformation of alfalfa. The stable gene expression in the putative transgenic tissue was confirmed using PCR amplification of both marker genes, indicating that gene expression in explants was not solely due to Agrobacterium, but also from transformed cells. The improved protocol could be used for generating transgenic alfalfa plants using genome editing techniques such as CRISPR/Cas9.Keywords: Medicago sativa l. Procedia PDF Downloads 3601777 Competitive DNA Calibrators as Quality Reference Standards (QRS™) for Germline and Somatic Copy Number Variations/Variant Allelic Frequencies Analyses
Authors: Eirini Konstanta, Cedric Gouedard, Aggeliki Delimitsou, Stefania Patera, Samuel Murray
Abstract:
Introduction: Quality reference DNA standards (QRS) for molecular testing by next-generation sequencing (NGS) are essential for accurate quantitation of copy number variations (CNV) for germline and variant allelic frequencies (VAF) for somatic analyses. Objectives: Presently, several molecular analytics for oncology patients are reliant upon quantitative metrics. Test validation and standardisation are also reliant upon the availability of surrogate control materials allowing for understanding test LOD (limit of detection), sensitivity, specificity. We have developed a dual calibration platform allowing for QRS pairs to be included in analysed DNA samples, allowing for accurate quantitation of CNV and VAF metrics within and between patient samples. Methods: QRS™ blocks up to 500nt were designed for common NGS panel targets incorporating ≥ 2 identification tags (IDTDNA.com). These were analysed upon spiking into gDNA, somatic, and ctDNA using a proprietary CalSuite™ platform adaptable to common LIMS. Results: We demonstrate QRS™ calibration reproducibility spiked to 5–25% at ± 2.5% in gDNA and ctDNA. Furthermore, we demonstrate CNV and VAF within and between samples (gDNA and ctDNA) with the same reproducibility (± 2.5%) in a clinical sample of lung cancer and HBOC (EGFR and BRCA1, respectively). CNV analytics was performed with similar accuracy using a single pair of QRS calibrators when using multiple single targeted sequencing controls. Conclusion: Dual paired QRS™ calibrators allow for accurate and reproducible quantitative analyses of CNV, VAF, intrinsic sample allele measurement, inter and intra-sample measure not only simplifying NGS analytics but allowing for monitoring clinically relevant biomarker VAF across patient ctDNA samples with improved accuracy. Keywords: calibrator, CNV, gene copy number, VAF

Authors: Jinkoo Kim, Minsung Kim
Abstract:
This study develops a hybrid seismic energy dissipation device composed of a viscoelastic damper and a steel slit damper connected in parallel. A cyclic loading test is conducted on a test specimen to validate the seismic performance of the hybrid damper. Then a moment-framed model structure is designed without seismic load so that it is retrofitted with the hybrid dampers. The model structure is transformed into an equivalent simplified system to find out optimum story-wise damper distribution pattern using genetic algorithm. The effectiveness of the hybrid damper is investigated by fragility analysis and the life cycle cost evaluation of the structure with and without the dampers. The analysis results show that the model structure has reduced probability of reaching damage states, especially the complete damage state, after seismic retrofit. The expected damage cost and consequently the life cycle cost of the retrofitted structure turn out to be significantly small compared with those of the original structure. Procedia PDF Downloads 5101773 MSIpred: A Python 2 Package for the Classification of Tumor Microsatellite Instability from Tumor Mutation Annotation Data Using a Support Vector Machine
Authors: Chen Wang, Chun Liang
Abstract:
Microsatellite instability (MSI) is characterized by high degree of polymorphism in microsatellite (MS) length due to a deficiency in mismatch repair (MMR) system. MSI is associated with several tumor types and its status can be considered as an important indicator for tumor prognostic. Conventional clinical diagnosis of MSI examines PCR products of a panel of MS markers using electrophoresis (MSI-PCR) which is laborious, time consuming, and less reliable. MSIpred, a python 2 package for automatic classification of MSI was released by this study. It computes important somatic mutation features from files in mutation annotation format (MAF) generated from paired tumor-normal exome sequencing data, subsequently using these to predict tumor MSI status with a support vector machine (SVM) classifier trained by MAF files of 1074 tumors belonging to four types. Evaluation of MSIpred on an independent 358-tumor test set achieved overall accuracy of over 98% and area under receiver operating characteristic (ROC) curve of 0.967. These results indicated that MSIpred is a robust pan-cancer MSI classification tool and can serve as a complementary diagnostic to MSI-PCR in MSI diagnosis. Keywords: microsatellite instability, pan-cancer classification, somatic mutation, support vector machine

