7 In vitro Plant Regeneration of Java Vetiver (Vetiveria zizanioides)
Authors: Iriawati, R. R. Esyanti, W. Natalia, N. Zahya
Abstract:
In vitro plant regeneration has been successfully obtained from basal shoot explant of Vetiveria zizanioides through indirect organogenesis. The explant was cultured in Murashige & Skoog’s (MS) media supplemented with 2,4-D, IAA, and kinetin in various concentrations. Callus was well induced in media supplemented with 2 ppm 2,4-D, 1 ppm IAA, and 1 ppm kinetin. This callus was then transferred to MS media supplemented with 1 - 5 ppm of BAP for shoot regeneration. The media supplemented with 3 ppm BAP was a suitable medium for shoot induction, as well as for shoot multiplication. Rooting was well developed in shoot following transferred to half MS media containing 0.2 ppm IBA. Plantlet was then transferred to husk charcoal for acclimatization, and almost all (90%) of plantlets were survived during acclimatization.

Authors: A. A. M. Azoddein, R. M. Yunus, N. M. Sulaiman, A. B. Bustary, K. Sabar
Abstract:
Microbes have been used to solve environmental problems for many years. The role of microorganism to sequester, precipitate or alter the oxidation state of various heavy metals has been extensively studied. Treatment using microorganism interacts with toxic metal are very diverse. The purpose of this research is to remove the mercury using Pseudomonas putida (P. putida), pure culture ATTC 49128 at optimum growth parameters such as techniques of culture, acclimatization time and speed of incubator shaker. Thus, in this study, the optimum growth parameters of P. putida were obtained to achieve the maximum of mercury removal. Based on the optimum parameters of P. putida for specific growth rate, the removal of two different mercury concentration, 1 ppm and 4 ppm were studied. From mercury nitrate solution, a mercuryresistant bacterial strain which is able to reduce from ionic mercury to metallic mercury was used to reduce ionic mercury. The overall levels of mercury removal in this study were between 80% and 89%. The information obtained in this study is of fundamental for understanding of the survival of P. putida ATTC 49128 in mercury solution. Thus, microbial mercury removal is a potential bioremediation for wastewater especially in petrochemical industries in Malaysia. Keywords: Pseudomonas putida, growth kinetic, biosorption, mercury, petrochemical wastewater.

Authors: Atef M. Ghaleb, Mohamed Z. Ramadan, Khalid Saad Aljaloud
Abstract:
The aim of this study is to assesses the lifting capabilities of persons experiencing hypoxia. It also examines the behavior of the physiological response induced through the lifting process related to changing in the hypoxia and lifting frequency variables. For this purpose, the study performed two consecutive tests by using; (1) training and acclimatization; and (2) an actual collection of data. A total of 10 male students from King Saud University, Kingdom of Saudi Arabia, were recruited in the study. A two-way repeated measures design, with two independent variables (ambient oxygen (15%, 18% and 21%)) and lifting frequency (1 lift/min and 4 lifts/min) and four dependent variables i.e., maximum acceptable weight of lift (MAWL), Electromyography (EMG) of four muscle groups (anterior deltoid, trapezius, biceps brachii, and erector spinae), rating of perceived exertion (RPE), and rating of oxygen feeling (ROF) were used in this study. The results show that lifting frequency has significantly impacted the MAWL and muscles’ activities. The oxygen content had a significant effect on the RPE and ROE. The study has revealed that acclimatization and training sessions significantly reduce the effect of the hypoxia on the human physiological parameters during the manual materials handling tasks.

Authors: L. Al-Mulla, N. R. Bhat, M. Khalil
Abstract:
A study was conducted in greenhouse environment to determine the response of five tissue-cultured date palm cultivars, Al- Ahamad, Nabusaif, Barhee, Khalas, and Kasab to irrigation water salinity of 1.6, 5, 10, or 20 dS/ m. The salinity level of 1.6dS/m, was used as a control. The effects of high salinity on plant survival were manifested at 360 days after planting (DAP) onwards. Three cultivars, Khalas, Kasab and Barhee were able to tolerate 10 dS/m salinity level at 24 months after the start of study. Khalas tolerated the highest salinity level of 20 dS/ m and 'Nabusaif' was found to be the least tolerant cv. The average heights of palms and the number of fronds were decreased with increasing salinity levels as time progressed.

