1116 Cloning of a β-Glucosidase Gene (BGL1) from Traditional Starter Yeast Saccharomycopsis fibuligera BMQ 908 and Expression in Pichia pastoris

Abstract:

β-Glucosidase is an important enzyme for production of ethanol from lignocellulose. With hydrolytic activity on cellooligosaccharides, especially cellobiose, β-glucosidase removes product inhibitory effect on cellulases and forms fermentable sugars. In this study, β-glucosidase encoding gene (BGL1) from traditional starter yeast Saccharomycosis fibuligera BMQ908 was cloned and expressed in Pichia pastoris. BGL1 of S. fibuligera BMQ 908 shared 98% nucleotide homology with the closest GenBank sequence (M22475) but identity in amino-acid sequences of catalytic domains. Recombinant plasmid pPICZαA/BGL1 containing the sequence encoding BGL1 mature protein and α-factor secretion signal was constructed and transformed into methylotrophic yeast P. pastoris by electroporation. The recombinant strain produced single extracellular protein with molecular weight of 120 kDa and cellobiase activity of 60 IU/ml. The optimum pH of the recombinant β-glucosidase was 5.0 and the optimum temperature was 50°C.

1115 All Types of Base Pair Substitutions Induced by γ-Rays in Haploid and Diploid Yeast Cells

Abstract:

We study the biological effects induced by ionizing radiation in view of therapeutic exposure and the idea of space flights beyond Earth's magnetosphere. In particular, we examine the differences between base pair substitution induction by ionizing radiation in model haploid and diploid yeast Saccharomyces cerevisiae cells. Such mutations are difficult to study in higher eukaryotic systems. In our research, we have used a collection of six isogenic trp5-strains and 14 isogenic haploid and diploid cyc1-strains that are specific markers of all possible base-pair substitutions. These strains differ from each other only in single base substitutions within codon-50 of the trp5 gene or codon-22 of the cyc1 gene. Different mutation spectra for two different haploid genetic trp5- and cyc1-assays and different mutation spectra for the same genetic cyc1-system in cells with different ploidy — haploid and diploid — have been obtained. It was linear function for dose-dependence in haploid and exponential in diploid cells. We suggest that the differences between haploid yeast strains reflect the dependence on the sequence context, while the differences between haploid and diploid strains reflect the different molecular mechanisms of mutations.

1114 Selection of Pichia kudriavzevii Strain for the Production of Single-Cell Protein from Cassava Processing Waste

Abstract:

A total of 115 yeast strains isolated from local cassava processing wastes were measured for crude protein content. Among these strains, the strain MSY-2 possessed the highest protein concentration (>3.5 mg protein/mL). By using molecular identification tools, it was identified to be a strain of Pichia kudriavzevii based on similarity of D1/D2 domain of 26S rDNA region. In this study, to optimize the protein production by MSY-2 strain, Response Surface Methodology (RSM) was applied. The tested parameters were the carbon content, nitrogen content, and incubation time. Here, the value of regression coefficient (R2) = 0.7194 could be explained by the model which is high to support the significance of the model. Under the optimal condition, the protein content was produced up to 3.77 g per L of the culture and MSY-2 strain contains 66.8 g protein per 100 g of cell dry weight. These results revealed the plausibility of applying the novel strain of yeast in single-cell protein production.

1112 Microbial Oil Production by Monoculture and Mixed Cultures of Microalgae and Oleaginous Yeasts using Sugarcane Juice as Substrate

Abstract:

Monoculture and mixed cultures of microalgae and the oleaginous yeast for microbial oil productions were investigated using sugarcane juice as carbon substrate. The monoculture of yeast Torulaspora maleeae Y30, Torulaspora globosa YU5/2 grew faster than that of microalgae Chlorella sp. KKU-S2. In monoculture of T. maleeae Y30, a biomass of 8.267g/L with lipid yield of 0.920g/L were obtained, while 8.333g/L of biomass with lipid yield of 1.141g/L were obtained for monoculture of T. globosa YU5/2. A biomass of 1.933g/L with lipid yield of 0.052g/L was found for monoculture of Chlorella sp. KKU-S2. The biomass concentration in the mixed culture of the oleaginous yeast with microalgae increased faster and was higher compared with that in the monocultures. A biomass of 8.733g/L with lipid yield of 1.564g/L was obtained for a mixed culture of T. maleeae Y30 with Chlorella sp. KKU-S2, while 8.010g/L of biomass with lipid yield of 2.424g/L was found for mixed culture of T. globosa YU5/2 with Chlorella sp. KKU-S2. Maximum cell yield coefficient (YX/S, g/L) was found of 0.323 in monoculture of Chlorella sp. KKU-S2 but low level of both specific yield of lipid (YP/X, g lipid/g cells) of 0.027 and volumetric lipid production rate (QP, g/L/d) of 0.003 were observed. While, maximum YP/X (0.303), QP (0.105) and maximum process product yield (YP/S, 0.061) were obtained in mixed culture of T. globosa YU5/2 with Chlorella sp. KKU-S2. The results obtained from the study shows that mixed culture of yeast with microalgae is a desirable cultivation process for microbial oil production.

