3 Effect of Two Entomopathogenic Fungi Beauveria bassiana and Metarhizium anisopliae var. acridum on the Haemolymph of the Desert Locust Schistocerca gregaria

Abstract:

Effect of Beauveria bassiana and Metarhizium anisopliae var. acridum on the 5th instar nymphs of Schistocerca gregaria was studied in the laboratory. Infection by these both entomopathogenic fungi caused reduction in the hemolymph total protein. The average amounts of total proteins were 2.3, 2.07, 2.09 µg/100 ml of haemolymph in the control and M. anisopliae var. acridum, and B. bassiana based-treatments, respectively. Three types of haemocytes were recognized and identified as prohaemocytes, plasmatocytes and granulocytes. The treatment caused significant reduction in the total haemocyte count and in each haemocyte type on the 9th day after its application.

2 Activities of Alkaline Phosphatase and Ca2+ATPase over the Molting Cycle of mud Crab (Scylla serrata)

Abstract:

The activities of alkaline phosphatase and Ca2+ATPase in mud crab (Scylla serrata) collected from a soft-shell crab farm in Chantaburi Province, Thailand, in several stages of molting cycle were observed. The results showed that the activity of alkaline phosphatase in gill after molting was highly significant (p<0.05) comparing to those at intermolt and premolt stages. The activity profiles of alkaline phosphatase in integument and haemolymph were similar showing a decrease from intermolt to 2- week premolt stage and increased during 2-day premolt to 6-h postmolt stage before dropping at 7-day postmolt stage, while this enzyme in the gill was quite low at intermolt and premolt stages. For Ca2+ATPase, the activity profiles in gill and integument corresponded to the molting variation, especially the activities increased during 5-7 day postmolt stage were at highly significant levels (p<0.05) comparing to those at premolt and early postmolt stages. The highest activity of Ca2+ATPase in haemolymph was found at 2-week premolt stage (p<0.05). Changes in alkaline phosphatase and Ca2+ATPase activities over the molting cycle clearly indicated their active functions on calcification.

1 Activation of Prophenoloxidase during Bacterial Injection into the Desert Locust, Schistocerca Gregaria

Abstract:

The present study has been conducted to characterize the prophenoloxidase (PPO) system of the desert locust, Schistocerca gregaria following injection of Bacillus thuringiensis kurstaki (Bt). The bulk of PPO system was associated with haemocytes and a little amount was found in plasma. This system was activated by different activators such as laminarin, lipopolysaccharide (LPS) and trypsin suggesting that the stimulatory mechanism may involve an enzyme cascade of one or more associated molecules. These activators did not activate all the molecules of the cascade. Presence of phenoloxidase activity (PO) coincides with the appearance of protein band with molecular weight (MW) 70.154 KD (Kilo Dalton).
