10 In vitro Effects of Viscum album on the Functionality of Rabbit Spermatozoa

Abstract:

This study aimed to assess the in vitro effects of different concentrations of the Viscum album extract on the motility, viability, and reactive oxygen species (ROS) production by rabbit spermatozoa during different time periods (0, 2, and 8h). Spermatozoa motility was assessed by using the CASA (Computer aided sperm analysis) system. Cell viability was evaluated by using the metabolic activity MTT assay, and the luminol-based luminometry was applied to quantify the ROS formation. The CASA analysis revealed that low Viscum concentrations were able to prevent a rapid decline of spermatozoa motility, especially in the case of concentrations ranging between 1 and 5 µg/mL (P<0.05 with respect to time 8h). At the same time, concentrations ranging between 1 and 100 µg/mL of the extract led to a significant preservation of the cell viability (P<0.05 in case of 5, 50 and 100 µg/mL; P<0.01 with respect to 1 and 10 µg/mL, time 8h). 1 and 5 µg/mL of the extract exhibited antioxidant characteristics, translated into a significant reduction of the ROS production, particularly notable at time 8h (P<0.01). The results indicate that the Viscum extract is capable of delaying the damage inflicted to the spermatozoon by the in vitro environment.

9 In vitro Effects of Amygdalin on the Functional Competence of Rabbit Spermatozoa

Abstract:

The present in vitro study was designed to reveal whether amygdalin (AMG) is able to cause changes to the motility, viability and mitochondrial activity of rabbit spermatozoa. New Zealand White rabbits (n = 10) aged four months were used in the study. Semen samples were collected from each animal and used for the in vitro incubation. The samples were divided into five equal parts and diluted with saline supplemented with 0, 0.5, 1, 2.5 and 5 mg/mL AMG. At times 0h, 3h and 5h spermatozoa motion parameters were assessed using the SpermVision™ computer-aided sperm analysis (CASA) system, cell viability was examined with the metabolic activity (MTT) assay, and the eosin-nigrosin staining technique was used to evaluate the viability of rabbit spermatozoa. All AMG concentrations exhibited stimulating effects on the spermatozoa activity, as shown by a significant preservation of the motility (P<0.05 with respect to 0.5 mg/mL and 1 mg/mL AMG; Time 5 h) and mitochondrial activity (P< 0.05 in case of 0.5 mg/mL AMG; P< 0.01 in case of 1 mg/mL AMG; P < 0.001 with respect to 2.5 mg/mL and 5 mg/mL AMG; Time 5 h). None of the AMG doses supplemented had any significant impact of the spermatozoa viability. In conclusion, the data revealed that short-term co-incubation of spermatozoa with AMG may result in a higher preservation of the sperm structural integrity and functional activity.

8 Architecture Exception Governance

Abstract:

The article presents the whole model of IS/IT architecture exception governance. As first, the assumptions of presented model are set. As next, there is defined a generic governance model that serves as a basis for the architecture exception governance. The architecture exception definition and its attributes follow. The model respects well known approaches to the area that are described in the text, but it adopts higher granularity in description and expands the process view with all the next necessary governance components as roles, principles and policies, tools to enable the implementation of the model into organizations. The architecture exception process is decomposed into a set of processes related to the architecture exception lifecycle consisting of set of phases and architecture exception states. Finally, there is information about my future research related to this area.

7 Outsourcing Opportunities for Internet Banking Solutions

Abstract:

The main goal of the article is to present new model of application architecture of banking IT solution providing the Internet Banking services that is particularly outsourced. At first, we propose business rationale and a SWOT analysis to explain the reasons for the model in the article. The most important factor for our model is nowadays- big boom around smart phones and tablet devices. As next, we focus on IT architecture viewpoint where we design application, integration and security model. Finally, we propose a generic governance model that serves as a basis for the specialized governance model. The specialized instance of governance model is designed to ensure that the development and the maintenance of different parts of the IT solution are well governed in time.

6 Combination of Electrodialysis and Electrodeionization for Treatment of Condensate from Ammonium Nitrate Production

Abstract:

Ammonium nitrate (AN) is produced by the reaction of ammonia and nitric acid, and a waste condensate is obtained. The condensate contains pure AN in concentration up to 10g/L. The salt content in the condensate is too high to discharge immediately into the river thus it must be treated. This study is concerned with the treatment of condensates from an industrial AN production by combination of electrodialysis (ED) and electrodeionization (EDI). The condensate concentration was in range 1.9–2.5g/L of AN. A pilot ED module with 25 membrane pairs following by a laboratory EDI module with 10 membrane pairs operated continuously during 800 hours. Results confirmed that the combination of ED and EDI is suitable for the condensate treatment.

