2015 Immature Palm Tree Detection Using Morphological Filter for Palm Counting with High Resolution Satellite Image

Abstract:
Accurate inventories of oil palm planted areas are crucial for plantation management as this would impact the overall economy and production of oil. One of the technological advancements in the oil palm industry is semi-automated palm counting, which is replacing conventional manual palm counting via digitizing aerial imagery. Most of the semi-automated palm counting method that has been developed was limited to mature palms due to their ideal canopy size represented by satellite image. Therefore, immature palms were often left out since the size of the canopy is barely visible from satellite images. In this paper, an approach using a morphological filter and high-resolution satellite image is proposed to detect immature palm trees. This approach makes it possible to count the number of immature oil palm trees. The method begins with an erosion filter with an appropriate window size of 3m onto the high-resolution satellite image. The eroded image was further segmented using watershed segmentation to delineate immature palm tree regions. Then, local minimum detection was used because it is hypothesized that immature oil palm trees are located at the local minimum within an oil palm field setting in a grayscale image. The detection points generated from the local minimum are displaced to the center of the immature oil palm region and thinned. Only one detection point is left that represents a tree. The performance of the proposed method was evaluated on three subsets with slopes ranging from 0 to 20° and different planting designs, i.e., straight and terrace. The proposed method was able to achieve up to more than 90% accuracy when compared with the ground truth, with an overall F-measure score of up to 0.91.Keywords: immature palm count, oil palm, precision agriculture, remote sensing

Abstract:
Mechanical index (MI) is used for quantifying acoustic cavitation and the relationship between acoustic pressure and the frequency. In this study, modeling of the MI was applied to provide treatment protocol and to understand the effective physical processes on reproducibility of stem cells. The acoustic pressure and MI equations are modeled and solved to estimate optimal MI for 28, 40, 150 kHz and 1 MHz frequencies. Radial and axial acoustic pressure distribution was extracted. To validate the results of the modeling, the acoustic pressure in the water and near field depth was measured by a piston hydrophone. Results of modeling and experiments show that the model is consistent well to experimental results with 0.91 and 0.90 correlation of coefficient (p<0.05) for 1 MHz and 40 kHz. Low intensity ultrasound with 0.40 MI is more effective on the proliferation rate of the spermatogonial stem cells during the seven days of culture, in contrast, high MI has a harmful effect on the spermatogonial stem cells. This model provides proper treatment planning in vitro and in vivo by estimating the cavitation phenomenon.Keywords: ultrasound, mechanical index, modeling, stem cell

Abstract:
The present study was conducted to investigate the relationship between career anchors and job satisfaction with emphasis on the mediating role of person-job fit. 502 managers and supervisors of ten operational areas of a large energy Company were selected as a cluster sample appropriate to the volume. The instruments used in this study were Career Anchor Questionnaire, Job Satisfaction Questionnaire and Person-job fit Questionnaire. Pearson correlation coefficient was used to analyze the data and AMOS software was used to determine the effect of career anchor variables and person-job fit on job satisfaction. Anchors of service and dedication, pure challenge and security and stability increase the person-job fit among managers and also the person-job fit plays a mediating role in relation to the effect it has on job satisfaction through these anchors. In contrast, the anchors of independence and autonomy reduce the person-job fit. Considering the importance of positive organizational attitudes and in order to have an optimal fit between job and worker, it is better that in human resources processes such as hiring and employing, the career anchors of the person should be considered so that the person can have more job satisfaction; and thus bring higher productivity for themselves and the organization.Keywords: career anchor, job satisfaction, person-job fit, energy company, managers

