58 Hydropriming and Osmopriming Effects on Cumin(Cuminum Cyminum L.) Seeds Germination
Authors: E. Neamatollahi, M. Bannayan, A. Souhani Darban, A. Ghanbari
Abstract:
In production of medicinal plants, seed germination is very important problem. The treated seeds (control, hydro priming and ZnSO4) of Cumin (Cuminum cyminum L.) were evaluated at germination and seedling growth for tolerance to salt (NaCl and Na2SO4) conditions at the same water potentials of 0.0, -0.3, -0.6, - 0.9 and -1.2MPa. Electrical conductivity (EC) values of the NaCl solutions were 0.0, 6.5, 12.7, 18.4 and 23.5 dSm-1, respectively. The objective of the study was to determine factors responsible for germination and early seedling growth due to salt toxicity or osmotic effect and to optimize the best priming treatment for these stress conditions. Results revealed that germination delayed in both solutions, having variable germination with different priming treatments. Germination, shoot and weight, root and shoot length were higher but mean germination time and abnormal germination percentage were lower in NaCl than Na2SO4 at the same water potential. The root / shoot weight and R/S length increased with increase in osmotic potential in both NaCl and Na2SO4 solutions. NaCl had less inhibitor effect on seedling growth than the germination. It was concluded that inhibition of germination at the same water potential of NaCl and Na2SO4 resulted from salt toxicity rather than osmotic effect. Hydro priming increased germination and seedling growth under salt stress. This protocol has practical importance and could be recommended to farmers to achieve higher germination and uniform emergence under field conditions. Keywords: Priming, Germination, Nacl, ZnSo4, Na2So4.

Abstract:
The use of engineered nanomaterials has increased as a result of their positive impact on many sectors of the economy, including agriculture. Silver nanoparticles (AgNPs) are now used to enhance seed germination, plant growth, and photosynthetic quantum efficiency and as antimicrobial agents to control plant diseases. In this study, we examined the effect of AgNP dosage on the seed germination of three plant species: corn (Zea mays L.), watermelon (Citrullus lanatus [Thunb.] Matsum. & Nakai) and zucchini (Cucurbita pepo L.). This experiment was designed to study the effect of AgNPs on germination percentage, germination rate, mean germination time, root length and fresh and dry weight of seedlings for the three species. Seven concentrations (0.05, 0.1, 0.5, 1, 1.5, 2 and 2.5 mg/ml) of AgNPs were examined at the seed germination stage. The three species had different dose responses to AgNPs in terms of germination parameters and the measured growth characteristics. The germination rates of the three plants were enhanced in response to AgNPs. Significant enhancement of the germination percentage values was observed after treatment of the watermelon and zucchini plants with AgNPs in comparison with untreated seeds. AgNPs showed a toxic effect on corn root elongation, whereas watermelon and zucchini seedling growth were positively affected by certain concentrations of AgNPs. This study showed that exposure to AgNPs caused both positive and negative effects on plant growth and germination. Keywords: Citrullus lanatus, Cucurbita pepo, seed germination, seedling growth, silver nanoparticles, Zea mays.

Abstract:
One problem of synthetic sunflower cultivation is an erratic germination of the seeds. To improve the germination, presowing seed treatment with an ultrasound was tested. All treatments were carried out at 40 kHz frequency with the intensities of 40, 60, 80 and 100% of the ultrasonic generator total power (250 W) for the durations of 5, 10, 15 and 20 minutes. Data on seed germination percentage, seed vigor index (SVI), root and shoot lengths of seedlings were collected. The results showed that germination, SVI, root and shoot lengths of ultrasonic treated seedlings were different from the control, depending on intensity of the ultrasound. The effects of ultrasonic treatment were significant on germination, resulting in a maximum increase of 43% at 40 and 60% intensities compared to that of the control seeds. In addition, seedlings of these 2 treatments had higher SVI and longer root and shoot lengths than that of the control seedlings. All treatment durations resulted in higher germination and SVI, longer root and higher shoot lenghts of seedlings than the control. Among the duration treatments, only SVI and seedling root length were significantly different. Keywords: Ultrasonic, germination, root length, shoot length, Helianthus annuus L.

