5026 Orthogonal Regression for Nonparametric Estimation of Errors-In-Variables Models

Abstract:
Two new algorithms for nonparametric estimation of errors-in-variables models are proposed. The first algorithm is based on penalized regression spline. The spline is represented as a piecewise-linear function and for each linear portion orthogonal regression is estimated. This algorithm is iterative. The second algorithm involves locally weighted regression estimation. When the independent variable is measured with error such estimation is a complex nonlinear optimization problem. The simulation results have shown the advantage of the second algorithm under the assumption that true smoothing parameters values are known. Nevertheless the use of some indexes of fit to smoothing parameters selection gives the similar results and has an oversmoothing effect.Keywords: grade point average, orthogonal regression, penalized regression spline, locally weighted regression

Abstract:
Landcover classification using automated classification techniques, while employing remotely sensed multi-spectral imagery, is one of the promising areas of research. Different land conditions at different time are captured through satellite and monitored by applying different classification algorithms in specific environment. In this paper, a SPOT-5 image provided by SUPARCO has been studied and classified in Environment for Visual Interpretation (ENVI), a tool widely used in remote sensing. Then, Artificial Neural Network (ANN) classification technique is used to detect the land cover changes in Abbottabad district. Obtained results are compared with a pixel based Distance classifier. The results show that ANN gives the better overall accuracy of 99.20% and Kappa coefficient value of 0.98 over the Mahalanobis Distance Classifier.Keywords: landcover classification, artificial neural network, remote sensing, SPOT 5

Abstract:
Self-driving vehicle require a high level of situational awareness in order to maneuver safely when driving in real world condition. This paper presents a LiDAR based real time perception system that is able to process sensor raw data for multiple target detection and tracking in dynamic environment. The proposed algorithm is nonparametric and deterministic that is no assumptions and priori knowledge are needed from the input data and no initializations are required. Additionally, the proposed method is working on the three-dimensional data directly generated by LiDAR while not scarifying the rich information contained in the domain of 3D. Moreover, a fast and efficient for real time clustering algorithm is applied based on a radially bounded nearest neighbor (RBNN). Hungarian algorithm procedure and adaptive Kalman filtering are used for data association and tracking algorithm. The proposed algorithm is able to run in real time with average run time of 70ms per frame.Keywords: lidar, segmentation, clustering, tracking

Abstract:
In France, the railway network is one of the main transport infrastructures and is the second largest European network. Therefore, railway inspection is an important task in railway maintenance to ensure safety for passengers using significant means in personal and technical facilities. Artificial vision has recently been applied to several railway applications due to its potential to improve the efficiency and accuracy when analyzing large databases of acquired images. In this paper, we present a vision system able to detect fastening elements based on artificial vision approach. This system acquires railway images using a CCD camera installed under a control carriage. These images are stitched together before having processed. Abstract:
In this study, it was tried to identify some heart rhythm disorders by electrocardiography (ECG) data that is taken from MIT-BIH arrhythmia database by subtracting the required features, presenting to artificial neural networks (ANN), artificial immune systems (AIS), artificial neural network based on artificial immune system (AIS-ANN) and particle swarm optimization based artificial neural network (PSO-NN) classifier systems. The main purpose of this study is to evaluate the performance of hybrid AIS-ANN and PSO-ANN classifiers with regard to the ANN and AIS. For this purpose, the normal sinus rhythm (NSR), atrial premature contraction (APC), sinus arrhythmia (SA), ventricular trigeminy (VTI), ventricular tachycardia (VTK) and atrial fibrillation (AF) data for each of the RR intervals were found. Then these data in the form of pairs (NSR-APC, NSR-SA, NSR-VTI, NSR-VTK and NSR-AF) is created by combining discrete wavelet transform which is applied to each of these two groups of data and two different data sets with 9 and 27 features were obtained from each of them after data reduction. Afterwards, the data randomly was firstly mixed within themselves, and then 4-fold cross validation method was applied to create the training and testing data. The training and testing accuracy rates and training time are compared with each other. As a result, performances of the hybrid classification systems, AIS-ANN and PSO-ANN were seen to be