3 The Effect of Curcumin on Cryopreserved Bovine Semen

Abstract:
Oxidative stress associated with semen cryopreservation may result in lipid peroxidation (LPO), DNA damage and apoptosis, leading to decreased sperm motility and fertilization ability. Curcumin (CUR), a natural phenol isolated from Curcuma longa Linn. has been presented as a possible supplement for a more effective semen cryopreservation because of its antioxidant properties. This study focused to evaluate the effects of CUR on selected oxidative stress parameters in cryopreserved bovine semen. 20 bovine ejaculates were split into two aliquots and diluted with a commercial semen extender containing CUR (50 μmol/L) or no supplement (control), cooled to 4 °C, frozen and kept in liquid nitrogen. Frozen straws were thawed in a water bath for subsequent experiments. Computer assisted semen analysis was used to evaluate spermatozoa motility, and reactive oxygen species (ROS) generation was quantified by using luminometry. Superoxide generation was evaluated with the NBT test, and LPO was assessed via the TBARS assay. CUR supplementation significantly (P<0.001) increased the spermatozoa motility and provided a significantly higher protection against ROS (P<0.001) or superoxide (P<0.01) overgeneration caused by semen freezing and thawing. Furthermore, CUR administration resulted in a significantly (P<0.01) lower LPO of the experimental semen samples. In conclusion, CUR exhibits significant ROS-scavenging activities which may prevent oxidative insults to cryopreserved spermatozoa and thus may enhance the post-thaw functional activity of male gametes.Keywords: bulls, cryopreservation, curcumin, lipid peroxidation, reactive oxygen species, spermatozoa

Abstract:
A semiflow is a triple consisting of a Hausdorff topological space $X$, a commutative topological monoid $T$ and a continuous monoid action of $T$ on $X$. The acting monoid $T$ is usually either the discrete monoid $\N_0$ of nonnegative integers (in which case the semiflow can be defined as a pair $(X,f)$ consisting of a phase space $X$ and a continuous function $f:X\to X$), or the monoid $\R_+$ of nonnegative real numbers (the so-called one-parameter monoid). However, it turns out that there are real-life situations where it is useful to consider the acting monoids that are a combination of discrete and continuous monoids. That, for example, happens, when we are observing certain dynamical system at discrete moments, but after some time realize that it would be beneficial to continue our observations in real time. The acting monoid in that case would be $T=\{0, t_0, 2t_0, \dots, (n-1)t_0\} \cup [nt_0,\infty)$ with the operation and topology induced from real numbers. This partly explains the motivation for the level of generality which is pursued in our research. We introduce the PSP monoids, which include all but ``pathological'' monoids, and most of our statements hold for them. The topic of our presentation are some recent results about chaos-related properties in semiflows, indecomposability and sensitivity of semiflows in the described general context.Keywords: chaos, indecomposability, PSP monoids, semiflow, sensitivity

Abstract:
The resource-efficient planning of the complex production planning processes in the meat industry and the reduction of food waste is a permanent challenge. The complexity of the production planning process occurs in every part of the supply chain, from agriculture to the end consumer. It arises from long and uncertain planning phases. Uncertainties such as stochastic yields, fluctuations in demand, and resource variability are part of this process. In the meat industry, waste mainly relates to incorrect storage, technical causes in production, or overproduction. The high amount of food waste along the complex supply chain in the meat industry could not be reduced by simple solutions until now. Therefore, resource-efficient production planning by conventional methods is currently only partially feasible. The realization of intelligent, automated production planning is basically possible through the application of machine learning algorithms, such as those of reinforcement learning. By applying the adapted design thinking method, machine learning methods (especially reinforcement learning algorithms) are used for the complex production planning process in the meat industry. This method represents a concretization to the application area. A resource-efficient production planning process is made available by adapting the design thinking method. In addition, the complex processes can be planned efficiently by using this method, since this standardized approach offers new possibilities in order to challenge the complexity and the high time consumption. It represents a tool to support the efficient production planning in the meat industry. This paper shows an elegant adaption of the design thinking method to apply the reinforcement learning method for a resource-efficient production planning process in the meat industry. Following, the steps that are necessary to introduce machine learning algorithms into the production planning of the food industry are determined. This is achieved based on a case study which is part of the research project ”REIF - Resource Efficient, Economic and Intelligent Food Chain” supported by the German Federal Ministry for Economic Affairs and Climate Action of Germany and the German Aerospace Center. Through this structured approach, significantly better planning results are achieved, which would be too complex or very time consuming using conventional methods.Keywords: change management, design thinking method, machine learning, meat industry, reinforcement learning, resource-efficient production planning