1111 Application of Thermoplastic Microbioreactor to the Single Cell Study of Budding Yeast to Decipher the Effect of 5-Hydroxymethylfurfural on Growth

Abstract:

Yeast cells are generally used as a model system of eukaryotes due to their complex genetic structure, rapid growth ability in optimum conditions, easy replication and well-defined genetic system properties. Thus, yeast cells increased the knowledge of the principal pathways in humans. During fermentation, carbohydrates (hexoses and pentoses) degrade into some toxic by-products such as 5-hydroxymethylfurfural (5-HMF or HMF) and furfural. HMF influences the ethanol yield, and ethanol productivity; it interferes with microbial growth and is considered as a potent inhibitor of bioethanol production. In this study, yeast single cell behavior under HMF application was monitored by using a continuous flow single phase microfluidic platform. Microfluidic device in operation is fabricated by hot embossing and thermo-compression techniques from cyclo-olefin polymer (COP). COP is biocompatible, transparent and rigid material and it is suitable for observing fluorescence of cells considering its low auto-fluorescence characteristic. The response of yeast cells was recorded through Red Fluorescent Protein (RFP) tagged Nop56 gene product, which is an essential evolutionary-conserved nucleolar protein, and also a member of the box C/D snoRNP complexes. With the application of HMF, yeast cell proliferation continued but HMF slowed down the cell growth, and after HMF treatment the cell proliferation stopped. By the addition of fresh nutrient medium, the yeast cells recovered after 6 hours of HMF exposure. Thus, HMF application suppresses normal functioning of cell cycle but it does not cause cells to die. The monitoring of Nop56 expression phases of the individual cells shed light on the protein and ribosome synthesis cycles along with their link to growth. Further computational study revealed that the mechanisms underlying the inhibitory or inductive effects of HMF on growth are enriched in functional categories of protein degradation, protein processing, DNA repair and multidrug resistance. The present microfluidic device can successfully be used for studying the effects of inhibitory agents on growth by single cell tracking, thus capturing cell to cell variations. By metabolic engineering techniques, engineered strains can be developed, and the metabolic network of the microorganism can thus be manipulated such that chemical overproduction of target metabolite is achieved along with the maximum growth/biomass yield.

1110 Screening of Antagonistic/Synergistic Effect between Lactic Acid Bacteria (LAB) and Yeast Strains Isolated from Kefir

Abstract:

Kefir is a traditional fermented refreshing beverage which is known for its valuable and beneficial properties for human health. Mainly yeast species, lactic acid bacteria (LAB) strains and fewer acetic acid bacteria strains live together in a natural matrix named “kefir grain”, which is formed from various proteins and polysaccharides. Different microbial species live together in slimy kefir grain and it has been thought that synergetic effect could take place between microorganisms, which belong to different genera and species. In this research, yeast and LAB were isolated from kefir samples obtained from Uludag University Food Engineering Department. The cell morphology of isolates was screened by microscopic examination. Gram reactions of bacteria isolates were determined by Gram staining method, and as well catalase activity was examined. After observing the microscopic/morphological and physical, enzymatic properties of all isolates, they were divided into the groups as LAB and/or yeast according to their physicochemical responses to the applied examinations. As part of this research, the antagonistic/synergistic efficacy of the identified five LAB and five yeast strains to each other were determined individually by disk diffusion method. The antagonistic or synergistic effect is one of the most important properties in a co-culture system that different microorganisms are living together. The synergistic effect should be promoted, whereas the antagonistic effect is prevented to provide effective culture for fermentation of kefir. The aim of this study was to determine microbial interactions between identified yeast and LAB strains, and whether their effect is antagonistic or synergistic. Thus, if there is a strain which inhibits or retards the growth of other strains found in Kefir microflora, this circumstance shows the presence of antagonistic effect in the medium. Such negative influence should be prevented, whereas the microorganisms which have synergistic effect on each other should be promoted by combining them in kefir grain. Standardisation is the most desired property for industrial production. Each microorganism found in the microbial flora of a kefir grain should be identified individually. The members of the microbial community found in the glue-like kefir grain may be redesigned as a starter culture regarding efficacy of each microorganism to another in kefir processing. The main aim of this research was to shed light on more effective production of kefir grain and to contribute a standardisation of kefir processing in the food industry.