close to the performance of the ANN system. Also, the results of the hybrid systems were much better than AIS, too. However, ANN had much shorter period of training time than other systems. In terms of training times, ANN was followed by PSO-ANN, AIS-ANN and AIS systems respectively. Satellite images in detail can serve an important role in the geographic study. Quantitative and qualitative information provided by the satellite and remote sensing images minimizes the complexity of work and time. Data/images are captured at regular intervals by satellite remote sensing systems, and the amount of data collected is often enormous, and it expands rapidly as technology develops. Interpreting remote sensing images, geographic data mining, and researching distinct vegetation types such as agricultural and forests are all part of satellite image categorization. One of the biggest challenge data scientists faces while classifying satellite images is finding the best suitable classification algorithms based on the available that could able to classify images with utmost accuracy. In order to categorize satellite images, which is difficult due to the sheer volume of data, many academics are turning to deep learning machine algorithms. As, the CNN algorithm gives high accuracy in image recognition problems and automatically detects the important features without any human supervision and the ANN algorithm stores information on the entire network (Abhishek Gupta., 2020), these two deep learning algorithms have been used for satellite image classification. This project focuses on remote sensing through Deep Neural Networks i.e., ANN and CNN with Deep Sat (SAT-4) Airborne dataset for classifying images. Thus, in this project of classifying satellite images, the algorithms ANN and CNN are implemented, evaluated & compared and the performance is analyzed through evaluation metrics such as Accuracy and Loss. In this paper we present the design and the implementation of a target tracking system where the target is set to be a moving person in a video sequence. The system can be applied easily as a vision system for mobile robot. The system is composed of two major parts the first is the detection of the person in the video frame using the SVM learning machine based on the “HOG” descriptors. The second part is the tracking of a moving person it’s done by using a combination of the Kalman filter and a modified version of the Camshift tracking algorithm by adding the target motion feature to the color feature, the experimental results had shown that the new algorithm had overcame the traditional Camshift algorithm in robustness and in case of occlusion.Keywords: camshift algorithm, computer vision, Kalman filter, object tracking

Abstract:
Due to the limited environmental parameters and the limited resolution of the sensor, the universal existence of the mixed pixels in the process of remote sensing images restricts the spatial resolution of the remote sensing images. Sub-pixel mapping technology can effectively improve the spatial resolution. As the bilinear interpolation algorithm inevitably produces the edge blur effect, which leads to the inaccurate sub-pixel mapping results. In order to avoid the edge blur effect that affects the sub-pixel mapping results in the interpolation process, this paper presents a new edge-directed interpolation algorithm which uses the covariance adaptive interpolation algorithm on the edge of the low-resolution image and uses bilinear interpolation algorithm in the low-resolution image smooth area. By using the edge-directed interpolation algorithm, the super-resolution of the image with low resolution is obtained, and we get the percentage of each sub-pixel under a certain type of high-resolution image. Then we rely on the probability value as a soft attribute estimate and carry out sub-pixel scale under the ‘hard classification’. Finally, we get the result of sub-pixel mapping. Through the experiment, we compare the algorithm and the bilinear algorithm given in this paper to the results of the sub-pixel mapping method. It is found that the sub-pixel mapping method based on the edge-directed interpolation algorithm has better edge effect and higher mapping accuracy. The results of the paper meet our original intention of the question. Abstract:
This research aims to investigate the artificial intelligence (AI) application Jung GPT and how helpful it is, as a therapy AI, to users. Jung GPT has the potential to make mental health care more accessible and cheaper while also providing tailored support and advice. However, it is not intended to be a substitute for human therapists. Jung GPT is instructed to understand a wide range of concepts, including emojis, sensitive subjects, and various languages. Furthermore, participants were asked to fill out a survey based on their experience with Jung GPT. Additionally, analysis of the responses indicated that Jung GPT was helpful in identifying and exploring challenges, and the use of Jung GPT by participants in the future is highly possible. The results demonstrate that Jung GPT does help in recognizing challenges or