10 ACTN3 R577X Polymorphism in Romany Children from Eastern Slovakia

Abstract:

The paper presents the results of the application of molecular genetics methods in sport research, with special emphasis on the most advanced methods and trends in diagnosing of motoric predispositions for the sake of identifying talented children. Genetic tests differ in principle from the traditional motoric tests, because the DNA of an individual does not change during life. Genetics is important in determining the capacity of an individual and for professional sport level. Genetic information can be used for individual genetic predispositions in early childhood. The phenotypes are influenced by a combination of genetic and environmental factors. The aim of the presented study was to examine physical condition, coordination skills, motoric docility and to determine the frequency of ACTN3 (R577X) gene in Romany children from Eastern Slovakia and compared their motoric performance with non-Romany children. This paper is not looking just for a performance, but also its association to genetic predispositions in relation to ACTN3 gene and its R577X polymorphism. Genotype data were obtained from 175 Romany children from 6 to 15 years old and 218 non-Romany children at the same age from Eastern Slovakia. Biological material for genetic analyses comprised samples of buccal swabs. Genotypes were determined using Real Time High resolution melting PCR method (Rotor Gene 6000 Corbett and LightCycler 480 Roche). Romany children of analyzed group legged to non-Romany children at the same age in all the compared tests. The % distribution of R and X alleles in children was different from controls. The frequency of XX genotype was 11,45% which is comparable to a frequency of an Indian population. Data were analysed with the ANOVA statistical programme and parametric and nonparametric tests. 9 Identification of Multiple Sclerosis Risk Groups in the Egyptian Population: A Cross-Sectional Study for Egypt’s Health

Abstract:

Background: Multiple Sclerosis (MS) is a chronic autoimmune disease that affects the central nervous system, with a growing prevalence in Egypt. The interplay between genetic predisposition, environmental factors, and immune dysregulation contributes to the disease's onset and progression. In Egypt, where healthcare access and environmental exposures differ from Western contexts, understanding the impact of vaccinations, particularly COVID-19 vaccines, on MS disease activity is crucial. This study examines the relationship between vaccination status, disease characteristics, and clinical outcomes in a cohort of Egyptian MS patients, providing region-specific insights. Additionally, it identifies the risk groups and most susceptible categories to MS in the Egyptian population. Methodology: A cross-sectional analysis was conducted on 100 Egyptian MS patients, with data collected on demographic variables, vaccination status, disease subtype, relapse frequency, Expanded Disability Status Scale (EDSS) scores, and disease duration. Patients were categorized based on their vaccination status: unvaccinated (NONE), partially vaccinated (PART VAC), and fully vaccinated (FULL VAC). Statistical analyses, including ANOVA and Chi-square tests, were performed to assess the impact of vaccination on EDSS scores, relapse rates, and disease onset relative to vaccination. The study also considered unique regional factors, such as pesticide exposure and familial MS history, which are prevalent in the Egyptian context. Results: The study found no significant difference in EDSS scores across vaccination groups (ANOVA, p > 0.05), indicating that vaccination status does not significantly influence disability progression in Egyptian MS patients. However, a Chi-square analysis revealed a lower relapse rate among fully vaccinated patients compared to unvaccinated and partially vaccinated groups (p < 0.05). Additionally, disease onset after vaccination was more common in fully vaccinated patients, with 16 out of 21 patients experiencing disease initiation post-vaccination, compared to 5 out of 15 in the unvaccinated group (p < 0.05). This suggests a potential temporal association between vaccination and disease onset in the Egyptian cohort, though causality cannot be established. Additionally, 80% of MS population are women, 5% of the population have pediatric multiple sclerosis, and 10% have familial MS. Conclusion: In the Egyptian context, vaccination does not appear to exacerbate disability progression in MS patients and may be associated with a reduced relapse rate. However, the observed temporal relationship between vaccination and disease onset highlights the need for further investigation into whether this association is coincidental or indicative of a vaccine-triggered immune response in this population. These findings emphasize the importance of monitoring Egyptian MS patients’ post-vaccination and conducting longitudinal studies to better understand the long-term effects of vaccines on MS disease activity in this region.