1109 Threshold Stress of the Soil Subgrade Evaluation for Highway Formations

Abstract:

The objective of this study is to evaluate the threshold stress of the clay with sand subgrade soil. Threshold stress can be defined as the stress level above which cyclic loading leads to excessive deformation and eventual failure. The thickness determination of highways formations using the threshold stress approach is a more realistic assessment of the soil behaviour because it is subjected to repeated loadings from moving vehicles. Threshold stress can be evaluated by plastic strain criterion, which is based on the accumulated plastic strain behaviour during cyclic loadings [1]. Several conditions of the all-round pressure the subgrade soil namely, zero confinement, low all-round pressure and high all-round pressure are investigated. The threshold stresses of various soil conditions are determined. Threshold stress of the soil are 60%, 31% and 38.6% for unconfined partially saturated sample, low effective stress saturated sample, high effective stress saturated sample respectively.

1108 Sophorolipids Production by Candida Bombicola using Synthetic Dairy Wastewater

Abstract:

Sophorolipids (SLs) production by the yeast Candida bombicola was studied in batch shake flasks using synthetic dairy wastewaters (SDWW) with or without any added external carbon and nitrogen sources. A maximum SLs production of 38.76 g/l was observed with the SDWW supplemented with low cost substrate of sugarcane molasses at 50 g/l and soybean oil at 50 g/l. When the SDWW was supplemented with more costly glucose, yeast extract, urea and soybean oil, the production, however, got lowered to only 29.49 g/l, but with a maximum biomass production of 17.38 g/l together with a complete utilization of the carbon sources.

1107 Identification of Binding Proteins That Interact with BVDV E2 Protein in Bovine Trophoblast Cell

Abstract:

Bovine viral diarrhea virus (BVDV) can cause lifelong persistent infection. One reason for the phenomena is attributed to BVDV infection to placenta tissue. However the mechanisms that BVDV invades into placenta tissue remain unclear. To clarify the molecular mechanisms, we investigated the possible means that BVDV entered into bovine trophoblast cells (TPC). Yeast two-hybrid system was used to identify proteins extracted from TPC, which interact with BVDV envelope glycoprotein E2. A PGbkt7-E2 yeast expression vector and TPC cDNA library were constructed. Through two rounds of screening, three positive clones were identified. Sequencing analysis indicated that all the three positive clones encoded the same protein clathrin. Physical interaction between clathrin and BVDV E2 protein was further confirmed by coimmunoprecipitation experiments. This result suggested that the clathrin might play a critical role in the process of BVDV entry into placenta tissue and might be a novel antiviral target for preventing BVDV infection.

1106 The Effect of Saccharomyces cerevisiae Live Yeast Culture on Microbial Nitrogen Supply to Small Intestine in Male Kivircik Yearlings Fed with Different Forage-Concentrate Ratios

Abstract:

The aim of the study was to investigate the effect of Saccharomyces cerevisiae (SC) live yeast culture on microbial protein supply to small intestine in Kivircik male yearlings when fed with different ratio of forage and concentrate diets. Four Kivircik male yearlings with permanent rumen canula were used in the experiment. The treatments were allocated to a 4x4 Latin square design. Diet I consisted of 70% alfalfa hay and 30% concentrate, Diet II consisted of 30% alfalfa hay and 70% concentrate, Diet I and II were supplemented with a SC. Daily urine was collected and stored at -20°C until analysis. Calorimetric methods were used for the determination of urinary allantoin and creatinine levels. The estimated microbial N supply to small intestine for Diets I, I+SC, II and II+SC were 2.51, 2.64, 2.95 and 3.43 g N/d respectively. Supplementation of Diets I and II with SC significantly affected the allantoin levels in μmol/W0.75 (p<0.05). Mean creatinine values in μmol/W0.75 and allantoin:creatinine ratios were not significantly different among diets. In conclusion, supplementation with SC live yeast culture had a significant effect on urinary allantoin excretion and microbial protein supply to small intestine in Kivircik yearlings fed with high concentrate Diet II (P<0.05). Hence urinary allantoin excretion may be used as a tool for estimating microbial protein supply in Kivircık yearlings. However, further studies are necessary to understand the metabolism of Saccharomyces cerevisiae live yeast culture with different forage:concentrate ratio in Kıvırcık Yearlings.

