1164 Effect of Capsule Storage on Viability of Lactobacillus bulgaricus and Streptococcus thermophilus in Yogurt Powder

Abstract:
Yogurt capsule was made by mixing 14% w/v of reconstitution of skim milk with 2% FOS. The mixture was fermented by commercial yogurt starter comprising Lactobacillus bulgaricus and Streptococcus thermophilus. These yogurts were made as yogurt powder by freeze-dried. Yogurt powder was put into capsule then stored for 28 days at 4oc. 8ml of commercial yogurt was found to be the most suitable inoculum size in yogurt production. After freeze-dried, the viability of L. bulgaricus and S. thermophilus reduced from 109 to 107 cfu/g. The precence of sucrose cannot help to protect cell from ice crystal formation in freeze-dried process, high (20%) sucrose reduced L. bulgaricus and S. thermophilus growth during fermentation of yogurt. The addition of FOS had reduced slowly the viability of both L. bulgaricus and S. thermophilus similar to control (without FOS) during 28 days of capsule storage. The viable cell exhibited satisfactory viability level in capsule storage (6.7x106cfu/g) during 21 days at 4oC.Keywords: Yogurt capsule, Lactobacillus bulgaricus, Streptococcus thermophilus, freeze-drying, sucrose.

Abstract:
Soil organic carbon (SOC) plays a key role in soil fertility, hydrology, contaminants control and acts as a sink or source of terrestrial carbon content that can affect the concentration of atmospheric CO2. SOC supports the sustainability and quality of ecosystems, especially in semi-arid region. This study was conducted to determine relative importance of 13 different exploratory climatic, soil and geometric factors on the SOC contents in one of the semiarid watershed zones in Iran. Two methods canonical discriminate analysis (CDA) and feed-forward back propagation neural networks were used to predict SOC. Stepwise regression and sensitivity analysis were performed to identify relative importance of exploratory variables. Results from sensitivity analysis showed that 7-2-1 neural networks and 5 inputs in CDA models output have highest predictive ability that explains %70 and %65 of SOC variability. Since neural network models outperformed CDA model, it should be preferred for estimating SOC.Keywords: Soil organic carbon, modeling, neural networks, CDA.

Abstract:
The aim of this paper is to perform an analytic solution of steady state natural convection in a narrow rectangular channel between two vertical parallel MTR-type fuel plates, imposed under a cosine shape heat flux to determine the margin of the nuclear core power at which the natural convection cooling mode can ensure a safe core cooling, where the cladding temperature should not be reach the specific safety limits (90 °C). For this purpose, a simple computer program is developed to determine the principal parameter related to the nuclear core safety such as the temperature distribution in the fuel plate and in the coolant (light water) as a function of the reactor power. Modelling of building processes of a multimodal freight transportation support information system is discussed based on modern CASE technologies. Functional efficiencies of ports in the eastern part of the Black Sea are analyzed taking into account their ecological, seasonal, resource usage parameters. By resources, we mean capacities of berths, cranes, automotive transport, as well as work crews and neighbouring airports. For the purpose of designing database of computer support system for Managerial (Logistics) function, using Object-Role Modeling (ORM) tool (NORMA–Natural ORM Architecture) is proposed, after which Entity Relationship Model (ERM) is generated in automated process. Currently, there are few user friendly Weigh-in- Motion (WIM) data analysis softwares available which can produce traffic input data for the recently developed AASHTOWare pavement Mechanistic-Empirical (ME) design software. However, these softwares have only rudimentary Quality Control (QC) processes. Therefore, they cannot properly deal with erroneous WIM data. As the pavement performance is highly sensible to the quality of WIM data, it is highly recommended to use more refined QC process on raw WIM data to get a good result. This study develops a userfriendly software, which can produce traffic input for the ME design software. This software takes the raw data (Class and Weight data) collected from the WIM station and processes it with a sophisticated QC procedure. In Indonesia, goat milk is often consumed and believed as anti-allergy. The objective of this research was to study the effect of goat milk and their fractions (casein and whey) supplementation on total serum IgE concentrations and leukocytes count in rat sensitized with contact allergen dinitrochlorobenzene (DNCB). Female Wistar rats 6-8 weeks old were divided into four groups: 1) whey, 2) casein, 3) whole milk supplementation and 4) phosphate-buffered saline/PBS (control). The results showed that supplementation of goat milk on rats did not affects on total serum IgE concentrations and number of leukocytes. After sensitized with DNCB, the monocyte percentage in rats was higher (P<0.01) than before. In conclusion, goat milk or their fractions supplementation unable to decrease the total serum IgE concentrations and also had no effect on leukocytes count. Abstract:
Laser Doppler flowmetry is a modern method of noninvasive microcirculation investigation. The aim of our study was to use this method in the examination of patients with secondary lymphedema of the lower extremities and obliterating atherosclerosis of lower extremities. In the analysis of the amplitude-frequency spectrum of secondary lymphedema patients we have identified remarkable changes. To describe the changes we used a special amplitude rate. In both of patients groups this rate was significally (p<0.05) different with the control group. So the marker phenomena of the amplitude-frequency spectrum of the LDF signal were identified. It is suggested that there is a limfodynamics contribution to the formation of the output signal of laser Doppler flowmetry. These data have fundamental meaning and are interesting for practical medicine, as they give an opportunity to further developments for the use of laser Doppler flowmetry in the diagnostics and monitoring the effectiveness of the treatment.Keywords: laser Doppler flowmetry, secondary lymphedema of the lower extremities, obliterating atherosclerosis, non-invasive diagnostics.

