49 Function of miR-125b in Zebrafish Neurogenesis

Abstract:
MicroRNAs are an important class of gene expression regulators that are involved in many biological processes including embryogenesis. miR-125b is a conserved microRNA that is enriched in the nervous system. We have previously reported the function of miR-125b in neuronal differentiation of human cell lines. We also discovered the function of miR-125b in regulating p53 in human and zebrafish. Here we further characterize the brain defects in zebrafish embryos injected with morpholinos against miR-125b. Our data confirm the essential role of miR-125b in brain morphogenesis particularly in maintaining the balance between proliferation, cell death and differentiation. We identified lunatic fringe (lfng) as an additional target of miR-125b in human and zebrafish and suggest that lfng may mediate the function of miR-125b in neurogenesis. Together, this report reveals new insights into the function of miR- 125b during neural development of zebrafish.Keywords: microRNA, miR-125b, neurogenesis, zebrafish.

Abstract:
5-FU is a chemotherapeutic agent that has been used in colorectal cancer (CRC) treatment. However, it is usually associated with the acquired resistance, which decreases the therapeutic effects of 5-FU. miR-200c is involved in chemotherapeutic drug resistance, but its mechanism is not fully understood. In this study, the effect of inhibition of miR-200c in sensitivity of HCT-116 CRC cells to 5-FU was evaluated. HCT-116 cells were transfected with LNA-anti- miR-200c for 48 h. mRNA expression of miR-200c was evaluated using quantitative real- time PCR. The protein expression of phosphatase and tensin homolog (PTEN) and E-cadherin were analyzed by western blotting. Annexin V and propidium iodide staining assay were applied for apoptosis detection. The caspase-3 activation was evaluated by an enzymatic assay. The results showed LNA-anti-miR-200c inhibited the expression of PTEN and E-cadherin protein, apoptosis and activation of caspase 3 compared with control cells. In conclusion, these results suggest that miR-200c as a prognostic marker can overcome to 5-FU chemoresistance in CRC.

Abstract:
The main features of NPP-2006/MIR-1200 design are described. Estimation of individual doses for population under normal operation and accident conditions is performed for Leningradskaya NPP – 2 as an example. The radiation effect on population and environment doesn-t exceed the established normative limit and is as low as reasonably achievable. Abstract:
It has been established that microRNAs (miRNAs) play an important role in gene expression by post-transcriptional regulation of messengerRNAs (mRNAs). However, the precise relationships between microRNAs and their target genes in sense of numbers, types and biological relevance remain largely unclear. Dissecting the miRNA-target relationships will render more insights for miRNA targets identification and validation therefore promote the understanding of miRNA function. In miRBase, miRanda is the key algorithm used for target prediction for Zebrafish. This algorithm is high-throughput but brings lots of false positives (noise). Since validation of a large scale of targets through laboratory experiments is very time consuming, several computational methods for miRNA targets validation should be developed. In this paper, we present an integrative method to investigate several aspects of the relationships between miRNAs and their targets with the final purpose of extracting high confident targets from miRanda predicted targets pool. This is achieved by using the techniques ranging from statistical tests to clustering and association rules. Our research focuses on Zebrafish. It was found that validated targets do not necessarily associate with the highest sequence matching. Besides, for some miRNA families, the frequency of their predicted targets is significantly higher in the genomic region nearby their own physical location. Finally, in a case study of dre-miR-10 and dre-miR-196, it was found that the predicted target genes hoxd13a, hoxd11a, hoxd10a and hoxc4a of dre-miR- 10 while hoxa9a, hoxc8a and hoxa13a of dre-miR-196 have similar characteristics as validated target genes and therefore represent high confidence target candidates.Keywords: MicroRNA targets validation, microRNA-target relationships, dre-miR-10, dre-miR-196.