1105 Effect of Nano-Silver on Growth of Saffron in Flooding Stress

Abstract:

Saffron (Crocus sativus) is cultivated as spices, medicinal and aromatic plant species. At autumn season, heavy rainfall can cause flooding stress and inhibits growth of saffron. Thus this research was conducted to study the effect of silver ion (as an ethylene inhibitor) on growth of saffron under flooding conditions. The corms of saffron were soaked with one concentration of nano silver (0, 40, 80 or 120 ppm) and then planting under flooding stress or non flooding stress conditions. Results showed that number of roots, root length, root fresh and dry weight, leaves fresh and dry weight were reduced by 10 day flooding stress. Soaking saffron corms with 40 or 80 ppm concentration of nano silver rewarded the effect of flooding stress on the root number, by increasing it. Furthermore, 40 ppm of nano silver increased root length in stress. Nano silver 80 ppm in flooding stress, increased leaves dry weight.

1104 Dissimilar Materials Joint and Effect of Angle Junction on Stress Distribution at Interface

Abstract:

in dissimilar material joints, failure often occurs along the interface between two materials due to stress singularity. Stress distribution and its concentration depend on materials and geometry of the junction. Inhomogenity of stress distribution at the interface of junction of two materials with different elastic modules and stress concentration in this zone are the main factors resulting in rupture of the junction. Effect of joining angle in the interface of aluminum-polycarbonate will be discussed in this paper. Computer simulation and finite element analysis by ABAQUS showed that convex interfacial joint leads to stress reduction at junction corners in compare with straight joint. This finding is confirmed by photoelastic experimental results.

1103 Estimation of Stress Intensity Factors from Near Crack Tip Field

Abstract:

All current experimental methods for determination of stress intensity factors are based on the assumption that the state of stress near the crack tip is plane stress. Therefore, these methods rely on strain and displacement measurements made outside the near crack tip region affected by the three-dimensional effects or by process zone. In this paper, we develop and validate an experimental procedure for the evaluation of stress intensity factors from the measurements of the out-of-plane displacements in the surface area controlled by 3D effects. The evaluation of stress intensity factors is possible when the process zone is sufficiently small, and the displacement field generated by the 3D effects is fully encapsulated by K-dominance region.

1102 Response of Chickpea (Cicer arietinum L.) Genotypes to Drought Stress at Different Growth Stages

Chickpea (Cicer arietinum L.) is one of the important grain legume crops in the world. However, drought stress is a serious threat to chickpea production, and development of drought-resistant varieties is a necessity. Field experiments were conducted to evaluate the response of 8 chickpea genotypes (MCC* 696, 537, 80, 283, 392, 361, 252, 397) and drought stress (S1: non-stress, S2: stress at vegetative growth stage, S3: stress at early bloom, S4: stress at early pod visible) at different growth stages. Experiment was arranged in split plot design with four replications. Difference among the drought stress time was found to be significant for investigated traits except biological yield. Differences were observed for genotypes in flowering time, pod information time, physiological maturation time and yield. Plant height reduced due to drought stress in vegetative growth stage. Stem dry weight reduced due to drought stress in pod visibly. Flowering time, maturation time, pod number, number of seed per plant and yield cause of drought stress in flowering was also reduced. The correlation between yield and number of seed per plant and biological yield was positive. The MCC283 and MCC696 were the high-tolerance genotypes. These results demonstrated that drought stress delayed phonological growth in chickpea and that flowering stage is sensitive.

1101 Analytical Solution of Stress Distribution ona Hollow Cylindrical Fiber of a Composite with Cylindrical Volume Element under Axial Loading

Abstract:

The study of the stress distribution on a hollow cylindrical fiber placed in a composite material is considered in this work and an analytical solution for this stress distribution has been constructed. Finally some parameters such as fiber-s thickness and fiber-s length are considered and their effects on the distribution of stress have been investigated. For finding the governing relations, continuity equations for the axisymmetric problem in cylindrical coordinate (r,o,z) are considered. Then by assuming some conditions and solving the governing equations and applying the boundary conditions, an equation relates the stress applied to the representative volume element with the stress distribution on the fiber has been found.

