3 Patients Undergoing Diagnostic Bronchoscopy at General Ahmad Yani Hospital in 2023: A Profile of Lung Cancer Types

Abstract:
Flexible fiberoptic bronchoscopy (FFB) is the essential method for both the diagnosis and staging of central lung cancer. Concerning central lesions, the sensitivity of endobronchial forceps biopsy of a visible endobronchial lesion is 74%. The aim is to determine the distribution of lung cancer types of patients diagnosed with and who underwent bronchoscopy at General Ahmad Yani Metro Hospital. Methods: This study is a cross-sectional descriptive study. Data are obtained from histopathological results of lung malignancy through bronchoscopy either from forceps biopsy, washing and brushing. Results: Lung cancer is more found in men (72.2%) than women (27.8%). The average age for men ranges 57 years old for Non-Small Cell and 56 years old for Small Cell case. Most histopathology found in non-small cell lung cancer (87.7%), with adenocarcinoma as the most common type (68.4%), followed by squamous cell carcinoma (29.1%). Findings for adenocarcinoma showed that men had an incidence of 64.8% and women had an incidence of 35.2%. Like NSCLC, men can have up to 90% of cases of SCLC compared to 10% in women. In addition to non-small cell and small cell types, other types, such as large cells and renal carcinoma metastases, were obtained. Conclusions: Adenocarcinoma lung cancer is the most widely obtained type with predominantly male patients.Keywords: bronchoscopy, forceps biopsy, washing, brushing

Abstract:
Methylene blue is a popular water-soluble dye that is used for dyeing a variety of substrates such as bacteria, wool, and silk. Methylene blue discharged into the aquatic environment will cause health problems for living things. Treatment method for industrial wastewater may be divided into three main categories: physical, chemical, and biological. Among them, adsorption technology is generally considered to be an effective method for quickly lowering the concentration of dissolved dyes in a wastewater. This has attracted considerable research into low-cost alternative adsorbents for adsorbing or removing coloring matter. In this research, pectin from durian seeds was utilized here to assess their ability for the removal of methylene blue. Adsorption parameters are contact time and dye concentration were examined in the batch adsorption processes. Pectin characterization was performed by FTIR spectrometry. Methylene blue concentration was determined by using UV-Vis spectrophotometer. FTIR results show that the samples showed the typical fingerprint in IR spectrogram. The adsorption result on 10 mL of 5 mg/L methylene blue solution achieved 95.12% when contact time 10 minutes and pectin 0.2 g.Keywords: pectin, methylene blue, adsorption, durian seed

Abstract:
Bacteria ability to survive in antibiotic is controlled by the presence of gene that encodes the antibiotic resistance protein. The instability of the antibiotic resistance gene can be observed by exposing the bacteria under the lethal dose of antibiotic. Low concentration of antibiotic can induce mutation, which may take a role in bacterial adaptation through the antibiotic concentration. Lactobacillus casei FNCC 0090 is one of the probiotic bacteria that has an ability to survive in tetracycline by expressing the tetM gene. The aims of this study are to observe the possibilities of mutation happened in L.casei FNCC 0090 by exposing in sub-lethal dose of tetracycline and also observing the instability of the tetM gene by comparing the sequence between the wild type and mutant. L.casei FNCC 0090 has a lethal dose in 60 µg/ml, low concentration is applied to induce the mutation, the range from 10 µg/ml, 15 µg/ml, 30 µg/ml, 45 µg/ml, and 50 µg/ml. L.casei FNCC 0090 is exposed to the low concentration from lowest to the highest concentration to induce the adaptation. Plasmid is isolated from the highest concentration culture which is 50 µg/ml by using modified alkali lysis method with the addition of lysozyme. The tetM gene is isolated by using PCR (Polymerase Chain Reaction) method, then PCR amplicon is purified and sequenced. Sequencing is done on both samples, wild type and mutant. Both sequences are compared and the mutations can be traced in the presence of nucleotides changes. The changing of the nucleotides means that the tetM gene is instable.Keywords: L. casei FNCC 0090, probiotic, tetM, tetracycline