Abstract:
Objective: Biliary atresia (BA) is characterized by progressive liver fibrosis. Epithelial-mesenchymal transition (EMT) has been implicated as a key mechanism in the pathogenesis of organ fibrosis. MiR-200a has been shown to repress EMT. We aim to explore the role of miR-200a in the fibrogenesis of BA. Methods: We obtained the plasma samples and liver samples from patients with BA or controls to examine the role of miR-200a. Histological liver fibrosis was assessed using the Ishak fibrosis scores. Reverse transcription quantitative polymerase chain reaction (RT-qPCR) was performed to detect the expression of miR-200a in plasma. We also evaluated the expression of miR-200a in liver tissues using tyramide signal amplification fluorescence in situ hybridization (TSA-FISH). The expression of EMT related proteins zinc finger E-box-binding homeobox 1 (ZEB1), E-cadherin and α-smooth muscle actin (α-SMA) in the liver sections were detected by immunohistochemical staining. Results: We found that the expression of miR-200a was both elevated in the plasma and liver tissues from BA patients compared with the controls. The hepatic expression of ZEB1 and α-SMA were markedly increased in the liver sections from BA patients compared to the controls, whereas E-cadherin was downregulated in the BA group. Simultaneously, we noted that the hepatic expression of miR-200a, E-cadherin and α-SMA were upregulated with the progression of liver fibrosis in the BA group, while ZEB1 was downregulated with the progression of liver fibrosis in BA patients. Conclusion: These findings suggest EMT has a critical effect on the fibrotic process of BA, and the interaction between miR-200a and ZEB1 may regulate EMT and eventually influence liver fibrogenesis of BA.Keywords: Biliary atresia, liver fibrosis, MicroRNA, epithelial-mesenchymal transition, zinc finger e-box-binding homeobox 1.

Abstract:
After the recent devastating flood in Kashmir in 2014, dredging of the local water bodies, especially Jhelum River has become a priority for the government. Local government under the project name of 'Comprehensive Flood Management Programme' plans to undertake an increase in discharge of existing flood channels by removal of encroachments and acquisition of additional land, dredging and other works of the water bodies. The total quantity of soil to be dredged will be 16.15 lac cumecs. Dredged soil is a major component that would result from the project which requires disposal/utilization. This study analyses the effect of cement and sand on the engineering properties of soil. The tests were conducted with variable additions of sand (10%, 20% and 30%), whereas cement was added at 12%. Samples with following compositions: soil-cement (12%) and soil-sand (30%) were tested as well. Laboratory experiments were conducted to determine the engineering characteristics of soil, i.e., compaction, strength, and CBR characteristics. The strength characteristics of the soil were determined by unconfined compressive strength test and direct shear test. Unconfined compressive strength of the soil was tested immediately and for a curing period of seven days. CBR test was performed for unsoaked, soaked (worst condition- 4 days) and cured (4 days) samples.

Iodine radionuclides in accident releases under severe accident conditions at NPP with VVER are the most radiationimportant with a view to population dose generation at the beginning of the accident. To decrease radiation consequences of severe accidents the technical solutions for severe accidents management have been proposed in MIR.1200 project, with consideration of the measures for suppression of volatile iodine forms generation in the containment. Music segmentation is a key issue in music information retrieval (MIR) as it provides an insight into the internal structure of a composition. Structural information about a composition can improve several tasks related to MIR such as searching and browsing large music collections, visualizing musical structure, lyric alignment, and music summarization. The authors of this paper present the MTSSM framework, a twolayer framework for the multi-track segmentation of symbolic music. The strength of this framework lies in the combination of existing methods for local track segmentation and the application of global structure information spanning via multiple tracks. The first layer of the MTSSM uses various string matching techniques to detect the best candidate segmentations for each track of a multi-track composition independently. The overall penumbra is usually defined as the distance, p20–80, separating the 20% and 80% of the dose on the beam axis at the depth of interest. This overall penumbra accounts also for the fact that some photons emitted by the distal parts of the source are only partially attenuated by the collimator. Medulloblastoma is the most common type of childhood brain tumor and often spreads to the spine. Current guidelines call for surgery to remove as much of the tumor as possible, followed by radiation of the brain and spinal cord, and finally treatment with chemotherapy. To investigate the applicability of the EDR-2 film for clinical radiation dosimetry, percentage depth-doses, profiles and distributions in open and dynamically wedged fields were measured using film and compared with data from a Treatment Planning system.The validity of the EDR2 film to measure dose in a plane parallel to the beam was tested by irradiating 10 cm×10 cm and 4 cm×4 cm fields from a Siemens, primus linac with a 6MV beam and a source-to-surface distance of 100 cm. The film was placed Horizontally between solid water phantom blocks and marked with pin holes at a depth of 10 cm from the incident beam surface. The film measurement results, in absolute dose, were compared with ion chamber measurements using a Welhoffer scanning water tank system and Treatment Planning system. Abstract:
Daylight utilization is a key factor in achieving visual and thermal comfort, and energy savings in integrated building design. However, lack of measured data related to this topic has become a major challenge with the increasing need for integrating lighting concepts and simulations in the early stages of design procedures. The current paper deals with the values of daylight illuminance on horizontal and south facing vertical surfaces; the data are estimated using IESNA model and measured values of the horizontal and vertical illuminance, and a regression model with an acceptable linear correlation is obtained. The resultant illuminance frequency curves are useful for estimating daylight availability on south facing surfaces in Tehran. In addition, the relationship between indirect vertical illuminance and the corresponding global horizontal illuminance is analyzed. A simple parametric equation is proposed in order to predict the vertical illumination on a shaded south facing surface. The equation correlates the ratio between the vertical and horizontal illuminance to the solar altitude and is used with another relationship for prediction of the vertical illuminance. Both equations show good agreement, which allows for calculation of indirect vertical illuminance on a south facing surface at any time throughout the year.