problems within the users. On this basis, it is recommended that individuals use Jung GPT to explore their thoughts, feelings, and challenges. The development of intelligent transportation systems (ITS) and artificial intelligence (AI) are spurring us to pave the way for the widespread adoption of autonomous vehicles (AVs). This is open again opportunities for smart roads, smart traffic safety, and mobility comfort. A highly intelligent decision-making system is essential for autonomous driving around dense, dynamic objects. It must be able to handle complex road geometry and topology, as well as complex multiagent interactions, and closely follow higher-level commands such as routing information. Autonomous vehicles have become a very hot research topic in recent years due to their significant ability to reduce traffic accidents and personal injuries. Using new artificial intelligence-based technologies handles important functions in scene understanding, motion planning, decision making, vehicle control, social behavior, and communication for AV. This paper focuses only on deep reinforcement learning-based methods; it does not include traditional (flat) planar techniques, which have been the subject of extensive research in the past because reinforcement learning (RL) has become a powerful learning framework now capable of learning complex policies in high dimensional environments. Abstract:
The increasing demand for sustainable construction materials has led to a growing interest in high-strength recycled concrete (HSRC). Utilizing recycled materials not only reduces waste but also minimizes the depletion of natural resources. This study explores the application of artificial intelligence (AI) techniques to model and predict the properties of HSRC. In the past two decades, the production levels in various industries and, consequently, the amount of waste have increased significantly. Continuing this trend will undoubtedly cause irreparable damage to the environment. For this reason, engineers have been constantly seeking practical solutions for recycling industrial waste in recent years. This research utilized the results of the compressive strength of 90-day high-strength recycled concrete. The method for creating recycled concrete involved replacing sand with crushed glass and using glass powder instead of cement. Subsequently, a feedforward artificial neural network was employed to model the compressive strength results for 90 days. In software development organizations, Component based Software engineering (CBSE) is emerging paradigm for software development and gained wide acceptance as it often results in increase quality of software product within development time and budget. In component reusability, main challenges are the right component identification from large repositories at right time. The major objective of this work is to provide efficient algorithm for storage and effective retrieval of components using neural network and parameters based on user choice through clustering. This research paper aims to propose an algorithm that provides error free and automatic process (for retrieval of the components) while reuse of the component. In this algorithm, keywords (or components) are extracted from software document, after by applying k mean clustering algorithm. Then weights assigned to those keywords based on their frequency and after assigning weights, ANN predicts whether correct weight is assigned to keywords (or components) or not, otherwise it back propagates in to initial step (re-assign the weights). In last, store those all keywords into repositories for effective retrieval. Abstract:
Grover’s search algorithm is the fastest possible quantum mechanical algorithm to search a certain element from an unstructured set of data of N items. The algorithm can determine the desired result in only O(√N) steps. It has been demonstrated theoretically and experimentally on two-qubit systems long ago. In this work, we investigate the fidelity of Grover’s search algorithm by implementing it on an open quantum system. In particular, we study with what accuracy one can estimate that the algorithm would deliver the searched state. In reality, every system has some influence on its environment. We include the environmental effects on the system dynamics by using a recently reported fluctuation-regulated quantum master equation (FRQME). We consider that the environment experiences thermal fluctuations, which leave its signature in the second-order term of the master equation through its appearance as a regulator. The FRQME indicates that in addition to the regular relaxation due to system-environment coupling, the applied drive also causes dissipation in the system dynamics. As a result, the fidelity is found to depend on both the drive-induced dissipative terms and the relaxation terms, and we find that there exists a competition between them, leading to an optimum drive amplitude for which the fidelity becomes maximum. For efficient implementation of the search algorithm, precise knowledge of this optimum drive amplitude is essential.Keywords: dissipation, fidelity, quantum master equation, relaxation, system-environment coupling