Abstract:
We present a simulation of a HEMT (high electron mobility transistor) structure with and without a field plate. We extract the device characteristics through the analysis of DC, AC and high frequency regimes, as shown in this paper. This work demonstrates the optimal device with a gate length of 15 nm, InAlN/GaN heterostructure and field plate structure, making it superior to modern HEMTs when compared with otherwise equivalent devices. This improves the ability to bear the burden of the current density passes in the channel. We have demonstrated an excellent current density, as high as 2.05 A/m, a peak extrinsic transconductance of 0.59S/m at VDS=2 V, and cutting frequency cutoffs of 638 GHz in the first HEMT and 463 GHz for Field plate HEMT., maximum frequency of 1.7 THz, maximum efficiency of 73%, maximum breakdown voltage of 400 V, leakage current density IFuite=1 x 10-26 A, DIBL=33.52 mV/V and an ON/OFF current density ratio higher than 1 x 1010. These values were determined through the simulation by deriving genetic and Monte Carlo algorithms that optimize the design and the future of this technology.Keywords: HEMT, silvaco, field plate, genetic algorithm, quantum

This paper presents the electrical effect of two neighboring distribution substation during the construction phase. The size of auxiliary grounding grid have an effect on entire grounding system. The bigger the size of auxiliary grounding grid, the lower the GPR and maximum touch voltage, with the exception that when the two grids are unconnected, i.e. the bigger the size of auxiliary grounding grid, the higher the maximum step voltage. The results in this paper could be served as design guideline of grounding system, and perhaps remedy of some troublesome grounding grids in power distribution’s system. Modeling and simulation is carried out on the Current Distribution Electromagnetic interference Grounding and Soil structure (CDEGS) program. Abstract:
The objective of this study was to develop a method to pretreat sugarcane leaf waste using microwave-assisted (MA) inorganic salt. The effects of process parameters of salt concentration, microwave power intensity and pretreatment time on reducing sugar yield from enzymatically hydrolysed sugarcane leaf waste were investigated. Pretreatment models based on MA-NaCl, MA-ZnCl2 and MA-FeCl3 were developed. Maximum reducing sugar yield of 0.406 g/g was obtained with 2 M FeCl3 at 700W for 3.5 min. Scanning electron microscopy (SEM) and Fourier Transform Infrared analysis (FTIR) showed major changes in lignocellulosic structure after MA-FeCl3 pretreatment with 71.5 % hemicellulose solubilization. This pretreatment was further assessed on sorghum leaves and Napier grass under optimal MA-FeCl3 conditions. A 2 fold and 3.1-fold increase in sugar yield respectively were observed compared to previous reports. This pretreatment was highly effective for enhancing enzymatic saccharification of lignocellulosic biomass.Keywords: acid, pretreatment, salt, sugarcane leaves

Abstract:
Wearable biosensor systems can be designed and developed for health monitoring. There is much interest in both scientific and industrial communities established since 2007. Fundamentally, the cost of healthcare has increased dramatically and the world population is aging. That creates the need to harvest technological improvements with small bio-sensing devices, wireless-communication, microelectronics and smart textiles, that leads to non-stop developments of wearable sensor based systems. There has been a significant demand to monitor patient's health status while the patient leaves the hospital in his/her personal environment. To address this need, there are numerous system prototypes which has been launched in the medical market recently, the aim of that is to provide real time information feedback about patient's health status, either to the patient himself/herself or direct to the supervising medical centre station, while being capable to give a notification for the patient in case of possible imminent health threatening conditions. Furthermore, wearable health monitoring systems comprise new techniques to address the problem of managing and monitoring chronic heart diseases for elderly people. Wearable sensor systems for health monitoring include various types of miniature sensors, either wearable or implantable. To be specific, our proposed system able to measure essential physiological parameter, such as heart rate signal which could be transmitted through Bluetooth to the cloud server in order to store, process, analysis and visualise the data acquisition. Abstract:
Determination of land use changing is an important component of regional planning for applications ranging from urban fringe change detection to monitoring change detection of land use. This data are very useful for natural resources management.On the other hand, the technologies and methods of change detection also have evolved dramatically during past 20 years. So it has been well recognized that the change detection had become the best methods for researching dynamic change of land use by multi-temporal remotely-sensed data. The objective of this paper is to assess, evaluate and monitor land use change surrounding the area of Najran city, Kingdom of Saudi Arabia (KSA) using Landsat images (June 23, 2009) and ETM+ image(June. 21, 2014). The post-classification change detection technique was applied. At last,two-time subset images of Najran city are compared on a pixel-by-pixel basis using the post-classification comparison method and the from-to change matrix is produced, the land use change information obtained.Three classes were obtained, urban, bare land and agricultural land from unsupervised classification method by using Erdas Imagine and ArcGIS software. Accuracy assessment of classification has been performed before calculating change detection for study area. The obtained accuracy is between 61% to 87% percent for all the classes. Change detection analysis shows that rapid growth in urban area has been increased by 73.2%, the agricultural area has been decreased by 10.5 % and barren area reduced by 7% between 2009 and 2014. The quantitative study indicated that the area of urban class has unchanged by 58.2 km〗^2, gained 70.3 〖km〗^2 and lost 16 〖km〗^2. For bare land class 586.4〖km〗^2 has unchanged, 53.2〖km〗^2 has gained and 101.5〖km〗^2 has lost. While agriculture area class, 20.2〖km〗^2 has unchanged, 31.2〖km〗^2 has gained and 37.2〖km〗^2 has lost.Keywords: land use, remote sensing, change detection, satellite images, image classification