Abstract:
Nanotechnology is widely applied in various areas so professionals in the related fields have to know more than nano knowledge. In the study, we focus on adopting ICT-assisted PBL in college general education to foster professionals who possess multiple abilities. The research adopted a pretest and posttest quasi-experimental design. The control group received traditional instruction, and the experimental group received ICT-assisted PBL instruction. Descriptive statistics will be used to describe the means, standard deviations, and adjusted means for the tests between the two groups. Next, analysis of covariance (ANCOVA) will be used to compare the final results of the two research groups after 6 weeks of instruction. Statistics gathered in the end of the research can be used to make contrasts. Therefore, we will see how different teaching strategies can improve students’ understanding about nanotechnology and learning skills.

Abstract:
Acaricides are commonly used to control ticks but are toxic, harmful to the environment and too expensive to resource-limited farmers. Traditionally, many communities in South Africa rely on a wide range of indigenous practices to keep their livestock healthy. One of these health care practices includes the use of medicinal plants and this offers an alternative to conventional medicine. An investigation was conducted at the CSIR in South Africa, and selected indigenous plants used in communities were scientifically evaluated for the management of ticks in animals. 17 plants were selected from 239 plants used traditionally in South Africa. Two different organic extracts were prepared from the 17 samples, resulting in 34 plant samples. These were tested for efficacy against two tick species, namely Rhipicephalus microplus and Rhipicephalus turanicus. The plant extracts were also screened against Vero cells and most were found to have low cytotoxicity. This study has shown that there is potential for the development of botanicals as natural acaricides against ticks that are non-toxic and environmentally benign.

Abstract:
Water contains dissolved oxygen that a fish needs to breathe. It is important to increase the amounts of separation of dissolved oxygen from water for diverse applications using the separation system. In this paper, a separation system of dissolved gases from water concurrently variable mixed with the exhalations using a compressor is proposed. This system takes use of exhalations to increase the amounts of separation of dissolved oxygen from water. A compressor with variable off-time and on-time is used to control the exhalations mixed with inlet water. Exhalations contain some portion of carbon dioxide, oxygen, and nitrogen. Separation of dissolved gases containing dissolved oxygen is enhanced by using exhalations. The amounts of separation and the compositions of carbon dioxide and oxygen are measured. Higher amounts of separation can make the size of the separation device smaller, and then, application areas are diversified.