In this paper, an Improved Cuckoo Search algorithm is applied as an appropriate optimization algorithm to determine the optimum location and size of a Static Var Compensator (SVC) in a transmission network. The main objectives are voltage stability improvement and total cost minimization. Over the last few years, the amount of data available on the globe has been increased rapidly. This came up with the emergence of recent concepts, such as the big data and the Internet of Things, which have furnished a suitable solution for the availability of data all over the world. However, managing this massive amount of data remains a challenge due to their large verity of types and distribution. Therefore, locating the required file particularly from the first trial turned to be a not easy task, due to the large similarities of names for different files distributed on the web. Consequently, the accuracy and speed of search have been negatively affected. This work presents a method using Electroencephalography signals to locate the files based on their contents. Giving the concept of natural mind waves processing, this work analyses the mind wave signals of different people, analyzing them and extracting their most appropriate features using multi-objective metaheuristic algorithm, and then classifying them using artificial neural network to distinguish among files with similar names. The aim of this work is to provide the ability to find the files based on their contents using human thoughts only. Implementing this approach and testing it on real people proved its ability to find the desired files accurately within noticeably shorter time and retrieve them as a first choice for the user.Keywords: artificial intelligence, data contents search, human active memory, mind wave, multi-objective optimization

Abstract:
Hierarchical classification is a special type of classification task where the class labels are organised into a hierarchy, with more generic class labels being ancestors of more specific ones. Meta-learning for classification-algorithm recommendation consists of recommending to the user a classification algorithm, from a pool of candidate algorithms, for a dataset, based on the past performance of the candidate algorithms in other datasets. Meta-learning is normally used in conventional, non-hierarchical classification. By contrast, this paper proposes a meta-learning approach for more challenging task of hierarchical classification, and evaluates it in a large number of bioinformatics datasets. Hierarchical classification is especially relevant for bioinformatics problems, as protein and gene functions tend to be organised into a hierarchy of class labels. This work proposes meta-learning approach for recommending the best hierarchical classification algorithm to a hierarchical classification dataset. This work’s contributions are: 1) proposing an algorithm for splitting hierarchical datasets into new datasets to increase the number of meta-instances, 2) proposing meta-features for hierarchical classification, and 3) interpreting decision-tree meta-models for hierarchical classification algorithm recommendation.Keywords: algorithm recommendation, meta-learning, bioinformatics, hierarchical classification

This study investigates the implications of Artificial Intelligence Marketing (AIM) marketing mix (PRD) Product, (PRC) Price, (PRM), Promotion and (PLC) Place on consumption patterns of halal food (CPHF). A quantitative approach was adopted in this study and responses were obtained from 350 Indonesian consumers. Using Partial Least Squares-Structural Equation Modeling (PLS-SEM), the results show that there is a direct support of marketing mix (PRD, PRC, PLC) to AIM and CPHF, while PRM does not play a significant role in CPHF. In addition, the findings reveal that AIM mediates significantly the relationship between PLC, PRC and PRM and CPHF, while AIM indicates no mediation between PRD and CPHF. Indonesian consumer’s exhibit serious concerns with consumption patterns of halal food. it is recommended that managers focus their attention on marketing strategies to predict consumer behavior in terms of consumption patterns of halal food through the integration of AIM.Keywords: marketing mix, consumption patterns, artificial intelligence marketing, Halal food

This paper proposes a mathematical model and examines the performance of an exact algorithm for a location–transportation problems in humanitarian relief. The model determines the number and location of distribution centers in a relief network, the amount of relief supplies to be stocked at each distribution center and the vehicles to take the supplies to meet the needs of disaster victims under capacity restriction, transportation and budgetary constraints. The computational experiments are conducted on the various sizes of problems that are generated. Branch and bound algorithm is applied for these problems. The results show that this algorithm can solve problem sizes of up to three candidate locations with five demand points and one candidate location with up to twenty demand points without premature termination.Keywords: disaster response, facility location, humanitarian relief, transportation