Abstract:
Rapid urbanization significantly change land use, urban nature, land surface vegetation cover, and heat distribution, leading to the formation of urban heat island (UHI) effect and affecting the healthy growth of cities and the comfort of human living style. Past information and present changes in Land Surface Temperature (LST) and urban landscapes could be useful to geographers, environmentalists, and urban planners in an attempt to shape the urban development process and mitigate the effects of urban heat islands (UHI). This study aims at using Satellite Remote Sensing (SRS) and GIS techniques to develop an approach for assessing the urban nature and UHI effects in Lahore, Pakistan. The study employed the Radiative Transfer Method (RTM) in estimating LST to assess the SUHI effect during the interval of 20 years (2000-2020). The assessment was performed by the available Landsat 7/ETM+ and Landsat 8/OIL_TIRs data for the years 2000, 2010, and 2020 respectively. Pearson’s correlation and normalized mutual information were applied to investigate the relationship between green space characteristics and LST. The result of this work revealed that the influence of urban heat island is not always at the city centers but sometimes in the outskirt where a lot of development activities were going on towards the direction of expansion of Lahore, Pakistan. The present study explores the usage of image processing and spatial analysis in the drive towards achieving urban greening of Lahore and a sustainable urban environment in terms of urban planning, policy, and decision making and promoting the healthy and sustainable urban environment of the city.Keywords: urban nature, urban heat islands, urban green space, land use, Lahore

Abstract:
A design problem of non-uniform circular antenna arrays for maximum reduction of both the side lobe level (SLL) and first null beam width (FNBW) is dealt with. This problem is modeled as a simple optimization problem. The method of Firefly algorithm (FFA) is used to determine an optimal set of current excitation weights and antenna inter-element separations that provide radiation pattern with maximum SLL reduction and much improvement on FNBW as well. Circular array antenna laid on x-y plane is assumed. FFA is applied on circular arrays of 8-, 10-, and 12- elements. Various simulation results are presented and hence performances of side lobe and FNBW are analyzed. Experimental results show considerable reductions of both the SLL and FNBW with respect to those of the uniform case and some standard algorithms GA, PSO, and SA applied to the same problem.Keywords: circular arrays, first null beam width, side lobe level, FFA