1100 Free Vibration and Buckling of Rectangular Plates under Nonuniform In-Plane Edge Shear Loads

Abstract:

A method for determining the stress distribution of a rectangular plate subjected to two pairs of arbitrarily distributed in-plane edge shear loads is proposed, and the free vibration and buckling of such a rectangular plate are investigated in this work.  The method utilizes two stress functions to synthesize the stress-resultant field of the plate with each of the stress functions satisfying the biharmonic compatibility equation. The sum of stress-resultant fields due to these two stress functions satisfies the boundary conditions at the edges of the plate, from which these two stress functions are determined. Then, the free vibration and buckling of the rectangular plate are investigated by the Galerkin method. Numerical results obtained by this work are compared with those appeared in the literature, and good agreements are observed.

1099 Intensity of Singular Stress Field at the Corner of Adhesive Layer in Bonded Plate

Abstract:

In this paper the strength of adhesive joint under tension and bending is discussed on the basis of intensity of singular stress by the application of FEM. A useful method is presented with focusing on the stress at the edge of interface between the adhesive and adherent obtained by FEM. After analyzing the adhesive joint strength with all material combinations, it is found that to improve the interface strength, thin adhesive layers are desirable because the intensity of singular stress decreases with decreasing the thickness.

1098 Effect of Commercial or Bovine Yeasts on the Performance and Blood Variables of Broiler Chickens Intoxicated with Aflatoxins

Abstract:

The effects of commercial or bovine yeasts on the performance and blood variables of broiler chickens intoxicated with aflatoxin were investigated in broilers. Four hundred eighty broilers (Arbor Acres; 3-wk-old) were randomly assigned to 4 groups. Each group (120 broiler chickens) was further randomly divided into 6 replicates of 20 chickens. The treatments were control diet without additives (treatment 1), 250 ppb AFB1 (treatment 2), commercial yeast, Saccharomyces cerevisiae, (CY 2.5 x 107 CFU/g) + 250 ppb AFB1 (treatment 3) and bovine yeast, Saccharomyces cerevisiae, (BY 2.5 x 107 CFU/g + 250 ppb AFB1 (treatment 4). Complete randomized design (CRD) was used in the experiment. Feed consumption and body weight were recorded at every five-day period. On day 42, carcass compositions were determined from 30 birds per treatment. While chicks were sacrificed, 3-4 ml blood sample was taken and stored frozen at (-20°C) for serum chemical analysis to determine effects of consumption of diets on blood chemistry (total protein, albumin, glucose, urea, cholesterol and triglycerides). There were no significant differences in ADFI among the treatments(P>0.05). However, BWG, FCR and mortality were highly significantly different (P<0.01) between treatments. ADG was significantly reduced (P<0.05) by aflatoxin but was unaffected by aflatoxin supplemented with either commercial or bovine yeasts (P>0.05). In terms of carcass portions, percentage of carcass was unaffected by the treatments, however, percentages of drumstick were reduced by aflatoxin and aflatoxin supplemented commercial yeast. Abdominal fat was significantly reduced (P<0.01) when commercial or bovine yeasts were added to the aflatoxin contaminated diets. Percentage of liver were significantly increased by aflatoxin contamination but were unaffected when yeasts were added to the diets. Blood chemical parameters, i.e. albumin, blood urea nitrogen and glucose were unaffected the treatments, while total protein, cholesterol and triglycerides were significantly decreased by aflatoxin. When yeasts were supplemented, such effect was not differed from the control. It is clearly indicated in the present study that supplementation of either commercial or bovine yeasts had beneficial effects on performance of broiler chickens intoxicated with aflatoxins.