Abstract:
In this paper, we propose a sign adaptive filter algorithm with the ability of dynamic selection of subband filters which leads to low computational complexity compared with conventional sign subband adaptive filter (SSAF) algorithm. Dynamic selection criterion is based on largest reduction of the mean square deviation at each adaption. We demonstrate that this simple proposed algorithm has the same performance of the conventional SSAF and somewhat faster than it. In the presence of impulsive interferences robustness of the simple proposed algorithm as well as the conventional SSAF and outperform the conventional normalized subband adaptive filter (NSAF) algorithm. Therefore, it is preferred for environments under impulsive interferences. Water is one of the most important resources for human society. The world is currently undergoing a wave of urban growth, and pollution problems are of a great impact. Monitoring water quality is a key task for the future of the environment and human species. In recent times, researchers, using Smart Cities technologies are trying to mitigate the problems generated by the population growth in urban areas. The availability of huge amounts of data collected by a pervasive urban IoT can increase the transparency of decision making. Several services have already been implemented in Smart Cities, but more and more services will be involved in the future. Water quality monitoring can successfully be implemented in the urban IoT. The combination of water quality sensors, cloud computing, smart city infrastructure, and IoT technology can lead to a bright future for environmental monitoring. In the past decades, lots of effort has been put on monitoring and predicting water quality using traditional approaches based on manual collection and laboratory-based analysis, which are slow and laborious. The present study proposes a methodology for implementing a water quality prediction model using artificial intelligence techniques and comparing the results obtained with different algorithms. Furthermore, a 3D numerical model will be created using the software D-Water Quality, and simulation results will be used as a training dataset for the artificial intelligence algorithm. This study derives the methodology and demonstrates its implementation based on information and data collected at the floating harbour in the city of Bristol (UK). The city of Bristol is blessed with the Bristol-Is-Open infrastructure that includes Wi-Fi network and virtual machines. Tracking of moving and flying targets is one of the most important issues in image processing topic. Estimating of trajectory of desired object in short-term and long-term scale is more important than tracking of moving and flying targets. In this paper, a new way of identifying and estimating of future trajectory of a moving ball in long-term scale is estimated by using synthesis and interaction of image processing algorithms including noise removal and image segmentation, Kalman filter algorithm in order to estimating of trajectory of ball in football game in short-term scale and intelligent adaptive neuro-fuzzy algorithm based on time series of traverse distance. The proposed system attain more than 96% identify accuracy by using aforesaid methods and relaying on aforesaid algorithms and data base video in format of synthesis and interaction. Although the present method has high precision, it is time consuming. Abstract:
The administration of veterinary treatments at higher doses than the recommended Varroa mite control in beehive matrices has the potential to generate residues in the honeybee colony and in the derived products for consumption. Honeybee colonies can also be indirectly exposed to residues of plant protection products when foraging in crops, wildflowers near the crops, or in urban gardens just after spraying. The study evaluates the presence of both types of residues, veterinary treatments, and pesticides in beeswax, bee bread, and honey. The study was carried out in apiaries located in agricultural zones and forest areas in Andalusia, Spain. Up to nineteen residues were identified above LOQ using gas chromatography-triple quadrupole-mass spectrometry analysis (GC-MS/MS). Samples were extracted by a modified QuEChERs method. Chlorfenvinphos was detected in beeswax and bee bread despite its use is not authorized for Varroa mite control. Residues of fluvalinate-tau, authorized as veterinary treatment, were detected in most of the samples of beeswax and bee bread, presumably due to overdose or also to its potential for accumulation associated with its marked liposolubility. Residues of plant protection products were also detected in samples of beeswax and bee bread. Pesticide residues were detected above the LOQ that was established at 5 µg.kg⁻¹, which is the minimum concentration that can be quantified with acceptable accuracy and precision, as described in the European guidelines for pesticide residue analysis SANTE/11945/2015. Real time