Abstract:
In this study, an Organic Rankine Cycle (ORC) with Cyclohexane working fluid is proposed for cogeneration in the cement industry. In this regard: first, a parametric study is conducted to evaluate the effects of some key parameters on the system performances. Next, single and multi-objective optimizations are performed to achieve the system optimal design. The optimization considers the exergy efficiency, the cost per exergy unit and the environmental impact of the net produced power as objective functions. Finally, exergy, exergoeconomic and exergoenvironmental analysis of the cycle is carried out at the optimum operating conditions. Abstract:
Diabetes mellitus is still known as one of diseases which give a big amount of death in the world. From 2012 to 2014, diabetes is estimated to have resulted in 1.5 to 4.9 million deaths each year. In this paper, we present our research in the analysis and dose determination of Tenebrio molitor (Mealworm) extract as an anti-diabetic agent which is believed by Indonesian people as a traditional treatment to prevent and treat diabetes. We found that Tenebrio molitor extract has a potential as an anti-diabetic agent by in vitro test to Mus musculus which were divided into six group of treatment. Our dose determination analysis gave a conclusion that at 2,5 g/mL of concentration of the extract would give the optimal result in healing a wound given to Mus musculus which were induced by aloxane monohydrate. Abstract:
Modular multilevel converters (MMC) exhibit a highly scalable and modular characteristic with good voltage/power expansion, fault tolerance capability, low output harmonic content, good redundancy, and a flexible front-end configuration. Fault detection, location, and isolation, as well as maintaining fault ride-through (FRT), are major challenges to MMC reliability and power supply sustainability. Different papers have been reviewed to seek the best MMC configuration with fault capability. DC faults are the most common fault, while the probability that AC fault occurs in a modular multilevel converter (MCC) is low; though, AC faults consequence are severe. This paper reviews several MMC topologies and modulation techniques in tackling faults. These fault control strategies are compared based on cost, complexity, controllability, and power loss. A meshed network of half-bridge (HB) MMC topology was optimal in rendering fault ride through than any other MMC topologies but only when combined with DC circuit breakers (CBS), AC CBS, and fault current limiters (FCL).Keywords: MMC-HVDC, DC faults, fault current limiters, control scheme

Abstract:
Kecombrang is a Zingiberaceae plant which has antioxidant properties. The high antioxidant content in kecombrang flowers has the potential to be processed as a functional beverage raw material so that it can be used as an ingredient in making herbal teas. The purpose of this study was to determine the chemical components, physicochemistry, antioxidant activity and sensory characteristics of kecombrang tea. The research methodology was carried out by using a completely randomized design with processing factors of kecombrang tea namely blanching and non-blanching, fermentation and non-fermentation, and the optimal time for drying kecombrang tea. The best treatment combination based on the effective index method is the treatment of the blanching process followed by drying at a temperature of 50ᵒC until the 2% moisture content can produce kecombrang tea with a total phenol content of 5.95 mg Tannic Acid Equivalent (TAE) / gram db, total flavonoid 3%, pH 4.5, and antioxidant activity 82.95%, red color, distinctive aroma of tea, fresh taste, and preferred by panelists.Keywords: kecombrang tea, blanching, fermentation, total phenol, and antioxidant activity