However, without effective and efficient regulations, cryptocurrency transactions are mostly used by criminals to commit crimes such as money laundering, theft, and blackmailing. And because law is one step behind technological developments, this paper discusses the importance of regulations and supervision for the BitCoin-system, to provide unified regulatory solutions for our digital future in the Middle East. It will provide a detailed analysis of the legal nature of BitCoin along with, its regulation with respect to criminal and civil law.

7 The Impact of Artificial Intelligence on Human Rights Development

Abstract:

The relationship between development and human rights has long been the subject of academic debate. To understand the dynamics between these two concepts, various principles are adopted, from the right to development to development-based human rights. Despite the initiatives taken, the relationship between development and human rights remains unclear. However, the overlap between these two views and the idea that efforts should be made in the field of human rights have increased in recent years. It is then evaluated whether the right to sustainable development is acceptable or not. The article concludes that the principles of sustainable development are directly or indirectly recognized in various human rights instruments, which is a good answer to the question posed above. This book therefore cites regional and international human rights agreements such as , as well as the jurisprudence and interpretative guidelines of human rights institutions, to prove this hypothesis.

6 Factors Affecting Mobile Internet Adoption in an Emerging Market

Abstract:

The objective of this research is to find an explanatory model to define the most important variables and factors that affect the acceptance of Mobile Internet in the Egyptian market. A qualitative exploratory research was conducted to support the conceptual framework followed with a quantitative research in the form of a survey distributed among 411 respondents. It was clear that relative advantage, complexity, compatibility, perceived price level and perceived playfulness have a dominant role in influencing consumers to adopt mobile internet, while observability is correlated to the adoption but when measured with the other factors it lost its value. The perceived price level has a negative relationship with the adoption as well the compatibility.

5 Scalable Cloud-Based LEO Satellite Constellation Simulator

Abstract:

Distributed applications deployed on LEO satellites and ground stations require substantial communication between different members in a constellation to overcome the earth coverage barriers imposed by GEOs. Applications running on LEO constellations suffer the earth line-of-sight blockage effect. They need adequate lab testing before launching to space. We propose a scalable cloud-based net-work simulation framework to simulate problems created by the earth line-of-sight blockage. The framework utilized cloud IaaS virtual machines to simulate LEO satellites and ground stations distributed software. A factorial ANOVA statistical analysis is conducted to measure simulator overhead on overall communication performance. The results showed a very low simulator communication overhead. Consequently, the simulation framework is proposed as a candidate for testing LEO constellations with distributed software in the lab before space launch.

4 ACTN3 Genotype Association with Motoric Performance of Roma Children

Abstract:

The paper presents the results of the molecular genetics analysis in sports research, with special emphasis to use genetic information in diagnosing of motoric predispositions in Roma boys from East Slovakia. The ability and move are the basic characteristics of all living organisms. The phenotypes are influenced by a combination of genetic and environmental factors. Genetic tests differ in principle from the traditional motoric tests, because the DNA of an individual does not change during life. The aim of the presented study was to examine motion abilities and to determine the frequency of ACTN3 (R577X) gene in Roma children. Genotype data were obtained from 138 Roma and 155 Slovak boys from 7 to 15 years old. Children were investigated on physical performance level in association with their genotype. Biological material for genetic analyses comprised samples of buccal swabs. Genotypes were determined using Real Time High resolution melting PCR method (Rotor-Gene 6000 Corbett and Light Cycler 480 Roche). The software allows creating reports of any analysis, where information of the specific analysis, normalized and differential graphs and many information of the samples are shown. Roma children of analyzed group legged to non-Romany children at the same age in all the compared tests. The % distribution of R and X alleles in Roma children was different from controls. The frequency of XX genotype was 9.26%, RX 46.33% and RR was 44.41%. The frequency of XX genotype was 9.26% which is comparable to a frequency of an Indian population. Data were analyzed with the ANOVA test.