Abstract:
Using effluent as a sustainable water resource for agriculture not only could provide part of water needs but also would save the existing water resources, durably. Vinasse, the effluent of ethyl alcohol factories, a by-product, which is derived from sugarcane molasses, is one of the water resources that could be effectively utilized for agricultural purposes. In the present study in order to investigate the application of different ratios of water: vinasse on germination and growth of barley seedlings an experiment was designed in pots with completely randomized design with three replications and control treatment. The consequences of four irrigation levels were studied with different water: effluent ratios (100% water, 90% water & 10% effluent, 75% water & 25% effluent, 50% water & 50% effluent) on germination and growth of barley seedling components in sandy-loam soil. The results showed that, with increasing the percentage of vinasse in the irrigation admixture, the germination percentage in barley seedlings decreased, significantly, so that the decrease in germination in comparison with the control samples in the second and third treatments was 20% and 93.33%, respectively. Seed germination percentage was about 46.66. The average stem length in seedlings was 14.3 mm and the average root length was 9.37 mm. The averages of the soils Electrical Conductivity (EC) and pH which were under irrigation with different ratios of vinasse (dSm-1) were 5.85 and 7.32, respectively, which showed a 76.2% increase in soil salinity.

Abstract:
The evaluation of non-conventional water resources on seed germination and seedling growth performance at early growth stages is still in progress especially in forage crops. This study was designed to test the effect of four types of water qualities (treated wastewater (TWW), industrial water (IW), grey water (GW), and Distilled water (DW)) on germination and early seedling vigor of Leucaena leucocephala. The results showed that the germination was not significantly affected by the different water qualities. Seed germination reached maximum after 17, 14, 14, and 21 days under GW, IW, TWW, and DW treatments, respectively. The highest mean of shoot length was scored under the GW treatment. And, the highest mean of root length was scored under DW which was not significant from GW treatment. The means of shoot fresh was the highest under the TWW. The means of root fresh weight was not significantly different from each other's under different treatments. The growth performance was in progress with no mortality during 21 days of growth. Thus, the best non-conventional water qualities alternatives based on the cleanness, nutrients, and toxicity are the GW, TWW and IW, respectively. Keywords: Seed germination, Growth performance, Leucaena, Multipurpose forest trees, Waste water, Grey water

Abstract:
Biologically active peptides are of particular interest in food science and human nutrition because they have been shown to play several physiological roles. In vitro gastrointestinal digestion of lentil and whey proteins in this study produced high angiotensin-I converting enzyme inhibitory activity with 75.5±1.9 and 91.4±2.3% inhibition, respectively. High ACE inhibitory activity was observed in lentil after 5 days of germination (84.3±1.2%). Fractionation by reverse phase chromatography gave inhibitory activities as high as 86.3±2.0 for lentil, 94.8±1.8% for whey and 93.7±1.7% at 5th day of germination. Further purification by HPLC resulted in several inhibitory peptides with IC50 values ranging from 0.064 to 0.164 mg/ml. These results demonstrate that lentil proteins are a good source of peptides with ACE inhibitory activity that can be released by germination or gastrointestinal digestion. Despite the lower bioactivity in comparison with whey proteins, incorporation of lentil proteins in functional food formulations and natural drugs look promising. Keywords: ACE inhibitory peptides, digestion, germination, lentil proteins, whey proteins