Authors: L. G. Karancsi
Abstract:
The research was set up on chernozem soil at the Látókép AGTC MÉK research area of the University of Debrecen in Hungary. We examined the yield, the yield production per 1kg NPK fertilizer and the water and nutrient utilization of hybrid PR37N01 and PR37M81 in 2013. We found that PR37N01 produced the most yield at the level of N120+P (17,476kg ha-1) while PR37M81 reached the highest yield at level N150+PK (16,754kg ha-1). Studies related to yield production per 1kg NPK indicated that the best results were achieved at level N30+PK compared to the control treatment. Yield production per 1kg NPK was17.6kg kg-1 by P37N01 and 44.2kg kg-1 by PR37M81. By comparing the water utilization of hybrids we found that the worst water utilization results were reached in the control treatment (PR37N01: 26.2kg mm-1, PR37M81: 19.5kg mm-1). The best water utilization values were produced at level N120+PK in the case of hybrid PR37N01 (32.1kg mm-1) and at N150+PK in the case of hybrid PR37M81 (30.8kg mm-1). We established the values of the nutrient reaction and the fertilizer optimum of hybrids. We discovered a strong relationship between the amount of fertilizer applied and the yield produced (r2= 0.8228–0.9515). The best nutrient response was induced by hybrid PR37N01, while the weakest results were reached by hybrid PR37M81.Keywords: hybrid, maize, nutrient, yield, water utilization

Authors: Mark Schudeleit, Christian Sieg, Ferit Küçükay
Abstract:
This paper describes how to dimension the electric components of a 48V hybrid system considering real customer use. Furthermore, it provides information about savings in energy and CO2 emissions by a customer-tailored 48V hybrid. Based on measured customer profiles, the electric units such as the electric motor and the energy storage are dimensioned. Furthermore, the CO2 reduction potential in real customer use is determined compared to conventional vehicles. Finally, investigations are carried out to specify the topology design and preliminary considerations in order to hybridize a conventional vehicle with a 48V hybrid system. The emission model results from an empiric approach also taking into account the effects of engine dynamics on emissions. We analyzed transient engine emissions during representative customer driving profiles and created emission meta models. The investigation showed a significant difference in emissions when simulating realistic customer driving profiles using the created verified meta models compared to static approaches which are commonly used for vehicle simulation. Keywords: customer use, dimensioning, hybrid electric vehicles, vehicle simulation, 48V hybrid system