The present study investigated the attitude of university students towards semester system and teachers with a specific objective of finding problems faced by students in semester system. 10 focus group discussions were conducted among students in five Universities of Hazara Division of KPK regarding their knowledge and attitudes about semester system and problems they faced due to this system and teacher’s attitude. The key findings were the problems like favoritism, gender biased ness, racial biased ness, biased ness in marking, relative marking, harassment, using students for personal tasks and authoritarian attitude from teachers’ side and the heavy tasks in less time which are causing stress among students. It was recommended that proper training and monitoring system should be maintained for evaluation of teachers to minimize the corruption in this sacred profession and maximize the optimal functioning. Crown enclosure high temperature flares damaged and reduced dimensions crown. Generally crown on EHTF could have a life time up to twenty years. Therefore, this study aims to increase the value of thermal resistance with the effect post treatment on NiCr coated arc spray method. The variation of post treatment temperature, was at 650°C, 750°C, and 850°C. Morphology on the surface and the adhesion strength was analyzed by SEM-EDX, Surface Roughness and Pull - off test. XRD testing was conducted to determine the contained in NiCr coated. Thermal stability of NiCr coated was tested by DSC-TGA. The most optimal results was owned by NiCr coating with post treated at 850°C. It has good thermal stability until 1000°C because of Cr2O3 formation in coated specimen. Growing concerns about the increasing consumption of fossil energy and the improved recognition of environmental protection require sustainable road transportation technology. Electric vehicles (EVs) can contribute to improve environmental sustainability and to solve the energy problem with the right infrastructure. The problem of where to locate electric vehicle charging station can be grouped as decision-making problems because of including many criteria and alternatives that have to be considered simultaneously. The purpose of this paper is to present an integrated AHP and TOPSIS model to rank the optimal sites of EVs charging station in Istanbul, Turkey. Ten different candidate points and three decision criteria are identified. The performances of each candidate points with respect to criteria are obtained according to AHP calculations. These performances are used as an input for TOPSIS method to rank the candidate points. The purpose of economic load dispatch is to allocate the required load demand between the available generation units such that the cost of operation is minimized. It is an optimization problem to find the most economical schedule of the generating units while satisfying load demand and operational constraints. The multiobjective optimization problem in which the engineer’s goal is to maximize or minimize not a single objective function but several objective functions simultaneously. The purpose of multiobjective problems in the mathematical programming framework is to optimize the different objective functions. Many approaches and methods have been proposed in recent years to solve multiobjective optimization problems. Weighting method has been applied to convert multiobjective optimization problems into scalar optimization. MATLAB 7.10 has been used to write the code for the complete algorithm with the help of genetic algorithm (GA). Inspired by endogenous protein motors, researchers have constructed various synthetic DNA motors based on the specificity and predictability of Watson-Crick base pairing. However, the application of DNA motors to signal amplification and biosensing is limited because of low mobility and difficulty in real-time monitoring of the walking process. The objective of our work was to construct a new type of DNA motor termed target-triggered DNA motors that can walk for hundreds of steps in response to a single target binding event. To improve the mobility and processivity of DNA motors, we used gold nanoparticles (AuNPs) as scaffolds to build high-density, three-dimensional tracks. Hundreds of track strands are conjugated to a single AuNP. To enable DNA motors to respond to specific protein and nucleic acid targets, we adapted the binding-induced DNA assembly into the design of the target-triggered DNA motors. In response to the binding of specific target molecules, DNA motors are activated to autonomously walk along AuNP, which is powered by a nicking endonuclease or DNAzyme-catalyzed cleavage of track strands. Each moving step restores the fluorescence of a dye molecule, enabling monitoring of the operation of DNA motors in real time. The motors can translate a single binding event into the generation of hundreds of oligonucleotides from a single nanoparticle. The motors have been applied to amplify the detection of proteins and nucleic acids in test tubes and live cells. The motors were able to detect low pM concentrations of specific protein and nucleic acid targets in homogeneous solutions without the need for separation. Target-triggered DNA motors are significant for broadening applications of DNA motors to molecular sensing, cell imagining, molecular interaction monitoring, and controlled delivery and release of therapeutics.Keywords: biosensing, DNA motors, gold nanoparticles, signal amplification

Abstract:
ᴅ-serine acts as an endogenous co-agonist for N-methyl-ᴅ-aspartate receptors in neuronal synapses. This makes it a key component in the development and function of a healthy brain, especially given its role in several neurodegenerative diseases such as Alzheimer’s disease and dementia. Despite such clear research motivations, the primary site and mechanism of ᴅ-serine release is still currently unclear. For this reason, we are developing a biosensor for the detection of ᴅ-serine utilizing a microelectrode in combination with a ᴅ-amino acid oxidase enzyme, which produces stoichiometric quantities of hydrogen peroxide in response to ᴅ-serine. For the fabrication of a biosensor with good selectivity, we use a permselective poly(meta-phenylenediamine) film to ensure only the target molecule is reacted, according to the size exclusion principle. In this work, we investigated the effect of the electrodeposition conditions used on the biosensor’s response time and selectivity. Careful optimization of the fabrication process allowed for enhanced biosensor response time. This allowed for the real time sensing of ᴅ-serine in a bulk solution, and also provided in means to map the efflux of ᴅ-serine in real time. This was done using scanning electrochemical microscopy (SECM) with the optimized biosensor to measure localized release of ᴅ-serine from an agar filled glass capillary sealed in an epoxy puck, which acted as a model system. The SECM area scan simultaneously provided information regarding the rate of ᴅ-serine flux from the model substrate, as well as the size of the substrate itself. This SECM methodology, which provides high spatial and temporal resolution, could be useful to investigate the primary site and mechanism of ᴅ-serine release in other biological samples.Keywords: ᴅ-serine, enzymatic biosensor, microelectrode, scanning electrochemical microscopy