Heat transfer from two cam shape cylinder in side-byside arrangement had been studied numerically. The transverse gap between the centers of cylinders (T) is allowed to vary to change the pitch ratio (T/Deq). The equivalent diameter of the cylinder (Deq) is 27.6 mm and pitch ratio varies in range of 1≤T/Deq≤3. The Reynolds numbers based on equivalent circular cylinder are within 50≤ Reeq≤150. The first part of the algorithm is like the other similar algorithms but second part of that is including a new easy implementing method which has reduced the vast number of inputoutput operations saliently. As decreasing processor operating time does not have any effect on main algorithm speed, any improvement in it should be done through decreasing the number of input-output operations. This paper propose an easy algorithm for choose the correct record location of the final list. Guard channels improve the probability of successful handoffs by reserving a number of channels exclusively for handoffs. This concept has the risk of underutilization of radio spectrum due to the fact that fewer channels are granted to originating calls even if these guard channels are not always used, when originating calls are starving for the want of channels. The penalty is the reduction of total carried traffic. The optimum number of guard channels can help reduce this problem. This paper presents fuzzy logic based guard channel scheme wherein guard channels are reorganized on the basis of traffic density, so that guard channels are provided on need basis. The equivalent diameter of the cylinder (Deq) is 27.6 mm and longitudinal pitch ratio varies in range 2<L/Deq<6. The Reynolds number based on equivalent circular cylinder are within 50< Reeq <300. Results show that Nusselt number of second cylinder increases about 5 to 33 times when longitudinal pitch ratio increases from 2 to 6.

In this paper a nonlinear model is presented to demonstrate the relation between production and marketing departments. By introducing some functions such as pricing cost and market share loss functions it will be tried to show some aspects of market modelling which has not been regarded before. The proposed model will be a constrained signomial geometric programming model. For model solving, after variables- modifications an iterative technique based on the concept of geometric mean will be introduced to solve the resulting non-standard posynomial model which can be applied to a wide variety of models in non-standard posynomial geometric programming form. Abstract:
Music Information Retrieval (MIR) and modern data mining techniques are applied to identify style markers in midi music for stylometric analysis and author attribution. Over 100 attributes are extracted from a library of 2830 songs then mined using supervised learning data mining techniques. Two attributes are identified that provide high informational gain. These attributes are then used as style markers to predict authorship. Using these style markers the authors are able to correctly distinguish songs written by the Beatles from those that were not with a precision and accuracy of over 98 per cent. The identification of these style markers as well as the architecture for this research provides a foundation for future research in musical stylometry.

Rapid progress in audio compression technology has contributed to the explosive growth of music available in digital form today. In a reversal of ideas, this work makes use of a recently proposed efficient audio compression scheme to develop three important applications in the context of Music Information Retrieval (MIR) for the effective manipulation of large music databases, namely automatic music recommendation (AMR), digital rights management (DRM) and audio finger-printing for song identification. The performance of these three applications has been evaluated with respect to a database of songs collected from a diverse set of genres.