5 Application of a Fracture-Mechanics Approach to Gas Pipelines

Abstract:

This study offers a new simple method for assessing an axial part-through crack in a pipe wall. The method utilizes simple approximate expressions for determining the fracture parameters K, J, and employs these parameters to determine critical dimensions of a crack on the basis of equality between the J-integral and the J-based fracture toughness of the pipe steel. The crack tip constraint is taken into account by the so-called plastic constraint factor C, by which the uniaxial yield stress in the J-integral equation is multiplied. The results of the prediction of the fracture condition are verified by burst tests on test pipes.

4 Determination of Material Properties for Biodegradable Polylactic Acid Plastic Used in 3D Printers

Abstract:

Within Rapid Prototyping technologies are used many types of materials. Many of them are recyclable but there are still as plastic like, so practically they do not degrade in the landfill. Polylactic acid (PLA) is one of the special plastic materials, which are biodegradable and available for 3D printing within Fused Deposition Modeling (FDM) technology. The question is, if the mechanical properties of produced models are comparable to similar technical plastic materials which are usual for prototype production. Presented paper shows the experiments results for tensile strength measurements for specimens prepared with different 3D printer settings and model orientation. Paper contains also the comparison of tensile strength values with values measured on specimens produced by conventional technologies as injection moulding.

3 Effects of Initial Moisture Content on the Physical and Mechanical Properties of Norway Spruce Briquettes

Abstract:

The moisture content of densified biomass is a limiting parameter influencing the quality of this solid biofuel. It influences its calorific value, density, mechanical strength and dimensional stability as well as affecting its production process. This paper deals with experimental research into the effect of moisture content of the densified material on the final quality of biofuel in the form of logs (briquettes or pellets). Experiments based on the singleaxis densification of the spruce sawdust were carried out with a hydraulic piston press (piston and die), where the densified logs were produced at room temperature. The effect of moisture content on the qualitative properties of the logs, including density, change of moisture, expansion and physical changes, and compressive and impact resistance were studied. The results show the moisture ranges required for producing good-quality logs. The experiments were evaluated and the moisture content of the tested material was optimized to achieve the optimum value for the best quality of the solid biofuel. The dense logs also have high-energy content per unit volume. The research results could be used to develop and optimize industrial technologies and machinery for biomass densification to achieve high quality solid biofuel.

2 Influence of Densification Process and Material Properties on Final Briquettes Quality from Fast-Growing Willows

Abstract:

Biomass treatment through densification is very suitable and helpful technology before its effective energy recovery. Densification process of biomass is significantly influenced by various technological and material variables, which are ultimately reflected on the final solid biofuels quality. The paper deals with the experimental research of the relationship between technological and material variables during densification of fast-growing trees, roundly fast-growing willows. The main goal of presented experimental research is to determine the relationship between compression pressure and raw material particle size from a final briquettes density point of view. Experimental research was realized by single-axis densification. The impact of particle size with interaction of compression pressure and stabilization time on the quality properties of briquettes was determined. These variables interaction affects the final solid biofuels (briquettes) quality. From briquettes production point of view and from densification machines constructions point of view is very important to know about mutual interaction of these variables on final briquettes quality. The experimental findings presented here are showing the importance of mentioned variables during the densification process.

1 Diagnostics of Existing Steel Structures of Winter Sport Halls

Abstract:

The paper deals with the diagnostics of steel roof structure of the winter sports halls built in 1970 year. The necessity of the diagnostics has been given by the requirement to the evaluation design of this structure, which has been caused by the new situation in the field of the loadings given by the validity of the European Standards in the Czech Republic from 2010 year. Due to these changes in the normative rules, in practice existing structures are gradually subjected to the evaluation design and depending on its results to the strengthening or reconstruction, respectively. Steel roof is composed of plane truss main girders, purlins and bracings and the roof structure is supported by two arch main girders with the span of L = 84 m. The in situ diagnostics of the roof structure was oriented to the following parts: (i) determination and evaluation of the actual material properties of used steel and (ii) verification of the actual dimensions of the structural members. For the solution the nondestructive methods have been used for in situ measurement. For the indicative determination of steel strengths the modified method based on the determination of Rockwell’s hardness has been used. For the verification of the member’s dimensions (thickness of hollow sections) the ultrasound method has been used. This paper presents the results obtained using these testing methods and their evaluation, from the viewpoint of the usage for the subsequent static assessment and design evaluation of the existing structure. For the comparison, the examples of the similar evaluations realized for steel structures of the stadiums in Olomouc and Jihlava cities are briefly illustrated, too.
