3 Analysis of Roasted and Ground Grains on the Seoul (Korea) Market for Their Contaminants of Aflatoxins, Ochratoxin A and Fusarium Toxins by LC-MS/MS

Abstract:

A sensitive and specific method for quantitative determination of aflatoxins(B1, B2, G1,G2), deoxynivalenol, fumonisin(B1,B2), ochratoxin A, zearalenone, T-2 and HT-2 in roasted and ground grains using liquid chromatography combined with tandem mass spectrometry. A double extraction using a phosphate buffer solution followed by methanol was applied to achieve effective co extraction of 11 mycotoxins. A multitoxin immunoaffinity column for all these mycotoxins was used to clean up the extract. The LODs of mycotoxins were 0.1~6.1 μg/kg, LOQs were 0.3~18.4 μg/kg. Forty seven samples collected from Seoul (Korea) for mycotoxin contamination monitoring. The results showed that the occurrence of zearalenone and deoxynivalenol were frequent. Zearalenone was detected in all samples and deoxynivalenol was detected in 80.9 % samples in the range 0.626 ~ 29.264 μg/kg and N.D ~ 48.332 μg/kg respectively. Fumonisins and ochratoxin A were detected in 46.8% samples and 17 % samples respectively, aflatoxins and T-2/HT-2 toxins were not detected all samples.

2 Induction of Hsp70 and Antioxidant Status in Porcine Granulosa Cells in Response to Deoxynivalenol and Zearalenone Exposure in vitro

Abstract:

The aim of this study was to determine the activity of superoxide dismutase (SOD), glutathione peroxidase (GPx), total antioxidant status (TAS) and accumulation of Hsp70 in porcine ovarian granulosa cells after deoxynivalenol (DON) and zearalenone (ZEA) exposure in vitro. Porcine ovarian granulosa cells were incubated with DON/ZEA administrations as follows: group A (10/10 ng/mL), group B (100/100 ng/mL), group C (1000/1000 ng/mL), and the control group without any additions for 24h. In this study mycotoxins developed stress reaction of porcine ovarian granulosa cells and increased accumulation of Hsp70 what resulted in increasing activities of SOD and GPx in groups with lower doses of mycotoxins. High dose of DON and ZEA had opposite effect on GPx activity than the lower doses. Slight increase in TAS of porcine granulosa cells was observed after mycotoxins exposure. These results contribute towards the understanding of cellular stress and its response.

1 The Comparation of Limits of Detection of Lateral Flow Immunochromatographic Strips of Different Types of Mycotoxins

Abstract:

Mycotoxins are secondary metabolic products of fungi. These are poisonous, carcinogens and mutagens in nature and pose a serious health threat to both humans and animals, causing severe illnesses and even deaths. The rapid, simple and cheap detection methods of mycotoxins are of immense importance and in great demand in the food and beverage industry as well as in agriculture and environmental monitoring. Lateral flow immunochromatographic strips (ICSTs) have been widely used in food safety, environment monitoring. 46 papers were identified and reviewed on Google Scholar and Scopus for their limit of detection and nanomaterial on Lateral flow ICSTs on different types of mycotoxins. The papers were dated 2001-2021. 25 papers were compared to identify the lowest limit of detection of among different mycotoxins (Aflatoxin B1: 10, Zearalenone: 5, Fumonisin B1: 5, Trichothecene-A: 5). Most of these highly sensitive strips are competitive. Sandwich structures are usually used in large scale detection. In conclusion, the limit of detection of Aflatoxin B1 is the lowest among these mycotoxins. Gold-nanoparticle based immunochromatographic test strips have the lowest limit of detection. Five papers involve smartphone detection and they all detect aflatoxin B1 with gold nanoparticles.