non-invasive Brain Computer Interfaces have a significant progressive role in restoring or maintaining a quality life for medically challenged people. This manuscript provides a comprehensive review of emerging research in the field of cognitive/affective computing in context of human neural responses. The perspectives of different emotion assessment modalities like face expressions, speech, text, gestures, and human physiological responses have also been discussed. Focus has been paid to explore the ability of EEG (Electroencephalogram) signals to portray thoughts, feelings, and unspoken words. An automated workflow-based protocol to design an EEG-based real time Brain Computer Interface system for analysis and classification of human emotions elicited by external audio/visual stimuli has been proposed. The front end hardware includes a cost effective and portable Emotive EEG Neuroheadset unit, a personal computer and a set of external stimulators. Primary signal analysis and processing of real time acquired EEG shall be performed using MATLAB based advanced brain mapping toolbox EEGLab/BCILab. This shall be followed by the development of MATLAB based self-defined algorithm to capture and characterize temporal and spectral variations in EEG under emotional stimulations. The extracted hybrid feature set shall be used to classify emotional states using artificial intelligence tools like Artificial Neural Network. Uncertain data mining algorithms use different ways to consider uncertainty in data such as by representing a data object as a sample of points or a probability distribution. Fuzzy methods have long been used for clustering traditional (certain) data objects. They are used to produce non-crisp cluster labels. For uncertain data, however, besides some uncertain fuzzy k-medoids algorithms, not many other fuzzy clustering methods have been developed. In this work, we develop a fuzzy kernel k-medoids algorithm for clustering uncertain data objects. Abstract:
The present paper introduces a modified adaptive mixed differential evolution (MAMDE) to select the main geometry parameters of specific cylindrical spur gear. The developed algorithm used the self-adaptive mechanism in order to update the values of mutation and crossover factors. The feasibility rules are used in the selection phase to improve the search exploration of MAMDE. Moreover, the elitism is performed to keep the best individual found in each generation. For the constraints handling the normalization method is used to treat each constraint design equally. The finite element analysis is used to confirm the optimization results for the maximum bending resistance. Abstract:
Traditional means of harvesting honey are often stressful for honeybees. Each time honey is collected a portion of the colony can die. In consequence, the colonies’ resilience to environmental stressors will decrease and this ultimately contributes to the global problem of honeybee colony losses. As part of the project HIVEOPOLIS, we design and build a different kind of beehive, incorporating technology to reduce negative impacts of beekeeping procedures, including honey harvesting. A first step in maintaining more sustainable honey harvesting practices is to design honey storage frames that can automate the honey collection procedures. This way, beekeepers save time, money, and labor by not having to open the hive and remove frames, and the honeybees' nest stays undisturbed.This system shows promising features, e.g., high reliability which could be a key advantage compared to current honey harvesting technologies.Our original concept of fractional honey harvesting has been to encourage the removal of honey only from "safe" locations and at levels that would leave the bees enough high-nutritional-value honey. In this abstract, we describe the current state of our honey harvester, its technology and areas to improve. The honey harvester works by separating the honeycomb cells away from the comb foundation; the movement and the elastic nature of honey supports this functionality. The honey sticks to the foundation, because of the surface tension forces amplified by the geometry. In the future, by monitoring the weight and therefore the capped honey cells on our honey harvester frames, we will be able to remove honey as soon as the weight measuring system reports that the comb is ready for harvesting. Higher viscosity honey or crystalized honey cause challenges in temperate locations when a smooth flow of honey is required. We use resistive heaters to soften the propolis and wax to unglue the moving parts during extraction. These heaters can also melt the honey slightly to the needed flow state. Precise control of these heaters allows us to operate the device for several purposes. We use ‘Nitinol’ springs that are activated by heat as an actuation method. Unlike conventional stepper or servo motors, which we also evaluated throughout development, the springs and heaters take up less space and reduce the overall system complexity. Honeybee acceptance was unknown until we actually inserted a device inside a hive. We not only observed bees walking on the artificial comb but also building wax, filling gaps with propolis and storing honey. This also shows that bees don’t mind living in spaces and hives built from 3D printed materials. We do not have data yet to prove that the plastic materials do not affect the chemical composition of the honey. We succeeded in automatically extracting stored honey from the device, demonstrating a useful extraction flow and overall effective operation this way.Keywords: honey harvesting, honeybee, hiveopolis, nitinol