Authors: E. Sargsyan, A. Vardanyan, L. Ghalachyan, S. Bulgadaryan
Abstract:
Our results showed that for the growth of qualitative seedling and vegetative raw material of ðó. marschallianus Willd. and T. serphyllum L. it is more profitable to use the in vitro and hydroponics combined method. In in vitro culture it is possible to do micro-propagation whole year with 98-99% rhizogenesis. 30000 micro-plants were obtained from one explant during 9 months. Hydroponic conditions provide the necessary microclimate for microplants where the survival rate without acclimatization was 93.3%. The essential oil content in hydroponic dry herb of both species in vegetative and blossom phase was 1.3% whereas in wild plants it was 1.2%, the content of extractive substances and vitamin C also exceeded wild plants. Our biochemical and radiochemical investigations indicated that the medicinal raw materials obtained from hydroponic and wild plants of Thymus species correspond to the demands of SPh XI, and the content of artificial radionuclides does not exceed the MACL.Keywords: Hydroponics, In vitro, Micro-propagation, Thymus

Authors: M. F. R. Zuthi, H. H. Ngo, W. S. Guo, T. T. Nguyen
Abstract:
A novel sponge submerged membrane bioreactor (SSMBR) was developed to effectively remove organics and nutrients from wastewater. Sponge is introduced within the SSMBR as a medium for the attached growth of biomass. This paper evaluates the effects of new and acclimatized sponges for dissolved organic carbon (DOC) removal from wastewater at different mixed liquor suspended solids- (MLSS) concentration of the sludge. It was observed in a series of experimental studies that the acclimatized sponge performed better than the new sponge whilst the optimum DOC removal could be achieved at 10g/L of MLSS with the acclimatized sponge. Moreover, the paper analyses the relationships between the MLSSsponge/MLSSsludge and the DOC removal efficiency of SSMBR. The results showed a non-linear relationship between the biomass parameters of the sponge and the sludge, and the DOC removal efficiency of SSMBR. A second-order polynomial function could reasonably represent these relationships. Keywords: Acclimatization, Dissolved organic carbon, Mathematical model, Sponge submerged membrane bioreactor.

Authors: T. Punjansing, M. Nakkuntod, S. Homchan, P. Inthima, A. Kongbangkerd
Abstract:
The influence of organic supplements on growth and multiplication efficiency of Phaius tankervilleae var. alba seedlings was investigated. 12 week-old seedlings were cultured on half-strength semi-solid Murashige and Skoog (MS) medium supplemented with 30 g/L sucrose, 8 g/L agar and various concentrations of coconut water (0, 50, 100, 150 and 200 mL/L) combined with potato extract (0, 25 and 50 g/L) and the pH was adjusted to 5.8 prior to autoclaving. The cultures were then kept under constant photoperiod (16 h light: 8 h dark) at 25 ± 2 °C for 12 weeks. The highest number of shoots (3.0 shoots/explant) was obtained when cultured on the medium added with 50 ml/L coconut water and 50 g/L potato extract whereas the highest number of leaves (5.9 leaves/explant) and roots (6.1 roots/explant) could receive on the medium supplemented with 150 ml/L coconut water and 50 g/L potato extract. with 150 ml/L coconut water and 50 g/L potato extract. Additionally, plantlets of P. tankervilleae var. alba were transferred to grow into seven different substrates i.e. soil, sand, coconut husk chip, soil-sand mix (1: 1), soil-coconut husk chip mix (1: 1), sand-coconut husk chip mix (1: 1) and soil-sand-coconut husk chip mix (1: 1: 1) for four weeks. The results found that acclimatized plants showed 100% of survivals when sand, coconut husk chip and sand-coconut husk chip mix are used as substrates. The number of leaves induced by sand-coconut husk chip mix was significantly higher than that planted in other substrates (P > 0.05). Meanwhile, no significant difference in new shoot formation among these substrates was observed (P < 0.05). This precursory developing protocol was likely to be applied for more large scale of plant production as well as conservation of germplasm of this orchid species.