Abstract:
A great number of existing buildings are designed without seismic design criteria and detailing rules for dissipative structural behavior. Thus, it is of critical importance that the structures that need seismic retrofitting are correctly identified, and an optimal retrofitting is conducted in a cost effective fashion. Among the retrofitting techniques available, steel braces can be considered as one of the most efficient solution among seismic performance upgrading methods of RC structures. This paper investigates the seismic behavior of RC buildings strengthened with different types of steel braces, X-braced, inverted V braced, ZX braced, and Zipper braced. Static non linear pushover analysis has been conducted to estimate the capacity of three story and six story buildings with different brace-frame systems and different cross sections for the braces. It is found that adding braces enhances the global capacity of the buildings compared to the case with no bracing and that the X and Zipper bracing systems performed better depending on the type and size of the cross section.Keywords: seismic design, strengthening, RC frames, steel bracing, pushover analysis

This paper presents an electromagnetic finite element model of permanent magnet synchronous linear motor and distortion rate of the air gap flux density waveform is analyzed in detail. By designing the sample space of the parameters, nonlinear regression modeling of the orthogonal experimental design is introduced. We put forward for possible air gap flux density waveform sine electromagnetic scheme. Parameters optimization of the permanent magnet synchronous linear motor is also introduced which is based on chaotic search and adaptation function. Simulation results prove that the pole shifting does not affect the motor back electromotive symmetry based on the structural parameters, it provides a novel way for the optimum design of permanent magnet synchronous linear motor and other engineering.Keywords: permanent magnet synchronous linear motor, finite element analysis, chaotic search, optimization design

Abstract:
Age-related macular degeneration (AMD) is one of the leading cause of blindness. It can cause severe visual loss due to damaged retinal pigment epithelium (RPE). RPE is an important component of the retinal tissue. It functions as a transducing boundary for visual perception making it an essential factor for sight. The RPE also functions as a metabolically complex and functional cell layer that is responsible for the local homeostasis and maintenance of the extra photoreceptor environment. Thus one of the suggested method of treating such diseases would be regenerating these RPE cells. As such, we intend to grow these cells using a synthetic scaffold to provide a stable environment that reduces the batch effects found in natural scaffolds. Stiffness of the scaffold will also be investigated to determine the optimal Young’s modulus for cultivating these cells. The cells will be generated into a monolayer cell sheet and their functions such as formation of tight junctions and gene expression patterns will be assessed to evaluate the cell sheet quality compared to a native RPE tissue.Keywords: RPE, scaffold, characterization, biomaterials, colloids and nanomedicine