In this article, we propose a protocol called DataGA-DRF (a protocol for Data collection using a Genetic Algorithm through Dynamic Reference Points) that collects data from Heterogeneous wireless sensor networks. This protocol is based on DGA (Destination selection according to Genetic Algorithm) to control the movement of the UAV (Unmanned aerial vehicle) between dynamic reference points that virtually represent the sensor node deployment. The dynamics of these points ensure an even distribution of energy consumption among the sensors and also improve network performance. Due to this, real-world clinical execution and implementation of AI have not yet aged. We believe that examining the determinants is key to addressing these challenges. Furthermore, the literature does not yet particularize determinants of AI capabilities and ways of empowering the healthcare ecosystem to develop AI capabilities in a developing economy. Thus, this study aims to position AI as a digital transformation weapon for the healthcare ecosystem by examining AI capability determinants and providing insights on better empowering the healthcare industry to develop AI capabilities. To do so, we base on the technology-organization-environment (TOE) model and will apply a mixed research approach. The localization information is crucial for the operation of WSN. There are principally two types of localization algorithms. The Range-based localization algorithm has strict requirements on hardware; thus, it is expensive to be implemented in practice. The Range-free localization algorithm reduces the hardware cost. However, it can only achieve high accuracy in ideal scenarios. In this paper, we locate unknown nodes by incorporating the advantages of these two types of methods. The proposed algorithm makes the unknown nodes select the nearest anchor using the Received Signal Strength Indicator (RSSI) and choose two other anchors which are the most accurate to achieve the estimated location. Our algorithm improves the localization accuracy compared with previous algorithms, which has been demonstrated by the simulating results.Keywords: WSN, localization, DV-Hop, RSSI

Abstract:
Cleaning and disinfection procedures are essential for maintaining the cleanliness status of the pharmaceutical manufacturing environment particularly of the cleanrooms and sterile unit area. The Good Manufacturing Practice (GMP) Annex 1 recommendation highly requires the implementation of the standard and validated cleaning and disinfection protocols. However, environmental monitoring has shown that even a validated cleaning method with certified agents may result in the presence of atypical microorganisms’ colony that exceeds GMP limits for a specific cleanroom area. In response to this issue, this case study aims to arrive at the root cause of the microbial contamination observed in the sterile production environment in Profarma pharmaceutical industry in Albania through applying a problem-solving practical approach that ensures the appropriate sterility grade. The guidelines and literature emphasize the importance of several factors in the prevention of possible microbial contamination occurring in cleanrooms, grade A and C. These factors are integrated into a practical framework, to identify the root cause of the presence of Aspergillus Niger colony in the sterile production environment in Profarma pharmaceutical industry in Albania. In addition, the application of a semi-automatic disinfecting system such as H2O2 FOG into sterile grade A and grade C cleanrooms has been an effective solution in eliminating the atypical colony of Aspergillus Niger. Selecting the appropriate detergents and disinfectants at the right concentration, frequency, and combination; the presence of updated and standardized guidelines for cleaning and disinfection as well as continuous training of operators on these practices in accordance with the updated GMP guidelines are some of the identified factors that influence the success of achieving sterility grade. However, to ensure environmental sustainability it is important to be prepared for identifying the source of contamination and making the appropriate decision. The proposed case-based practical approach may help pharmaceutical companies to achieve sterile production and cleanliness environmental sustainability in challenging situations. Apart from the integration of valid agents and standardized cleaning and disinfection protocols according to GMP Annex 1, pharmaceutical companies must be careful and investigate the source and all the steps that can influence the results of an abnormal situation.