Authors: H. Sakamoto, J. Tochimoto, S. Kurosawa, M. Suzuki, S. Oguri
Abstract:
Salt stress adversely affects plant growth at various stages of development including seed germination, seedling establishment, vegetative growth and finally reproduction. Because of their immobile nature, plants have evolved mechanisms to sense and respond to salt stress. Seed dormancy is an adaptive trait that enables seed germination to coincide with favorable environmental conditions. We identified a novel locus of Arabidopsis, designated SHG1 (salt hypersensitive germination 1), whose disruption leads to reduced germination rate under moderate salt stress conditions. SHG1 encodes a transmembrane protein with an ankyrin-repeat motif that has been implicated in diverse cellular processes such as signal transduction. The shg1-disrupted Arabidopsis mutant died at the cotyledon stage when sown on salt-containing medium, although wild-type plants could form true leaves under the same conditions. On the other hand, this mutant showed similar phenotypes to wild-type plants when sown on medium without salt and transferred to salt-containing medium at the vegetative stage. These results suggested that SHG1 played indispensable role in the seed germination and seedling establishment under moderate salt stress conditions. SHG1 may be involved in the release of seed dormancy.

Seed germination percentage, root length and shoot length in these conditions were measured. This experiment was conducted as a factorial completely randomized design with four replications in laboratory conditions. The results showed that each of aging treatments used in this experiment can be used to detect differences in vigor of wheat varieties. Wheat cultivars illustrated significant differences in germination percentage, root length and shoot length in terms of premature aging. The wheat cultivars; Astar and Vynak had maximum germination percentage and Karkheh, respectively Kavir and Chamran had lowest percentage of seed germination. Reactions of root and shoot length of wheat cultivars was also different. The results showed that the seeds with a stronger vigor affected less in premature aging condition and the difference between the percentage of seed germination under normal conditions and stress was significant and the seeds with the weaker vigor were more sensitive to the premature aging stress and the premature aging had more severe negative impact on seed vigor. Keywords: Wheat cultivars, seed vigor, premature aging effects.

Abstract:
Wheat is an important cereal crop for food security. Boosting the wheat production and productivity is the major challenge across the nation. Good quality of seed is required for maintaining optimum plant stand which ultimately increases grain yield. Ensuring a good germination is one of the key steps to ensure proper plant stand and moisture assurance during seed germination may help to speed up the germination. The tiny size of nanoparticles may help in entry of water into seed without disturbing their internal structure. Considering above, a laboratory experiment was conducted during 2012-13 at G.B. Pant University of Agriculture and Technology, Pantnagar, India. The completely randomized design was used for statistical analysis. The experiment was conducted in two phases. In the first phase, the appropriate concentration of nanoparticles for seed treatment was screened. In second phase seed soaking hours of nanoparticles for better seed germination were standardized. Wheat variety UP2526 was taken as test crop. Four nanoparticles (TiO2, ZnO, nickel and chitosan) were taken for study. The crop germination studies were done in petri dishes and standard package and practices were used to raise the seedlings. The germination studies were done by following standard procedure. In first phase of the experiment, seeds were treated with 50 and 300 ppm of nanoparticles and control was also maintained for comparison. In the second phase of experiment, seeds were soaked for 4 hours, 6 hours and 8 hours with 50 ppm nanoparticles of TiO2, ZnO, nickel and chitosan along with control treatment to identify the soaking time for better seed germination. Experiment revealed that the application of nanoparticles help to enhance seed germination. The study revealed that seed treatment with nanoparticles at 50 ppm concentration increases root length, shoot length, seedling length, shoot dry weight, seedling dry weight, seedling vigour index I and seedling vigour index II as compared to seed soaking at 300 ppm concentration. This experiment showed that seed soaking up to 4 hr was better as compared to 6 and 8 hrs. Seed soaking with nanoparticles specially TiO2, ZnO, and chitosan proved to enhance germination and seedling growth indices of wheat crop.

Pollen of ‘Black’, ‘Thyto’ resulted in high fruit weight, fruit diameter, fruit length, bigger flesh thickness, better total soluble solids as compared with other pollens. The observation of pollen-growth in vitro revealed that pollen germination at 15% sucrose concentration are required for optimum pollen germination with the high pollen germination were found in ‘Black’, ‘Thyto’. From the result, we concluded that ‘Black’, ‘Thyto’ were proved to be good pollinizers in ‘Long Red B’. Therefore, artificial cross-pollination using ‘Black’, ‘Thyto’ as pollinizers were strongly recommended for ‘Long Red B’ cultivar in wax apple orchard.