Authors: Ebrahim Asadi-Gangraj
Abstract:
Hybrid flow shop scheduling problem (HFS) contains sequencing in a flow shop where, at any stage, there exist one or more related or unrelated parallel machines. This production system is a common manufacturing environment in many real industries, such as the steel manufacturing, ceramic tile manufacturing, and car assembly industries. In this research, a mixed integer linear programming (MILP) model is presented for the hybrid flow shop scheduling problem, in which, the objective consists of minimizing the maximum completion time (makespan). For this purpose, a Benders Decomposition (BD) method is developed to solve the research problem. The proposed approach is tested on some test problems, small to moderate scale. The experimental results show that the Benders decomposition approach can solve the hybrid flow shop scheduling problem in a reasonable time, especially for small and moderate-size test problems. Keywords: hybrid flow shop, mixed integer linear programming, Benders decomposition, makespan
Procedia PDF Downloads 1881762 Parametric Study for Optimal Design of Hybrid Bridge Joint
Authors: Bongsik Park, Jae Hyun Park, Jae-Yeol Cho
Abstract:
Mixed structure, which is a kind of hybrid system, is incorporating steel beam and prestressed concrete beam. Hybrid bridge adopting mixed structure have some merits. Main span length can be made longer by using steel as main span material. In case of cable-stayed bridge having asymmetric span length, negative reaction at side span can be restrained without extra restraining devices by using weight difference between main span material and side span material. However angle of refraction might happen because of rigidity difference between materials and stress concentration also might happen because of abnormal loading transmission at joint in the hybrid bridge. Therefore the joint might be a weak point of the structural system and it needs to pay attention to design of the joint. However, design codes and standards about the joint in the hybrid-bridge have not been established so the joint designs in most of construction cases have been very conservative or followed previous design without extra verification. In this study parametric study using finite element analysis for optimal design of hybrid bridge joint is conducted. Before parametric study, finite element analysis was conducted based on previous experimental data and it is verified that analysis result approximated experimental data. Based on the finite element analysis results, parametric study was conducted. The parameters were selected as those have influences on joint behavior. Based on the parametric study results, optimal design of hybrid bridge joint has been determined. Keywords: parametric study, optimal design, hybrid bridge, finite element analysis

Procedia PDF Downloads 3591756 The Economic Value of Mastitis Resistance in Dairy Cattle in Kenya
Authors: Caleb B. Sagwa, Tobias O. Okeno, Alexander K. Kahi
Abstract:
Dairy cattle production plays an important role in the Kenyan economy. However, high incidences of mastitis is a major setback to the productivity in this industry. The current dairy cattle breeding objective in Kenya does not include mastitis resistance, mainly because the economic value of mastitis resistance has not been determined. Therefore this study aimed at estimating the economic value of mastitis resistance in dairy cattle in Kenya. Initial input parameters were obtained from literature on dairy cattle production systems in the tropics. Selection index methodology was used to derive the economic value of mastitis resistance. Somatic cell count (SCC) was used an indicator trait for mastitis resistance. The economic value was estimated relative to milk yield (MY). Economic values were assigned to SCC in a selection index such that the overall gain in the breeding goal trait was maximized. The option of estimating the economic value for SCC by equating the response in the trait of interest to its index response was considered. The economic value of mastitis resistance was US $23.64 while maximum response to selection for MY was US $66.01. The findings of this study provide vital information that is a pre-requisite for the inclusion of mastitis resistance in the current dairy cattle breeding goal in Kenya. Keywords: somatic cell count, milk quality, payment system, breeding goal