Abstract:
The emergence and neutralization of illegal waste dumps represent a global concern for waste management ecosystems with a particularly pronounced impact on disadvantaged communities. All over the world, and in this particular case in Romania, a relevant number of people resided in houses lacking any legal forms such as land ownership documents or building permits. These areas are referred to as “informal settlements”. An increasing number of regions and cities in Romania are struggling to manage their waste dumps, especially in the context of increasing poverty and lack of regulation related to informal settlements. An example of such informal settlement can be found at the terminus of Bistra Street in Câlnic, which falls under the jurisdiction of the Municipality of Reșița in Caras Severin County. The article presents a case study that focuses on employing remote sensing techniques and spatial data to monitor and map illegal waste practices, with subsequent integration into a geographic information system tailored for the Reșița community. In addition, the paper outlines the steps involved in devising strategies aimed at enhancing waste management practices in disadvantaged areas, aligning with the shift toward a circular economy. Results presented in the paper contain a spatial mapping and visualization methodology calibrated with in situ data collection applicable for identifying illegal landfills. The emergence and neutralization of illegal dumps pose a challenge in the field of waste management. Water splitting and rechargeable air-based batteries are emerging as new renewable energy storage and conversion technologies. However, the discovery of suitable catalysts with high activity and low cost remains a great challenge. In this work, we report a single-atom trifunctional catalyst, namely Ti₃C₂O₂ supported single Pd atom (Pd1@Ti₃C₂O₂), for the hydrogen evolution reaction (HER), oxygen evolution reaction (OER) and oxygen reduction reaction (ORR). This catalyst is selected from 12 candidates and possesses low overpotentials of 0.22 V, 0.31 V and 0.34 V for the HER, OER and ORR, respectively, making it an excellent electrocatalyst for both overall water splitting and rechargeable air-based batteries. The superior OER and ORR performance originates from the optimal d band center of the supported Pd atom. Moreover, the excellent activity can be maintained even if the single Pd atoms aggregate into small clusters. This work offers new opportunities for advancing the renewable energy storage and conversion technologies and paves a new way for the development of multifunctional electrocatalysts.Keywords: DFT, SACs, OER, ORR, HER

This paper studies the comparisons of individual and group replacement policies for a series connection system with two machines. Suppose that manufacturer’s production system is a series connection system which is combined by two machines. For two machines, when machines fail within the operating time, minimal repair is performed for machines by the manufacturer. The manufacturer plans to a preventive replacement for machines at a pre-specified time to maintain system normal operation. Under these maintenance policies, the maintenance cost rate models of individual and group replacement for a series connection system with two machines is derived and further, optimal preventive replacement time is obtained such that the expected total maintenance cost rate is minimized. Abstract:
We report the label-free electrochemical detection of Escherichia coli O157:H7 (ATCC 43895) in potable water using a DNA probe as a sensing molecule targeting the open reading frame marker. Indium tin oxide (ITO) surface was modified with organosilane and, glutaraldehyde was applied as a linker to fabricate the DNA sensor chip. Non-Faradic electrochemical impedance spectroscopy (EIS) behavior was investigated at each step of sensor fabrication using cyclic voltammetry, impedance, phase, relative permittivity, capacitance, and admittance. Atomic force microscopy (AFM) and scanning electron microscopy (SEM) revealed significant changes in surface topographies of DNA sensor chip fabrication. The decrease in the percentage of pinholes from 2.05 (Bare ITO) to 1.46 (after DNA hybridization) suggested the capacitive behavior of the DNA sensor chip. The results of non-Faradic EIS studies of DNA sensor chip showed a systematic declining trend of the capacitance as well as the relative permittivity upon DNA hybridization. DNA sensor chip exhibited linearity in 0.5 to 25 pg/10mL for E. coli O157:H7 (ATCC 43895). The limit of detection (LOD) at 95% confidence estimated by logistic regression was 0.1 pg DNA/10mL of E. coli O157:H7 (equivalent to 13.67 CFU/10mL) with a p-value of 0.0237. Moreover, the fabricated DNA sensor chip used for detection of E. coli O157:H7 showed no significant cross-reactivity with closely and distantly related bacteria such as Escherichia coli MTCC 3221, Escherichia coli O78:H11 MTCC 723 and Bacillus subtilis MTCC 736. Consequently, the results obtained in our study demonstrated the possible application of developed DNA sensor chips for E. coli O157:H7 ATCC 43895 in real water samples as well.Keywords: capacitance, DNA sensor, Escherichia coli O157:H7, open reading frame marker