Abstract:
The application of biochar to soils is becoming more and more common. Its application which is generally reported to improve the physical, chemical, and biological properties of soils, has an indirect effect on soil health and increased crop yields. However, many of the previous results are highly variable and dependent mainly on the initial soil properties, biochar characteristics, and production conditions. In this study, two biochars which are biochar II (BC II) derived from a blend of paper sludge and wheat husks and biochar 005 (BC 005) derived from sewage sludge with a KCl additive, are used, and the physical and chemical properties of BC II are characterized. To determine the potential impact of salt stress and toxic and volatile substances, the second part of this study focused on the effect biochars have on germination of salad (Lactuca sativa L.), barley (Hordeum vulgare), and cress (Lepidium sativum) respectively. Our results indicate that Biochar II showed some unique properties compared to the soil, such as high EC, high content of K, Na, Mg, and low content of heavy metals. Concerning salad and barley germination test, no negative effect of BC II and BC 005 was observed. However, a negative effect of BC 005 at 8% level was revealed. The test of the effect of volatile substances on germination of cress revealed a positive effect of BC II, while a negative effect was observed for BC 005. Moreover, the water holding capacities of biochar-sand mixtures increased with increasing biochar application. Collectively, BC II could be safely used for agriculture and could provide the potential for a better plant growth. Keywords: Biochar, phytotoxic tests, seedlings growth, water holding capacity.

Authors: Mohamed Rashad, Mohamed Hafez, Mohamed Emran, Emad Aboukila, Ibrahim Nassar
Abstract:
Environment-friendly organic wastes of Brewers' spent grain, a byproduct of the brewing process, have recently used as soil amendment to improve soil fertility and plant production. In this work, treatments of 1% (T1) and 2% (T2) of spent grains, 1% (C1) and 2% (C2) of compost and mix of both sources (C1T1) were used and compared to the control for growing Zea mays L. on sandy soil under arid Mediterranean climate. Soils were previously incubated at 65% saturation capacity for a month. The most relevant soil physical and chemical parameters were analysed. Water holding capacity and soil organic matter (OM) increased significantly along the treatments with the highest values in T2. Soil pH decreased along the treatments and the lowest pH was in C1T1. Bicarbonate decreased by 69% in C1T1 comparing to control. Total nitrogen (TN) and available P varied significantly among all treatments and T2, C1T1 and C2 treatments increased 25, 17 and 11 folds in TN and 1.2, 0.6 and 0.3 folds in P, respectively related to control. Available K showed the highest values in C1T1. Soil micronutrients increased significantly along all treatments with the highest values in T2. After corn germination, significant variation was observed in the velocity of germination coefficients (VGC) among all treatments in the order of C1T1>T2>T1>C2>C1>control. The highest records of final germination and germination index were in C1T1 and T2. The spent grains may compensate deficiencies of macro and micronutrients in newly reclaimed sandy soils without adverse effects to sustain crop production with a rider that excessive or continuous use need to be circumvented. Keywords: Spent grain, compost, micronutrients, macronutrients, water holding capacity, plant growth.

Authors: R. Pudasaini, R. B. Thapa, P. R. Poudel
Abstract:
An experiment was conducted to determine the effect of pollination on seed quality of rapeseed in Chitwan, Nepal during 2012-2013. The experiment was designed in Randomized Complete Block with four replications and five treatments. The rapeseed plots were caged with mosquito nets at 10% flowering except natural pollination. Two-framed colonies of Apis mellifera L. and Apis cerana F. were introduced separately for pollination, and control plot caged without pollinators. The highest germination percent was observed on Apis cerana F. pollinated plot seeds (90.50% germination) followed by Apis mellifera L. pollinated plots (87.25 %) and lowest on control plots (42.00% germination) seeds. Similarly, seed test weight of Apis cerana F. pollinated plots (3.22 gm/ 1000 seed) and Apis mellifera L. pollinated plots (2.93 gm/1000 seed) were and lowest on control plots (2.26 gm/ 1000 seed) recorded. Likewise, oil content was recorded highest on pollinated by Apis cerana F. (36.1%) followed by pollinated by Apis mellifera L. (35.4%) and lowest on control plots (32.8%). This study clearly indicated pollination increases the seed quality of rapeseed and therefore, management of honeybee is necessary for producing higher quality of rapeseed under Chitwan condition.