This paper presents a new type of mechanism and trajectory planning strategy for bipedal walking robot. The newly designed mechanism is able to improve the performance of bipedal walking robot in terms of energy efficiency and weight reduction by utilizing minimum number of actuators. The usage of parallelogram mechanism eliminates the needs of having an extra actuator at the knee joint. This mechanism works together with the joint space trajectory planning in order to realize straight legged walking which cannot be achieved by conventional inverse kinematics trajectory planning due to the singularity. The effectiveness of the proposed strategy is confirmed by computer simulation results.

Abstract:
Although it is fully impossible to ensure that a software system is quite secure, developing an acceptable secure software system in a convenient platform is not unreachable. In this paper, we attempt to analyze software development life cycle (SDLC) models from the hardware systems and circuits point of view. To date, the SDLC models pay merely attention to the software security from the software perspectives. In this paper, we present new features for SDLC stages to emphasize the role of systems and circuits in developing secure software system through the software development stages, the point that has not been considered previously in the SDLC models.

Abstract:
Service Oriented Architecture (SOA) allows modeling of dynamic interaction between incongruous providers, which enables governing the development of complex applications. However, implementation of SOA comes with some challenges, including its adaptability and robustness. Dynamism is inherent to the nature of service based applications and of their running environment. These factors lead to necessity for dynamic adaptation. In this paper we try to describe basics and main structure of SOA adaptation process with a conceptual view to this issue. In this survey we will review the relevant adaptation approaches. This paper allows studying how different approaches deal with service oriented architecture adaptation life-cycle and provides basic guidelines for their analysis, evaluation and comparison.

Abstract:
In this investigative research paper, we have presented the simulation results of a three-phase rectifier circuit to improve the input side current using the passive filters, such as capacitors and inductors at the output and input terminals of the rectifier circuit respectively. All simulation works were performed in a personal computer using the PSPICE simulator software, which is a virtual circuit design and simulation software package. The output voltages and currents were measured across a resistive load of 1 k. We observed that the output voltage levels, input current wave shapes, harmonic contents through the harmonic spectrum, and total harmonic distortion improved due to the use of such filters.

Abstract:
The pressure drag from a cam shaped tube in cross flows have been investigated experimentally using pressure distribution measurement. The range of angle of attack and Reynolds number based on an equivalent circular tube are within 0≤α≤360° and 2×104< Reeq < 3.4 ×104, respectively. It is found that the pressure drag coefficient is at its highest at α=90° and 270° over the whole range of Reynolds number. Results show that the pressure drag coefficient of the cam shaped tube is lower than that of circular tube with the same surface area for more of the angles of attack. Furthermore, effects of the diameter ratio and finite length of the cam shaped tube upon the pressure drag coefficient are discussed.

Abstract:
Efficient handoff algorithms are a cost-effective way of enhancing the capacity and QoS of cellular system. The higher value of hysteresis effectively prevents unnecessary handoffs but causes undesired cell dragging. This undesired cell dragging causes interference or could lead to dropped calls in microcellular environment. The problems are further exacerbated by the corner effect phenomenon which causes the signal level to drop by 20-30 dB in 10-20 meters. Thus, in order to maintain reliable communication in a microcellular system new and better handoff algorithms must be developed. A fuzzy based handoff algorithm is proposed in this paper as a solution to this problem. Handoff on the basis of ratio of slopes of normal signal loss to the actual signal loss is presented. The fuzzy based solution is supported by comparing its results with the results obtained in analytical solution.Keywords: Slope ratio, handoff, corner effect, fuzzy logic.

Abstract:
Prime Factorization based on Quantum approach in two phases has been performed. The first phase has been achieved at Quantum computer and the second phase has been achieved at the classic computer (Post Processing). At the second phase the goal is to estimate the period r of equation xrN ≡ 1 and to find the prime factors of the composite integer N in classic computer. In this paper we present a method based on Randomized Approach for estimation the period r with a satisfactory probability and the composite integer N will be factorized therefore with the Randomized Approach even the gesture of the period is not exactly the real period at least we can find one of the prime factors of composite N.