2 Activity of Commonly Used Intravenous Nutrient and Bisolvon in Neonatal Intensive Care Units against Biofilm Cells and Their Synergetic Effect with Antibiotics

Abstract:

The purpose of this study was to investigate the efficacy of intravenous nutrient(soluvit, vitalipid, aminoven infant, lipovenos) and bisolvon commonly used in neonatal intensive care units against biofilm cells of staphylococcus aureus, Staphylococcus epidermidis, Pseudomonas aerguinosa and klebseilla pneumonia as they are the most commonly isolated organisms and are biofilm producers. Also, the synergetic acticity of soluvit, heparin, bisolvon with antibiotics and its effect on minimum biofilm eradication concentration(MBEC) was tested. Intravenous nutrient and bromohexine are widely used in newborns. Numbers of viable cell count released from biofilm after treatment with intravenous nutrient and bromohexine were counted to compare the efficacy. The percentage of reduction in biofilm regrowth in case of using soluvit was 43-51% and 36-42 % for Gram positive and Gram negative respectively, on adding the vitalipid the percentage was 45-50 %and 37-41% for Gram positive and Gram negative respectively. While, in case of using bisolvon the percentage was 46-52% and 47-48% for Gram positive and Gram negative respectively. Adding lipovenos had a reduction percentage of 48-52% and 48-49% for Gram positive and Gram negative respectively. While, adding aminoven infant the percentage was 10-15% and 9-11% for Gram positive and Gram negative respectively. Adding soluvit, heparin and bisolvon to antibiotics had synergic effect. soluvit with ciprofloxacin has 8-16 times decrease than minimum biofilm eradication concentration (MBEC) for ciprofloxacin alone. While, by adding soluvit to vancomycin the MBEC reduced by 16 times than MBEC of vancomycin alone. In case of combination soluvit with cefotaxime, amikacin and gentamycin the reduction in MBEC was 16, 8 and 6-32 times respectively. The synergetic effect of adding heparin to ciprofloxacin, vancomycin, cefotaxime, amikacin and gentamicin was 2 times reduction with all except in case of gram negative the range of reduction was 0-2 with both gentamycin and ciprofloxacin. Bisolvon exihited synergetic effect with ciprofloxacin, vancomycin, cefotaxime, amikacin and gentamicin by 16, 32, 32, 8, 32-64 and 32 times decrease in MBEC respectively.

1 Predictive Modelling of Curcuminoid Bioaccessibility as a Function of Food Formulation and Associated Properties

Abstract:

Background: The bioaccessibility of bioactive compounds is a critical determinant of the nutritional quality of various food products. Despite its importance, there is a limited number of comprehensive studies aimed at assessing how the composition of a food matrix influences the bioaccessibility of a compound of interest. This knowledge gap has prompted a growing need to investigate the intricate relationship between food matrix formulations and the bioaccessibility of bioactive compounds. One such class of bioactive compounds that has attracted considerable attention is curcuminoids. These naturally occurring phytochemicals, extracted from the roots of Curcuma longa, have gained popularity owing to their purported health benefits and also well known for their poor bioaccessibility Project aim: The primary objective of this research project is to systematically assess the influence of matrix composition on the bioaccessibility of curcuminoids. Additionally, this study aimed to develop a series of predictive models for bioaccessibility, providing valuable insights for optimising the formula for functional foods and provide more descriptive nutritional information to potential consumers. Methods: Food formulations enriched with curcuminoids were subjected to in vitro digestion simulation, and their bioaccessibility was characterized with chromatographic and spectrophotometric techniques. The resulting data served as the foundation for the development of predictive models capable of estimating bioaccessibility based on specific physicochemical properties of the food matrices. Results: One striking finding of this study was the strong correlation observed between the concentration of macronutrients within the food formulations and the bioaccessibility of curcuminoids. In fact, macronutrient content emerged as a very informative explanatory variable of bioaccessibility and was used, alongside other variables, as predictors in a Bayesian hierarchical model that predicted curcuminoid bioaccessibility accurately (optimisation performance of 0.97 R2) for the majority of cross-validated test formulations (LOOCV of 0.92 R2). These preliminary results open the door to further exploration, enabling researchers to investigate a broader spectrum of food matrix types and additional properties that may influence bioaccessibility. Conclusions: This research sheds light on the intricate interplay between food matrix composition and the bioaccessibility of curcuminoids. This study lays a foundation for future investigations, offering a promising avenue for advancing our understanding of bioactive compound bioaccessibility and its implications for the food industry and informed consumer choices.