Authors: T. A. Abegunde, O. B. Oyewole, T. A. Sanni
Abstract:
A process of conversion of flour from three varieties of cassava, namely Odongbo, ofege and TMS30752 to ethanol using α-amylase locally sourced from germinated unhusked paddy rice and yeast isolated from palm wine was developed. It involves the germination of paddy rice for a period of 15days to produce α-amylase for starch hydrolysis and isolation of yeast from palm wine for fermentation. The results showed that optimum amylase yield of “ofada” rice paddy was at 6th day germination which was 576.9ml/g. Ethanol yield for TMS30572 (440.3%) was significantly higher than “Odongbo” (160.2%) and “Ofege’’ (115.1%), Sugar conversion efficiency were 311.0%v/v, 268.2%v/v and 186.84%v/v for TMS30572, “Odongbo” and “Ofege” respectively. The ethanol boiling points were 78oC, 76oC and 80oC for TMS30572, “Odongbo” and “Ofege” respectively. This study showed that cassava varieties affects quality of ethanol produced and germination of “ofada” rice for 6 days ensures optimum production of crude amylase enzyme.

Authors: Stella O. Olubodun, George E. Eriyamremu
Abstract:
The study examined the effect of Bonny Light whole crude oil (WC) and its water soluble fraction (WSF) on the activities of antioxidant enzymes (catalase (CAT) and superoxide dismutase (SOD)) and crude mitochondria ATPases in the radicle of germinating bean (Vigna unguiculata). The percentage germination, level of lipid peroxidation, antioxidant enzyme and mitochondria Ca2+ and Mg2+ ATPase activities were measured in the radicle of bean after 7, 14 and 21 days post germination. Viable bean seeds were planted in soils contaminated with 10ml, 25ml and 50ml of whole crude oil (WC) and its water soluble fraction (WSF) to obtain 2, 5 and 10% v/w crude oil contamination. There was dose dependent reduction of the number of bean seeds that germinated in the contaminated soils compared with control (p<0.001). The activities of the antioxidant enzymes, as well as, adenosine triphosphatase enzymes, were also significantly (p<0.001) altered in the radicle of the plants grown in contaminated soil compared with the control. Generally, the level of lipid peroxidation was highest after 21 days post germination when compared with control. Stress to germinating bean caused by Bonny Light crude oil or its water soluble fraction resulted in adaptive changes in crude mitochondria ATPases in the radicle.

Authors: E. Hosseini, M. Kadivar, M. Shahedi
Abstract:
Malting is usually carried out on intact barley seed, while hull is still attached to it. In this study, oat grain with and without hull was subjected to controlled germination to optimize its enzymes activity, in such a way that lipase has the lowest and α- amylase and proteinase the highest activities. Since pH has a great impact on the activity of the enzymes, the pH of germination media was set up to 3 to 8. In dehulled oats, lipase and α-amylase had the lowest and highest activities in pHs 3 and 6, respectively whereas the highest proteinase activity was evidenced at pH 7 and 4 in the oats with and without hull respectively. While measurements indicated that the effect of hull on the enzyme activities particularly in lipase and amylase at each level of the pH are significantly different, the best results were obtained in those samples in which their hull had been removed. However, since the similar lipase activity in germinated dehulled oat were recorded at the pHs 4 and 5, therefore it was concluded that pH 5 in dehulled oat seed may provide the optimum enzyme activity for all the enzymes. Keywords: Enzyme activity, malting, oat, optimization.