The paper deals with the perspectives and possibilities of "smart solutions" to critical infrastructure protection. It means that common computer aided technologies are used from the perspective of new, better protection of selected infrastructure objects. The paper is focused on the co-product of the Czech Defence Research Project - ADAPTIV. This project is carrying out by the University of Defence, Faculty of Economics and Management at the Department of Civil Protection. The project creates system and technology for adaptive cybernetic camouflage of armed forces objects, armaments, vehicles and troops and of mobilization infrastructure. These adaptive camouflage system and technology will be useful for army tactic activities protection and for decoys generation also. The fourth chapter of the paper concerns the possibilities of using the introduced technology to the protection of selected civil (economically important), critical infrastructure objects. Abstract:
In this paper the performance of unified power flow controller is investigated in controlling the flow of po wer over the transmission line. Voltage sources model is utilized to study the behaviour of the UPFC in regulating the active, reactive power and voltage profile. This model is incorporated in Newton Raphson algorithm for load flow studies. Simultaneous method is employed in which equations of UPFC and the power balance equations of network are combined in to one set of non-linear algebraic equations. It is solved according to the Newton raphson algorithm. Case studies are carried on standard 5 bus network. Simulation is done in Matlab. Abstract:
Municipal solid waste (MSW) generation is among the most significant sources which threaten the global environmental health. Solid Waste Management has been an important environmental problem in developing countries because of the difficulties in finding sustainable solutions for solid wastes. Therefore, more efforts are needed to be implemented to overcome this problem. Lebanon has suffered a severe solid waste management problem in 2015, and a new landfill site was proposed to solve the existing problem. The study aims to identify and locate the most suitable area to construct a landfill taking into consideration the sustainable development to overcome the present situation and protect the future demands. Throughout the article, a landfill site selection methodology was discussed using Geographic Information System (GIS) and Multi Criteria Decision Analysis (MCDA). Several environmental, economic and social factors were taken as criterion for selection of a landfill. Soil, geology, and LUC (Land Use and Land Cover) indices with the Sustainable Development Index were main inputs to create the final map of Environmentally Sensitive Area (ESA) for landfill site. Different factors were determined to define each index. Input data of each factor was managed, visualized and analyzed using GIS. GIS was used as an important tool to identify suitable areas for landfill. Spatial Analysis (SA), Analysis and Management GIS tools were implemented to produce input maps capable of identifying suitable areas related to each index. Weight has been assigned to each factor in the same index, and the main weights were assigned to each index used. The combination of the different indices map generates the final output map of ESA. The output map was reclassified into three suitability classes of low, moderate, and high suitability. Results showed different locations suitable for the construction of a landfill. Results also reflected the importance of GIS and MCDA in helping decision makers finding a solution of solid wastes by a sanitary landfill.

This paper presents a review on vision aided systems and proposes an approach for visual rehabilitation using stereo vision technology. The proposed system utilizes stereo vision, image processing methodology and a sonification procedure to support blind navigation. The developed system includes a wearable computer, stereo cameras as vision sensor and stereo earphones, all moulded in a helmet. The image of the scene infront of visually handicapped is captured by the vision sensors. The captured images are processed to enhance the important features in the scene in front, for navigation assistance. The image processing is designed as model of human vision by identifying the obstacles and their depth information. The processed image is mapped on to musical stereo sound for the blind-s understanding of the scene infront. The developed method has been tested in the indoor and outdoor environments and the proposed image processing methodology is found to be effective for object identification.Keywords: Blind navigation, stereo vision, image processing, object preference, music tones.

Information Security is the most describing problem in present times. To cop up with the security of the information, the passwords were introduced. The alphanumeric passwords are the most popular authentication method and still used up to now. However, text based passwords suffer from various drawbacks such as they are easy to crack through dictionary attacks, brute force attacks, keylogger, social engineering etc. Graphical Password is a good replacement for text password. Psychological studies say that human can remember pictures better than text. So this is the fact that graphical passwords are easy to remember. But at the same time due to this reason most of the graphical passwords are prone to shoulder surfing. In this paper, we have suggested a shoulder-surfing resistant graphical password authentication method. The system is a combination of recognition and pure recall based techniques. Proposed scheme can be useful for smart hand held devices (like smart phones i.e. PDAs, iPod, iPhone, etc) which are more handy and convenient to use than traditional desktop computer systems.

Abstract:
Visualizing sound and noise often help us to determine an appropriate control over the source localization. Near-field acoustic holography (NAH) is a powerful tool for the ill-posed problem. However, in practice, due to the small finite aperture size, the discrete Fourier transform, FFT based NAH couldn-t predict the activeregion- of-interest (AROI) over the edges of the plane. Theoretically few approaches were proposed for solving finite aperture problem. However most of these methods are not quite compatible for the practical implementation, especially near the edge of the source. In this paper, a zip-stuffing extrapolation approach has suggested with 2D Kaiser window. It is operated on wavenumber complex space to localize the predicted sources. We numerically form a practice environment with touch impact databases to test the localization of sound source. It is observed that zip-stuffing aperture extrapolation and 2D window with evanescent components provide more accuracy especially in the small aperture and its derivatives.Keywords: Acoustic source localization, Near-field acoustic holography (NAH), FFT, Extrapolation, k-space wavenumber errors.