Abstract:
Algerian cities are confronted with large quantities of waste generated by the disposal of household and similar residues in technical landfills (CET), such as the one in the location of Batna. The interaction between waste components and incoming water generates leachates rich in organic matter and trace elements, which require treatment before discharge. The aim of this study was to propose an effective process for treating the leachates, which were subjected to an initial chemical treatment using the (H₂O₂/UV) system. Optimal treatment conditions were determined at [H₂O₂] of 0.3 M and pH of 8.6. Next, two hybrid biological treatment systems were applied: hybrid system I (H₂O₂/UV/bacteria) and hybrid system II (H₂O₂/UV/bacteria/microalgae). The three processes resulted in the following degradation rates, expressed in terms of total organic carbon (TOC) 27.4% for the (H₂O₂/UV) system; 58.1% for the hybrid system I (H₂O₂/UV/Bacteria); 67.86% for the hybrid system II (H₂O₂/UV/Bacteria/Microalgae). This study demonstrates that a hybrid approach combining advanced oxidation processes and biological treatments is a highly effective alternative to achieve satisfactory treatment. Keywords: leachate, landfill, advanced oxidation processes, biological treatment, bacteria, microalgae, total organic carbon
Procedia PDF Downloads 691754 Efficient Mercury Sorbent: Activated Carbon and Metal Organic Framework Hybrid
Authors: Yongseok Hong, Kurt Louis Solis
Abstract:
In the present study, a hybrid sorbent using the metal organic framework (MOF), UiO-66, and powdered activated carbon (pAC) is synthesized to remove cationic and anionic metals simultaneously. UiO-66 is an octahedron-shaped MOF with a Zr₆O₄(OH)₄ metal node and 1,4-benzene dicarboxylic acid (BDC) organic linker. Zr-based MOFs are attractive for trace element remediation in wastewaters, because Zr is relatively non-toxic as compared to other classes of MOF and, therefore, it will not cause secondary pollution. Most remediation studies with UiO-66 target anions such as fluoride, but trace element oxyanions such as arsenic, selenium, and antimony have also been investigated. There have also been studies involving mercury removal by UiO-66 derivatives, however these require post-synthetic modifications or have lower effective surface areas. Activated carbon is known for being a readily available, well-studied, effective adsorbent for metal contaminants. Solvothermal method was employed to prepare hybrid sorbent from UiO66 and activated carbon, which could be used to remove mercury and selenium simultaneously. The hybrid sorbent was characterized using FSEM-EDS, FT-IR, XRD, and TGA. The results showed that UiO66 and activated carbon are successfully composited. From BET studies, the hybrid sorbent has a SBET of 1051 m² g⁻¹. Adsorption studies were performed, where the hybrid showed maximum adsorption of 204.63 mg g⁻¹ and 168 mg g⁻¹ for Hg (II) and selenite, respectively, and follows the Langmuir model for both species. Kinetics studies have revealed that the Hg uptake of the hybrid is pseudo-2nd order and has rate constant of 5.6E-05 g mg⁻¹ min⁻¹ and the selenite uptake follows the simplified Elovich model with α = 2.99 mg g⁻¹ min⁻¹, β = 0.032 g mg⁻¹.Keywords: adsorption, flue gas wastewater, mercury, selenite, metal organic framework

Abstract:
Discrete Event Simulation (DES) and Agent Based Simulation (ABS) are two simulation approaches that have been proposed to support decision-making in the construction industry. Despite the wide use of these simulation approaches in the construction field, their applications for production and material planning is still limited. This is largely due to the dynamic and complex nature of construction material supply chain systems. Moreover, managing the flow of construction material is not well integrated with site logistics in traditional construction planning methods. This paper presents a hybrid of DES and ABS to simulate on-site and off-site material supply processes. DES is applied to determine the best production scenarios with information of on-site production systems, while ABS is used to optimize the supply chain network. A case study of a construction piling project in New Zealand is presented illustrating the potential benefits of using the proposed hybrid simulation model in construction material flow planning. The hybrid model presented can be used to evaluate the impact of different decisions on construction supply chain management. Keywords: construction supply-chain management, simulation modeling, decision-support tools, hybrid simulation

Abstract:
This work is focused on the preparation and characterization of poly (vinyl alcohol)/silica gel/Nano-TiO₂, and the study of titanium dioxide (TiO₂) nanoparticles 1% on the properties of poly (vinyl alcohol) (PVA)/silica films. Fourier transform infrared (FT-IR), water contact angle, ultraviolet-visible spectrometry (UV-VIS)) were used to characterize the hybrid films obtained. The PVA/SiO₂/Nano-TiO₂ films were successfully synthesized. Owing to the FT-IR Analysis, the chemical bonds have clearly shown that the PVA backbone is linked to the (SiO₂-TiO₂) network. UV-VIS tests indicated that the hybrid films' UV shielding properties were drastically enhanced as a result of the addition of TiO₂. The water contact angle results revealed that TiO₂ nanoparticles used as a doping compound possess an important influence on the hydrophilicity of PVA/SiO₂ as thin films. Keywords: sol-gel method, hybrid materials, PVA/SIO₂/TiO₂, spectroscopical characterization