1097 Ethanol Yield of Three Varieties of Cassava (Odongbo, Ofege, and TMS 30572) Using α-Amylase from Germinated Paddy Rice and Yeast from Palm Wine

Abstract:

A process of conversion of flour from three varieties of cassava, namely Odongbo, ofege and TMS30752 to ethanol using α-amylase locally sourced from germinated unhusked paddy rice and yeast isolated from palm wine was developed. It involves the germination of paddy rice for a period of 15days to produce α-amylase for starch hydrolysis and isolation of yeast from palm wine for fermentation. The results showed that optimum amylase yield of “ofada” rice paddy was at 6th day germination which was 576.9ml/g. Ethanol yield for TMS30572 (440.3%) was significantly higher than “Odongbo” (160.2%) and “Ofege’’ (115.1%), Sugar conversion efficiency were 311.0%v/v, 268.2%v/v and 186.84%v/v for TMS30572, “Odongbo” and “Ofege” respectively. The ethanol boiling points were 78oC, 76oC and 80oC for TMS30572, “Odongbo” and “Ofege” respectively. This study showed that cassava varieties affects quality of ethanol produced and germination of “ofada” rice for 6 days ensures optimum production of crude amylase enzyme.

1096 Pulsating Flow of an Incompressible Couple Stress Fluid Between Permeable Beds

Abstract:

The paper deals with the pulsating flow of an incompressible couple stress fluid between permeable beds. The couple stress fluid is injected into the channel from the lower permeable bed with a certain velocity and is sucked into the upper permeable bed with the same velocity. The flow between the permeable beds is assumed to be governed by couple stress fluid flow equations of V. K. Stokes and that in the permeable regions by Darcy-s law. The equations are solved analytically and the expressions for velocity and volume flux are obtained. The effects of the material parameters are studied numerically and the results are presented through graphs.

1095 Relational Impact of Job Stress on Gender Based Managerial Effectiveness in Ghanaian Organizations

Abstract:

This study explored the relationship between occupational stress and the perceived effectiveness of men and women managers in Ghanaian organizations. The exploration is underlined by attempt to understand the degree to which male and female managers in Ghanaian organizations experience occupational stress at the workplace. The purpose is to examine the sources and extents of occupational stress experienced by male and female managers in Ghana. Data was collected using questionnaires and analyzed using both descriptive statistics and correlation analysis. The results showed that female managers in Ghana are more likely to report of more stress experiences in the workplace than their male counterparts. The female managers are more likely to perceive role conflict and alienation as job stressors while the male managers perceived blocked career as a major source of workplace stress. It is concluded that despite the female managers experiencing enormous level of occupational stress, there was no significant differences between their managerial effectiveness and that of the male.

1094 Determination of Stress Concentration Factors of a Steam Turbine Rotor by FEA

Abstract:

Stress Concentration Factors are significant in machine design as it gives rise to localized stress when any change in the design of surface or abrupt change in the cross section occurs. Almost all machine components and structural members contain some form of geometrical or microstructural discontinuities. These discontinuities are very dangerous and lead to failure. So, it is very much essential to analyze the stress concentration factors for critical applications like Turbine Rotors. In this paper Finite Element Analysis (FEA) with extremely fine mesh in the vicinity of the blades of Steam Turbine Rotor is applied to determine stress concentration factors. A model of Steam Turbine Rotor is shown in Fig. 1.

1093 Bi-axial Stress Effects on Barkhausen-Noise

Abstract:

Mechanical stress has a strong effect on the magnitude of the Barkhausen-noise in structural steels. Because the measurements are performed at the surface of the material, for a sample sheet, the full effect can be described by a biaxial stress field. The measured Barkhausen-noise is dependent on the orientation of the exciting magnetic field relative to the axis of the stress tensor. The sample inhomogenities including the residual stress also modifies the angular dependence of the measured Barkhausen-noise. We have developed a laboratory device with a cross like specimen for bi-axial bending. The measuring head allowed performing excitations in two orthogonal directions. We could excite the two directions independently or simultaneously with different amplitudes. The simultaneous excitation of the two coils could be performed in phase or with a 90 degree phase shift. In principle this allows to measure the Barkhausen-noise at an arbitrary direction without moving the head, or to measure the Barkhausen-noise induced by a rotating magnetic field if a linear superposition of the two fields can be assumed.

1092 Extracellular Laccase Production by Co-culture between Galactomyces reesii IFO 10823 and Filamentous Fungal Strains Isolated from Fungus Comb Using Natural Inducer

Abstract:

Extracellular laccases are copper-containing microbial enzymes with many industrial biotechnological applications. This study evaluated the ability of nutrients in coconut coir to enhance the yield of extracellular laccase of Galactomyces reesii IFO 10823 and develop a co-culture between this yeast and other filamentous fungi isolated from the fungus comb of Macrotermes sp. The co-culture between G. reesii IFO 10823 and M. indicus FJ-M-5 (G3) gave the highest activity at 580.20 U/mL. When grown in fermentation media prepared from coconut coir and distilled water at 70% of initial moisture without supplement addition, G3 produced extracellular laccase of 113.99 U/mL.