Abstract:
A. niger XP isolated from Vietnam produces very low amount of acidic phytase with optimal pH at 2.5 and 5.5. The phytase production of this strain was successfully improved through gene cloning and expression. A 1.4 - kb DNA fragment containing the coding region of the phyA gene was amplified by PCR and inserted into the expression vector pPICZαA with a signal peptide α- factor, under the control of AOX1 promoter. The recombined plasmid was transformed into the host strain P. pastoris KM71H and X33 by electroporation. Both host strains could efficiently express and secret phytase. The multicopy strains were screened for over expression of phytase. All the selected multicopy strains of P. pastoris X33 were examined for phytase activity, the maximum phytase yield of 1329 IU/ml was obtained after 4 days of incubation in medium BMM. The recombinant protein with MW of 97.4 KW showed to be the only one protein secreted in the culture broth. Multicopy transformant P. pastoris X33 supposed to be potential candidate for producing the commercial preparation of phytase.

The main aim is to perform mutational analysis of CTLA4 gene Exon 1 in SLE patients. A total of 61 SLE patients fulfilling “American College of Rheumatology (ACR) criteria" and 61 controls were enrolled in this study. The region of CTLA4 gene exon 1 was amplified by using Step-down PCR technique. Extracted DNA of band 354 bp was sequenced to analyze mutations in the exon-1 of CTLA-4 gene. Further, protein sequences were identified from nucleotide sequences of CTLA4 Exon 1 by using Expasy software and through Blast P software it was found that CTLA4 protein sequences of Pakistani SLE patients were similar to that of Chinese SLE population. No variations were found after patients sequences were compared with that of the control sequence. Furthermore it was found that CTLA4 protein sequences of Pakistani SLE patients were similar to that of Chinese SLE population. Thus CTLA4 gene may not be responsible for an autoimmune disease SLE.

Abstract:
Computerized lip reading has been one of the most actively researched areas of computer vision in recent past because of its crime fighting potential and invariance to acoustic environment. However, several factors like fast speech, bad pronunciation, poor illumination, movement of face, moustaches and beards make lip reading difficult. In present work, we propose a solution for automatic lip contour tracking and recognizing letters of English language spoken by speakers using the information available from lip movements. Level set method is used for tracking lip contour using a contour velocity model and a feature vector of lip movements is then obtained. Character recognition is performed using modified k nearest neighbor algorithm which assigns more weight to nearer neighbors. The proposed system has been found to have accuracy of 73.3% for character recognition with speaker lip movements as the only input and without using any speech recognition system in parallel. Abstract:
In this paper, we present a microcontroller-based liquid level controller which identifies the various levels of a liquid, carries out certain actions and is capable of communicating with the human being and other devices through the GSM network. This project is useful in ensuring that a liquid is not wasted. It also contributes to the internet of things paradigm, which is the future of the internet. The method used in this work includes designing the circuit and simulating it. The circuit is then implemented on a solderless breadboard after which it is implemented on a strip board. A C++ computer program is developed and uploaded into the microcontroller. This program instructs the microcontroller on how to carry out its actions. In other to determine levels of the liquid, an ultrasonic wave is sent to the surface of the liquid similar to radar or the method for detecting the level of sea bed. Message is sent to the phone of the user similar to the way computers send messages to phones of GSM users. It is concluded that the routine of observing the levels of a liquid in a tank, refilling the tank when the liquid level is too low can be entirely handled by a programmable device without wastage of the liquid or bothering a human being with such tasks.

A large variety of methods, different in concept and performance, have been newly developed and described. This paper analyzes the comparative merits of Sinusoidal Pulse Width Modulation (SPWM) and Space Vector Pulse Width Modulation (SVPWM) techniques and the suitability of these techniques in a Shunt Active Filter (SAF). The objective is to select the scheme that offers effective utilization of DC bus voltage and also harmonic reduction at the input side. The effectiveness of the PWM techniques is tested in the SAF configuration with a non linear load. The performance of the SAF with the SPWM and (SVPWM) techniques are compared with respect to the THD in source current. The study reveals that in the context of closed loop SAF control with the SVPWM technique there is only a minor improvement in THD. The utilization of the DC bus with SVPWM is also not significant compared to that with SPWM because of the non sinusoidal modulating signal from the controller in SAF configuration.Keywords: Voltage source inverter, Shunt active filter, SPWM, SVPWM, Matlab/SIMULINK.

