5 Molecular Profiling and Potential Bioactive Characteristics of Endophytic Fungi Isolated from Leptadenia Pyrotechnica

Abstract:
Endophytes are organisms that colonize internal plant tissues without causing apparent harm to their host. Almost all groups of microorganisms have been found in endophytic association with plants may be fungi. They stimulate the production of secondary metabolites with a diverse range of biological activities. Leptadenia pyrotechnica is a more or less leafless, erect shrub with straight stems which is highly distributed in Saudi Arabia. Four endophytes fungi were isolated from Leptadenia pyrotechnica and identified using 18S ribosomal RNA sequences, which revealed four fungi genuses, namely Aspergillus terreus; Aspergillus welwitschiae; Aspergillus fumigatus and Aspergillus flavus. In this present study, four endophytic fungi from Leptadenia pyrotechnica were used for obtaining crude aqueous and ethyl acetate extracts for antimicrobial screening against 6 human pathogens, the antibacterial tests presented satisfactory results, where the pathogenic bacteria were inhibited by the four extracts tested, except for Escherichia coli that was inhibited by all extracts except ethyl acetate extract of Aspergillus terreus. Analysis of variance showed that the extract produced by endophyte Leptadenia pyrotechnica was the most effective against all bacteria, either gram-negative or positive. However, the extract was not efficient against pathogenic fungi. Therefore, this study indicates that endophytes from medicinal plant Leptadenia pyrotechnica could be potential sources of antibacterial substances.Keywords: antimicrobial activity, Aspergillus sp, endophytes, Leptadenia pyrotechnica

Abstract:
This study investigated the effects of Acacia Senegal, Calotropis procera, Leptadenia pyrotechnica, Ziziphus spina Christi, Balanites aegyptiaca, Indigofera oblongigolia, Arachis hypogea and Sesimum indicum grown in the western region of White Nile State on soil properties of the 0-10, 10-30, 30-60 and 60-90 cm depths. Soil properties were: pH(paste), electrical conductivity of the saturation extract (ECe), total N (TN), organic carbon (OC), soluble K, available P, aggregate stability and water holding capacity. Triplicate Soil samples were collected after the end of the rainy season using 5 cm diameter auger. Results indicated that pH, ECe and TN were not significantly different among plant cover types. In the top 10-30 cm depth, OC under all types was significantly higher than the control (4.1 to 7.7 fold). The highest (0.085%) OC was found under the Z. spina Christi and A. Senegal whereas the lowest (0.045%) was reported under the A. hypogea. In the 10-30 cm depth, with the exception of A. hypogea, Z. spina christi and S. indicum, P content was almost similar but significantly higher than the control by 72 to 129%. In the 10-30 cm depth, K content under the S. indicum (0.46 meq/L) was exceptionally high followed by Z. spina christi (0.102 meq/L) as compared to the control (0.029 meq/L). Water holding capacity and aggregate stability of the top 0-10 cm depth were not significantly different among plant cover types. Based on the fact that accumulation of organic matter in the soil profile of any ecosystem is an important indicator of soil quality, results of this study may conclude that (1) cultivation of A.senegal, B.aegyptiaca and Z. spina Christi improved soil quality whereas (2) cultivation of A. hypogea or soil that is solely invaded with C. procera and L.pyrotechnica may induce soil degradation.Keywords: canopy, crops, shrubs, soil properties, trees

Abstract:
This project involved screening for antibacterial activity, phytochemical and mineral properties of Leptadenia hastata by flame photometry. The result of phytochemical screening reveals that the presence of flavonoids, tannins, saponins, alkaloids, steroidal, and anthraquinones while the cardiac glycoside was absent. This justifies the plant been used as anti-bleeding and anti-inflammatory agents. The result of flame photometry revealed that 1.85 % (Na), 0.65% (K) and 1.85 % (Ca) which indicates the safe nature of the plant extract as such could be used to lower high blood pressure. The antibacterial properties of both the aqueous and ethanolic extract were studied against some bacteria, Escherichia coli, Bacillus Cercus, Pseudomonas aeruginas, and Enterobacter aerogegens, by disc diffusion method and the result reveals that there are very good activities against the organism while the ethanolic extract at concentration 1.0 – 1.2 mg/ml. the ethanolic extract showed in considerable zone inhibition against bacteria’s; Escherichia coli, Bacillus Cercus, pseudomonas aeruginosa andklebsellapnemuoniae. Minimum inhibitory concentration (MIC) and minimum Bacterial concentration (MBC) were conducted with fairly good significant effect of inhibition on the organism, therefore, plant extract could be a potential source of antibacterial agent.Keywords: antibacterial activity, Leptadenia hastata, infectious diseases, phytochemical screening

Abstract:
The study was conducted to assess heifer camel behavior and live weight changes on native range supplemented with different energy levels. A total of nine camels aged between 2 and 3 years were randomly allotted into three groups and supplemented with 3400, 3600 and 3800 Kcal and designated A, B and C, respectively. The data obtained was analyzed for variance in a Completely Randomized Design. The heifers utilized average of 371.70 min/day (64% of daylight time) browsing on native pasture and 2.30 min/day (6%) sand bathing. A significantly higher mean time was spent by heifers on browsing Leptadenia hastata (P<0.001), Dichrostachys cinerea (P<0.01), Acacia nilotica (P<0.001) and Ziziphus spina-christi (P<0.05) in early dry season (January). No significant difference was recorded on browsing time on Tamarindus indica, Adansonia digitata, Piliostigma reticulatum, Parkia biglobosaand Azadirachta indica. No significant (P<0.05) liveweight change was recorded on she-camels due to the three energy levels. It was concluded that nutritive browse species in the study area could meet camel nutrient requirements including energy. Further research on effect of period on camel nutrients requirement in different physiological conditions is recommended.Keywords: heifer, camel, grazing, pasture

Abstract:
The Thar Desert of Rajasthan covers a wide geographical area spreading between 23.3° to 30.12°, North latitude and 69.3◦ to 76◦ Eastern latitudes; having a unique spectrum of arid zone vegetation. This desert is spreading over 12 districts having a rich source of economically important/threatened plant diversity interacting and growing with adverse climatic conditions of the area. Due to variable geological, physiographic, climatic, edaphic and biotic factors, the arid zone medicinal flora exhibit a wide collection of angiosperm families. The herbal diversity of this arid region is medicinally important in household remedies among tribal communities as well as in traditional systems. The on-going increasing disturbances in natural ecosystems are due to climatic and biological, including anthropogenic factors. The unique flora and subsequently dependent faunal diversity of the desert ecosystem is losing its biotic potential. A large number of plants have no future unless immediate steps are taken to arrest the causes, leading to their biological improvement. At present the potential loss in ecological amplitude of various genera and species is making several plant species as red listed plants of arid zone vegetation such as Commmiphora wightii, Tribulus rajasthanensis, Calligonum polygonoides, Ephedra foliata, Leptadenia reticulata, Tecomella undulata, Blepharis sindica, Peganum harmala, Sarcostoma vinimale, etc. Mostly arid zone species are under serious pressure against prevailing ecosystem factors to continuation their life cycles. Genetic, molecular, cytological, biochemical, metabolic, reproductive, germination etc. are the several points where the floral diversity of the arid zone area is facing severe ecological influences. So, there is an urgent need to conserve them. There are several opportunities in the field to carry out remarkable work at particular levels to protect the native plants in their natural habitat instead of only their in vitro multiplication.Keywords: ecology, evaluation, xerophytes, economically, threatened plants, conservation