1091 Optimization of New 25A-size Metal Gasket Design Based on Contact Width Considering Forming and Contact Stress Effect

Abstract:

At the previous study of new metal gasket, contact width and contact stress were important design parameter for optimizing metal gasket performance. However, the range of contact stress had not been investigated thoroughly. In this study, we conducted a gasket design optimization based on an elastic and plastic contact stress analysis considering forming effect using FEM. The gasket model was simulated by using two simulation stages which is forming and tightening simulation. The optimum design based on an elastic and plastic contact stress was founded. Final evaluation was determined by helium leak quantity to check leakage performance of both type of gaskets. The helium leak test shows that a gasket based on the plastic contact stress design better than based on elastic stress design.

1090 Utilization of Wheat Bran as Bed Material in Solid State Bacterial Production of Lactic Acid with Various Nitrogen Sources

Abstract:

The present experimental investigation brings about a comparative study of lactic acid production by pure strains of Lactobacilli (1) L. delbreuckii (NCIM2025), (2) L. pentosus (NCIM 2912), (3) Lactobacillus sp.(NCIM 2734, (4) Lactobacillus sp. (NCIM2084) and coculture of strain-1 and Stain-2 in solid bed of wheat bran, under the influence of different nitrogen sources such as baker-s yeast, meat extract and proteose peptone. Among the pure cultures, strain-3 attained lowest pH value of 3.44, hence highest acid formation 46.41 g/L, while the coculture attained an overall maximum value 47.56 g/L lactic acid (pH 3.38) at 15 g/L and 20 g/L level of baker-s yeast, respectively.

1089 Drought Stress Indices in Some Silage Maize Cultivars

Abstract:

Several yield-based stress indices have been developed that may be more applicable to work on drought tolerance. In this study, we investigate possibility of using stress susceptibility index (SSI), tolerance index (TOL), yield stability index (YSI), yield index (YI), stress tolerance index (STI), geometric mean productivity (GMP), harmonic mean (HARM), mean productivity (MP) to identify genotypic performance of some maize cultivars under normal and stressed condition. The results indicate that it was possible to identify superior genotypes for drought tolerance based on their stress indices and generally SSI indices which showed the lowest negative correlation with dry matter yield can be used as the best index for maize breeding programs to introduce drought tolerant hybrids. It was found that SC 647 showed the best behavior under drought stress condition based on TOL and SSI. A higher STI, GMP, and HARM values were attained for ko6. It can be suggested that ko6 should be cultivated in moderate stressful environment of Iran.

1088 Effects of Thread Dimensions of Functionally Graded Dental Implants on Stress Distribution

Abstract:

In this study, stress distributions on dental implants made of functionally graded biomaterials (FGBM) are investigated numerically. The implant body is considered to be subjected to axial compression loads. Numerical problem is assumed to be 2D, and ANSYS commercial software is used for the analysis. The cross section of the implant thread varies as varying the height (H) and the width (t) of the thread. According to thread dimensions of implant and material properties of FGBM, equivalent stress distribution on the implant is determined and presented with contour plots along with the maximum equivalent stress values. As a result, with increasing material gradient parameter (n), the equivalent stress decreases, but the minimum stress distribution increases. Maximum stress values decrease with decreasing implant radius (r). Maximum von Mises stresses increases with decreasing H when t is constant. On the other hand, the stress values are not affected by variation of t in the case of H = constant.

1087 Leaf Pigments Help Almond Explants Tolerating Osmotic Stress

Abstract:

This study was conducted to evaluate the response of almond genotypes to osmotic stress in vitro in order to screen drought tolerance. Explants subjected to polyethyleneglycol osmotic stress (0, 3.5, and 7.0% WV) on the MS medium. Concentrations of photosynthesis pigments, anthocyanins, and carothenoids were significantly reduced under osmotic stress. Under osmotic stress, leaf water content, cellular membrane stability and pigments concentrations were significantly higher in the leaves of drought tolerant genotypes. The results revealed that carotenoids and anthocyanins may act as photoprotectant compounds in almond leaves and involved in drought tolerance system of the plant.