Abstract:
Nonspecific protein adsorption generally occurs on any solid surfaces and usually has adverse consequences. Adsorption of proteins onto a solid surface is believed to be the initial and controlling step in biofouling. Surfaces modified with end-tethered poly(ethylene glycol) (PEG) have been shown to be protein-resistant to some degree. In this study, the adsorption of β-casein and lysozyme was performed on 6 different types of surfaces where PEG was tethered onto stainless steel by polyethylene imine (PEI) through either OH or NHS end groups. Protein adsorption was also performed on the bare stainless steel surface as a control. The adsorption was conducted at 23 °C and pH 7.2. In situ QCM-D was used to determine PEG adsorption kinetics, plateau PEG chain densities, protein adsorption kinetics and plateau protein adsorbed quantities. PEG grafting density was the highest for a NHS coupled chain, around 0.5 chains / nm2. Interestingly, lysozyme which has smaller size than β-casein, appeared to adsorb much less mass than that of β- casein. Overall, the surface with high PEG grafting density exhibited a good protein rejection.Keywords: QCM-D, PEG, stainless steel, β-casein, lysozyme.

Abstract:
Organic farming systems still depend on intensive, mechanical soil tillage. Frequent passes by machinery traffic cause substantial soil compaction that threatens soil health. Adopting practices as reduced tillage and organic matter retention on the soil surface are considered effective ways to control soil compaction. In tropical regions, however, the acceleration of soil organic matter decomposition and soil carbon turnover on the topsoil layer is influenced more rapidly by the oscillation process of drying and wetting. It is hypothesized therefore, that rapid reduction in soil organic matter hastens the potential for compaction to occur in organic farming systems. Compaction changes soil physical properties and as a consequence it has been implicated as a causal agent in the inhibition of natural disease suppression in soils. Here we describe relationships between soil management in organic vegetable systems, soil compaction, and declining soil capacity to suppress pathogenic microorganisms.

In this study, water monitoring was performed from Nov. 2012 to Oct. 2013 to assess water quality and evaluate the spatial and temporal distribution of physicochemical and biological variables in water. Water samples were collected from 10 coastal water stations of West Port. In the case of water-quality assessment, multi-metric indices and operational indicators have been proposed to classify the trophic status at different stations. The trophic level of West Port coastal water ranges from eutrophic to hypertrophic. Chl-a concentration was used to estimate the biological response of phytoplankton biomass and indicated eutrophic conditions in West Port and mesotrophic conditions at the control site. During the study period, no eutrophication events or secondary symptoms occurred, which may be related to hydrodynamic turbulence and water exchange, which prevent the development of eutrophic conditions in the West Port.Keywords: Water quality, multi-metric indices, operational indicator, Malaysia, West Port.

Abstract:
Graph coloring is an important problem in computer science and many algorithms are known for obtaining reasonably good solutions in polynomial time. One method of comparing different algorithms is to test them on a set of standard graphs where the optimal solution is already known. This investigation analyzes a set of 50 well known graph coloring instances according to a set of complexity measures. These instances come from a variety of sources some representing actual applications of graph coloring (register allocation) and others (mycieleski and leighton graphs) that are theoretically designed to be difficult to solve. The size of the graphs ranged from ranged from a low of 11 variables to a high of 864 variables. The method used to solve the coloring problem was the square of the adjacency (i.e., correlation) matrix. The results show that the most difficult graphs to solve were the leighton and the queen graphs. Complexity measures such as density, mobility, deviation from uniform color class size and number of block diagonal zeros are calculated for each graph. The results showed that the most difficult problems have low mobility (in the range of .2-.5) and relatively little deviation from uniform color class size.Keywords: graph coloring, complexity, algorithm.

Abstract:
Reburning is a useful technology in reducing nitric oxide through injection of a secondary hydrocarbon fuel. In this paper, an experimental study has been conducted to evaluate the effect of fuel lean reburning on NOx/CO reduction in LNG flame. Experiments were performed in flames stabilized by a co-flow swirl burner, which was mounted at the bottom of the furnace. Tests were conducted using LNG gas as the reburn fuel as well as the main fuel. The effects of reburn fuel fraction and injection manner of the reburn fuel were studied when the fuel lean reburning system was applied. The paper reports data on flue gas emissions and temperature distribution in the furnace for a wide range of experimental conditions. At steady state, temperature distribution and emission formation in the furnace have been measured and compared. This paper makes clear that in order to decrease both NOx and CO concentrations in the exhaust when the pulsated fuel lean reburning system was adapted, it is important that the control of some factors such as frequency and duty ratio. Also it shows the fuel lean reburning is also effective method to reduce NOx as much as reburning.

The evolutionary design of electronic circuits, or evolvable hardware, is a discipline that allows the user to automatically obtain the desired circuit design. The circuit configuration is under the control of evolutionary algorithms. Several researchers have used evolvable hardware to design electrical circuits. Every time that one particular algorithm is selected to carry out the evolution, it is necessary that all its parameters, such as mutation rate, population size, selection mechanisms etc. are tuned in order to achieve the best results during the evolution process. This paper investigates the abilities of evolution strategy to evolve digital logic circuits based on programmable logic array structures when different mutation rates are used. Several mutation rates (fixed and variable) are analyzed and compared with each other to outline the most appropriate choice to be used during the evolution of combinational logic circuits. Pomace, a by-product from fruit processing industry is the potential source of valuable bioactive. Cookies are popular, ready to eat and low price foods; therefore, enrichment of these products is of great importance. In this work, bioactive compounds extracted from cherry pomace, encapsulated in soy and whey proteins, have been incorporated in cookies, replacing 10 (SP10 and WP10) and 15% of wheat flour (SP15 and WP15). Cookie geometry (diameter (D), thickness (T) and spread ratio (D/T)), cookie weight, cookie hardness and cookie surface colour were measured. Sensory characteristics are also examined. The results show that encapsulated cherry pomace bioactives have positively influenced the cookie mass. Diameter, redness (a* value) and cookie hardness increased. Sensory evaluation of cookies, revealed that up to 15% substitution of wheat flour with WP encapsulate produced acceptable cookies similar to the control (100% wheat flour) cookies.Keywords: Cherry pomace, polyphenols, microencapsulation, cookies, physical characteristics.

Abstract:
Fasting dissolving magnesium (DM) alloy technology has contributed significantly to the “Shale Revolution” in oil and gas industry. This application requires DM downhole tools dissolving initially at a slow rate, rapidly accelerating to a high rate after certain period of operation time (typically 8 h to 2 days), a contradicting requirement that can hardly be addressed by traditional Mg alloying or processing itself. Premature disintegration has been broadly reported in downhole DM tool from field trials. To address this issue, “temporary” thin polymers of various formulations are currently coated onto DM surface to delay its initial dissolving. Due to conveying parts, harsh downhole condition, and high dissolving rate of the base material, the current delay coatings relying on pure polymers are found to perform well only at low temperature (typical < 100 ℃) and parts without sharp edges or corners, as severe geometries prevent high quality thin film coatings from forming effectively. In this study, a coating technology combining Plasma Electrolytic Oxide (PEO) coatings with advanced thin film deposition has been developed, which can delay DM complex parts (with sharp corners) in corrosive fluid at 150 ℃ for over 2 days. Synergistic effects between porous hard PEO coating and chemical inert elastic-polymer sealing leads to its delaying dissolution improvement, and strong chemical/physical bonding between these two layers has been found to play essential role. Microstructure of this advanced coating and compatibility between PEO and various polymer selections has been thoroughly investigated and a model is also proposed to explain its delaying performance. This study could not only benefit oil and gas industry to unplug their High Temperature High Pressure (HTHP) unconventional resources inaccessible before, but also potentially provides a technical route for other industries (e.g., bio-medical, automobile, aerospace) where primer anti-corrosive protection on light Mg alloy is highly demanded.

Abstract:
Atherosclerosis was identified as a chronic inflammatory process resulting from interactions between plasma lipoproteins, cellular components (monocyte, macrophages, T lymphocytes, endothelial cells and smooth muscle cells) and the extracellular matrix of the arterial wall. Several types of genes were known to express during formation of atherosclerosis. This study is carried out to identify unknown differentially expressed gene (DEG) in atherogenesis. Rabbit’s aorta tissues were stained by H&E for histomorphology. GeneFishing™ PCR analysis was performed from total RNA extracted from the aorta tissues. The DNA fragment from DEG was cloned, sequenced and validated by Real-time PCR. Histomorphology showed intimal thickening in the aorta. DEG detected from ACP-41 was identified as cathepsin B gene and showed upregulation at week-8 and week-12 of atherogenesis. Therefore, ACP-based GeneFishing™ PCR facilitated identification of cathepsin B gene which was differentially expressed during development of atherosclerosis.
